Journal of International Oncology 2011;38(10):741-744
Macrophage migration-inhibitory factor (MIF) is a cytokine which can promote tumorigenesis.MIF can promote tumorigenesis by directly inducing oncogene transformation, stimulating cell division,as well as indirectly inhibiting tumor apoptosis, promoting its invasiveness and angiogenesis.MIF-targeted biological therapy may become a new therapeutic strategy for malignant tumor.
2.Expression of aquaporin 8 in human fetal membrane and placenta of idiopathic polyhydramnios
Chinese Journal of Obstetrics and Gynecology 2009;44(1):19-22
Objective To determine the expression of Aquaporin 8 (AQP8) in the fetal membrane and placenta of idiopathic polyhydramnios. Methods The amnion, chorion and placenta were collected from 12 term pregnancies with idiopathic polyhydramnios(polyhydramnios group) and 12 term pregnancies who were normal (control group). The expression of AQP8 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression of AQP8 protein was detected by immunohistochemistry. Results The expression of AQP8 mRNA in amnion, chorion and placenta of polyhydramnios group was (0.78±0.13), (0.58±0.10), and (0.86±0.15) respectively, and that of control group was (0.39±0.07 ), (0.45±0.09),and (0.34±0.09) respectively. The expression of AQP8 protein in amnion, chorion and placenta of polyhydramnios group was (0.195±0.024), (0. 170±0.028), and (0. 193±0.024) respectively, and that of control group was (0. 151±0.018), (0.156±0.024), and (0. 152±0.023) respectively. In all 3 types of tissues the expression of AQP8 mRNA of polyhydramnios group was higher than that of control group (P<0.05). In anmion and placenta the expression of AQP8 protein of polyhydramnios group was also increased compared to that of control group (P<0.05), but in chorion the difference in AQP8 protein expression between the two groups was not significant(P>0.05). Conclusion The expression of AQP8 mRNA and protein is significantly increased in the anmion and placenta of polyhydramnios, suggesting that AQP8 may play an important role in the regulation of amniotic fluid volume.

Objective To investigate the effect and mechanism of low-dose remifentanil(RF)on systemic artery tension of baby rabbit with septic shock. Methods Thirty-six systemic artery strips were prepared from 12 baby rabbits and randomly divided into 6 groups:control group,lipopoly-saccharide(LPS)group,RF group,LPS + RF group,NG-nitro-L-arginine(L-NNA)pretreated group,LPS + L-NNA pretreated group. Each group had six rings(n =6).The effect of low-dose RF on the systemic artery tension of baby rabbit with septic shock was observed with isolated vascular ring technique. The change was also obversed after L-NNA pretreatment. Results In control group:the relaxant rates after low-dose RF at the time point of 5 min,10 min,20 min were(18.48±3.96)%,(23.63±4.42)%,(28.33±3.73)%(P<0.05),compared with NS group. After pretreatment with L-NNA,RF-related relaxant rates of systemic arteries decreased significantly to(8.15 ± 1.01)%,(13.08 ± 1.46)%,(18.54 ±2.94)%(P < 0.05).In LPS group, low dose RF(4 μg/L)did not bring out any response to systemic arteries(P >0.05).The tension was not affected with pretreatment of L-NNA(P > 0.05).Conclusion These results suggest that low dose RF has relaxant effect on systemic arteries of baby rabbits, and NO may be involved.Low dose RF has no relaxant effect on LPS-pretreated systemic arteries of baby rabbits, and it has no association with NO.

Objective The purpose of this study is to determine whether atrial expression of hepatocyte growth factor (HGF), connective tissue growth factor (CTGF) and transforming growth factor-beta1 (TGF-13,) is altered in patients with rheumatic heart disease (RHD) and chronic atrial fibrillation (AF).Methods Atrial tissue was obtained from the right atrial appendage during heart surgery from 35 patients. In 27 patients with rheumatic heart disease, 8 patients had no history of AF, and 19 had chronic persistent AF (≥6 months cAF). Other 8 patients with congenital heart disease had no history of AF were the control group. The mRNA expression of HGF, CTGF, TGF-β1, collagen Ⅰ and collagen Ⅲ was measured by the real-time fluorescence quota polymerase chain reaction (real time PCR). The fibrosis of right atrial appendage was detected by HE and Masson staining. Results The amount of CTGF, TGF-β1, collagen Ⅰ and collagen Ⅲ was significantly increased in patients with sinus rhythm compared with the control group (P<0.05) and further increased in patients with chronic AF compared with patients with sinus rhythm (P<0.05). The amount of HGF was significantly decreased in patients with chronic AF compared with patients with sinus rhythm and the control group (P<0.05). But the difference of the latter two groups was not statistically significant.Correlation analysis demonstrated that the mRNA expression of collagen Ⅰ , collagen Ⅲ, TGF-β1, and CTGF in right atrial appendage of RHD and AF was positively correlated with the left atrial diameter and the area of myocardial fibrosis. Conclusion In human atrium with RHD, the mRNA expression of collagen Ⅰ , collagen Ⅲ, CTGF and TGF-β1, is up-regulated. HGF has anti-fibrosis function and the down-regulation of its mRNA expression in patients with RHD may be an important factor in the initiation or maintenance of AF.
