ObjectiveToobserve the killing effect of herpes simplex virus thymidine kinase/ganciclovir (HSV tk/GCV) suicide gene therapy system on human pancreatic carcinoma cell line. MethdsHSV tk gene was constructed into a retroviral vector pDOR neo. The recombinant plasmid pDOR tk neo was transfected into the retroviral packaging cell PA317. Finally HSV tk gene was transferred into human pancreatic cancer cell line PC 2. The sensitivity to GCV and bystander effect of PC 2/tk cells was tested in vitro. Antitumor effects were also observed after the administration of GCV in nude mice bearing tumor derived from PC 2/tk cells. ResultsHSV tk gene was successfully integrated into host cell. The killing effect of GCV on PC 2/tk cells was in a dose depandent and time depandent manners. The PC 2/tk cells showed a significant "bystander effect". Intraperitoneal injection of GCV postponed the formation of the implanted tumors. ConclusionPancreatic cancer cells expressing HSV tk can be effectively killed by GCV both in vitro and in vivo.

BACKGROUND:Although the preparation of bone tissue engineering scaffolds can achieve satisfactory results by solvent casting/particulate leaching, in situ molding method, electrospinning, phase seperation/freeze drying, gas foaming, there are stil some deficiencies in the accuracy, pore uniformity, spatial structure complexity, personalized stents.
OBJECTIVE:To prepareβ-tricalcium phosphate bone tissue engineering scaffolds using 3D printing.
METHODS:Drug-loadedβ-tricalcium phosphate scaffolds were prepared with 3D printing, and the structure was observed to measure its porosity and mechanical strength. The scaffold was immersed in simulated body fluid for 15 weeks to observe the quality change. The scaffold was co-cultured with rat bone marrow mesenchymal stem cells for 7 days to observe celladhesion and morphological changes. Rat bone marrow mesenchymal stem cells were cultured in extracts of drug-loadedβ-tricalcium phosphate scaffold and low-glucose Dulbecco's modified Eagle’s medium containing 15%fetal bovine serum for 24, 48, and 72 hours, to determine the absorbance values and cytotoxicity grading, respectively. Meanwhile, the cells were subjected to osteogenic culture for 1 week, and
the alkaline phosphatase activities in two groups were detected.
RESULTS AND CONCLUSION:The prepared scaffold showed irregular micropores, high porosity, uniform pore distribution, high pore connectivity rate, and large compressive strength. The drug-loadedβ-tricalcium phosphate scaffold degraded completely with 15 weeks, and cancellous bone defect repair was completed in the same period. Rat bone marrow mesenchymal stem cells adhered to the surface of drug-loadedβ-tricalcium phosphate scaffold and went deep into the scaffold, showing good growth and proliferation. The activity of alkaline phosphatase was also improved. These findings indicate that the drug-loadedβ-tricalcium phosphate scaffold has good biocompatibility.

The simple and effective controlled needle devices were provided for acupuncture clinical trial so that the double blind acupuncture trial could be conducted possibly. The basic structure of filiform needle was modified. The needle handle was designed as hollow, the upper 1/3 of needle body as hollow and the lower 2/3 part as solid. The upper and lower parts were linked with an elastic unit. At the same time, the needle pad was attached. With this method, the new sham-acupuncture device was designed and developed. The new sham-acupuncture device could simulate needle insertion, manipulation and the techniques of reinforcing and reducing, without inserting into the human body. This innovation lays the foundation for perfecting the clinical randomized controlled blind trial of acupuncture.

BACKGROUND: Following peripheral nerve transplantation, the time phases and law of growth associated protein 43 (GAP-43) expression in sensory neuron soma of the corresponding dorsal root ganglia remain unclear.OBJECTIVE: To investigate the expression of GAP-43 at the corresponding dorsal root ganglia following sciatic nerve transplantation in rats.METHODS: Adult male Wistar rats were randomly divided into 2 groups: control group (sham-operated group) and experimental group (nerve transplantation group). The rats were killed at 3 days, 1, 2, 4, 6, and 8 weeks after operation and the dorsal root ganglia U-s of every rat was taken for observation. The expression of GAP-43 mRNA and protein in the corresponding segments of spinal cord were detected by RT-PCR and Western-blot techniques.RESULTS AND CONCLUSION: GAP-43 mRNA showed a low level of expression in the control group, which had no obvious changes in different phases. In the experimental group, GAP-43 mRNA expression was observed remarkably in the dorsal root ganglia at the first week, reached a peak at the second week and gradually descended from the sixth week. The GAP-43 protein expression showed the same laws as that of GAP-43 mRNA. The results demonstrated that there exists damage reactivity change in the regeneration ability of neurons.
