Objective To study the correlation between chronic infection of three microorganism and coronary artery lesion by detecting their antibodies and high-sensitivity C-reactive protein(hs-CRP) .Methods According to their severity of coronary artery lesion ,patients enrolled in the study were divided into 4 groups ,including normal control group(n=37) ,single-branch lesion group (n=36) ,double-branch lesion group(n= 30) and multi-branch lesion group(n= 29) .Serum Chlamydia pneumonia IgG(CPN-IgG) ,Helicobacter pylori IgG(HP-IgG) ,human cytomegalo virus IgG (HCMV-IgG) and hs-CRP tests were performed .Results CPN-IgG ,HP-IgG ,HCMV-IgG positive rates and the mixed infection positive rates increased sequentially from normal group to multi-branch group .There were significant differences of CPN-IgG ,HP-IgG ,HCMV-IgG and hs-CRP contents among the 4 groups (P<0 .05) .Logistic analysis showed that CPN-IgG ,HP-IgG and HCMV-IgG were dangerous factors to coronary artery lesion . Conclusion Chronic infections of three microorganism might affect the development of coronary artery lesion .

Objective To investigate the effects of HRT on endometrium of postmenopausal women.Methods 53 cases of postmenopausal vaginal bleeding(PMB),from Jan,1995 to Dec.1999 treated by HRT were selected to make a comparison with the other 60 cases of PMB without HRT during the corresponding period.Both groups had integrated ultrasonic and pathological data and getting information from succedent inspection.Results The rates of the endometrial carcinorma incidences that happen in the HRT group were non-HRT group were 5 66% and 8 33% respectively,which had little significant.While the occurring rates of inflammation were 1 89% and 16 67% respectively,a significant difference between the both existed here.Conclusions With HRT,the various symptoms of post menopause women can be efficiently improved.It is safe and effective in short term.But a close monitoring is absolutely necessary for its long term effect(5 years or more).

AIM:To investigate the aberrant methylation in the promoter of p16 in plasma,sputum,bronchoalveolar lavage fluid(BALF),pleural effusion and biopsy specimens from suspected lung cancer patients and to evaluate the clinical significance in the early diagnosis of lung cancer.METHODS:Using methylation specific PCR(MSP)for the detection of promoter methylation of p16 gene in plasma,sputum,BALF,pleural effusion and biopsy specimens from suspected lung cancer patients.RESULTS:Of the 67 cases of suspected lung cancer patients,42 were proved by pathology.The positive percentages of p16 gene promoter methylation of the lung cancer patients are as follows:52.4%(22/42)in plasma,47.6%(20/42)in sputum,59.5%(25/42)in BALF,71.4%(10/14)in pleural effusion and 61.9%(26/42)in biopsy specimens,respectively;while promoter methylation in p16 gene was found only one in plasma and one in pleural effusion in 25 patients with various benign lesions(P0.05).CONCLUSION:Detection of the aberrant methylation in the promoter of p16 gene in plasma,sputum,BALF,pleural effusion and biopsy specimens from lung cancer patients by MSP method is a kind of rising technology with development potential for lung cancer early diagnosis.

BACKGROUND: The effect of Chinese traditional medicine 3'-meisoindigo as well as indimbin derivatives on normal immunocytes is less reported while it is used for antitumor.OBJECTIVE: To investigate the effect of 3'-meisoindigo on the proliferation of the splenocytes and thymocytes of C56BL/6 mouse.DESIGN, TIME AND SETTING: The randomized cytopathology observation was performed between August 2007 and January 2008 at Zhujiang Hospital of Southern Medical University, Guangzhou, Guangdong Province, China.MATERIALS: C57BL/6 mice, clean, male, 6-8 weeks old, weighing (20±2) g.METHODS: The thymus gland and the spleen of C57BL/6 mice were ground to get the single-cell suspension and cells were treated by 5, 10, 15, 20 and 25 μ mol/L 3'-meisoindigo,respectively. Cells without any drug treatment were used as blank control and cells treated by concanavalin A were used as positive control.MAIN OUTCOME MEASURES: Proliferation of splenocytes and thymocytes detected using MTT method; IL-12 activity in culture supematant detected using ELISA method; the cell cycle, apoptosis rate, cell death rate and intracellular reactive oxygen species level detected using flow cytometry; the mRNA level of bcl-2 and cdk2 detected using reverse transcription-polymerase chain reaction; the expression rate of Bcl-2, CDK2 and Bax detected using fluorescence microscope.RESULTS: After treating for 24 hours, 15, 20 and 25 μ mol/L 3'-meisoindigo can significantly inhibit the proliferation of thymocytes and splenocytes (P < 0.05) and the inhibition was dose-dependent and time-dependent. Cells resumed proliferation after removing the 3'-meisoindigo, although they had been treated by 25 μ mol/L 3'-meisoindigo for 72 hours. The secretion of IL-12 was markedly reduced in all 3'-meisoindigo groups versus control groups at each time point (P < 0.05). 15 μ mol/L 3'-meisoindigo could decrease the mRNA expression level of apoptosis-related protein bcl-2 and cyclin cdk2 gene, decrease the expression level of BCL-2 protein and CDK protein, increase Bax expression level, decrease Bcl-2/Bax ratio markedly and started apoptosis.15 μ mol/L 3'-meisoindigo arrested cells in the G2/M stage of the thymocytes and splenocytes, and intracellular reactive oxygen species level elevated dose-dependently and time-dependently.CONCLUSION: In certain concentration range, 3'-meisoindigo can reversibly inhibit the proliferation of thymocytes and splenocytes of C57BL/6 mouse and can inhibit IL-12 secretion in parallel, and start apoptosis.

AIM: To explore whether the inhibitory effect of triptolide on IL - 1β production by PBMC is asso ciated with IL - 1β gene polymorphisms. METHODS: IL - 1β gene polymorphism was analyzed in 31 healthy volunteers. From genomic DNA, the C - T polymorphism at IL - 1 β - 511 was typed by PCR - RFLP. Meanwhile the IL - 1 β was also measured in the supernatants of the cultured and stimulated peripheral blood mononuclear cells (PBMC) by ELISA. RE SULTS: After LPS stimulation in PBMC cultures of healthy subjects, the secretion levels of IL - 1 β in 9 volunteers who carried IL - 1β -511 T/T genotype were higher than in volunteers who are not T/T genotype (P ＜0.05). Triptolide sup pressed the production of IL - 1β significantly in LPS - treated human PBMC carried C/C and C/T genotype ( P ＜ 0.05 ), but this significant inhibitory effect of triptolide was not seen in T/T genotype ( P ＞ 0.05 ). CONCLUSION: The gene polymorphism at IL - 1β - 511 was related to the production of IL - 1β, and the inhibitory effect of triptolide on the produc tion of IL - 1β was different in C/C, C/T, T/T genotype of IL - 1β -511, which may be one of the reasons for the phe nomenon that people respond differently to triptolide.

Objective To investigate the clinical efficacy of selenium-enriched garlic on patients with oligoasthenospermia.Methods From October 2012 to August 2013,160 patients with oligoasthenospermia were randomly divided into two groups.Treatment measurements for 80 patients in the control group included oral treatment of Shengjing capsules 1.6 g/time Tid,Vit-E soft capsules 100 mg/time Tid,and took Vit-B 0.2 g/time Tid,for three months.Treatment measurements for 80 patients in the treatment group included oral treatment of selenium-enriched garlic in addition to all the above,for three months.We observed the changes of the semen quality indexes (including semen volume,sperm density,motility,survival rate,etc.) of two groups before and after treatment.Results Sperm quality was improved after treatment in both groups.All indices in the treatment group were significantly higher than those in the control group (P ＜ 0.05).Conclusions For patients with male infertility caused by oligoasthenospermia,The use of combination of selenium-enriched garlic and Shengjing capsules can improve the quality of sperm.

Objective The aim of the study is to discuss the application value of HA330-Ⅱ microporous resin plasma adsorption in liver support and the treatment of complications of chronic severe hepatitis.Methods The treatment group has 41 patients of chronic severe hepatitis B,which were treated for 3 times by HA330-Ⅱ microporous resin plasma adsorption.We compared the improvement of symptom,the fadeamay and rebound of icterus,the change of hepatic encepllalOpathy and brain oedema,the change of blood routine test,blood ammonia level,blood-gas analyzing result,cruor index,inflammatory factors(IL-1b,IL-6,IL-8,TNF-α,etc.),side effects,etc.The control group has 37 patients of the same type.Results HA330-Ⅱ microporous resin plasma adsorptioin is partly effective for the improvement of symptom,improvernent rate 43.9%,and without new discomfort due to the treatment. It obtains some extent fadeaway of icterus which has a low rebound,reduce TBiL(17.83±3.65)% and there be no statistics difference of rebound.It lowers the blood ammonia level and improves the hepatic encephalopathy and brain oedema.It absorbs cytokine IL-8.Atthough it has some extent effect to albunfin,blood paltelet,haemoglobin.Conclusion HA330-Ⅱ microporous resin plasma adsorption has good application value in the treatment of chronic severe hepatitis B.It is a new treatment choice of severe hepatitis.

Objective To investigate the expression level of peripheral blood mononuclear cell(PBMC)human leucocyte anti-gen G(HLA-G)in patients with hepatocellular carcinoma(HCC).Methods The HLA-G mRNA in PBMC from 44 patients with HCC,21 patients with liver cirrhosis and 40 healthy subjects were measured by reverse transcription real time fluores-cent relative quantitative PCR.Results HLA-G mRNA expression level were 1.71±0.39,1.05±0.38 and 1.01±0.47 in HCC group,liver cirrhosis group and healthy control group respectively.HCC group was higher than the other two groups, the difference was statistically significant (F=33.657,P<0.001).The survival rate of HCC patients in HLA-G mRNA high-expression group was lower than HLA-G mRNA low-expression group,the difference was statistically significant (χ2=5.972,P=0.015).Conclusion PBMC HLA-G mRNA in HCC was closely correlated with tumorigenesis.It can proviede a novel diagnosis and research tool for HCC.

Objective To assess the effect of magnetic resonance imaging (MRI) on preoperative staging of endometrial carcinoma and depth of myometrial invasion. Methods Fifty-six patients with endometrial carcinoma were underwent conventional and contrast-enhanced MR scan. The results of pathology , MRI examinations of preoperative staging and depth of myometrial invasion were assessed and compared. Results The accurate rate of MRI diagnosis in preoperative staging diagnosis was 82.1% (46/56) and the consistency check as Kappa was 0.771 (P < 0.001). Conclusions MRI may be a good diagnostic tool with high sensitivity and specificity for assessing myometrial invasion and detecting tumour extension and should be considered as a routine part of the preoperative assessment in all patients with endometrial carcinoma.
