1.Recent progress in the investigation of cytokine and diabetes retinopathy

Diabetic retinopathy is a progressive vision-threatening complication of diabetes mellitus.The characters of diabetic retinopathy in clude capillary occlusion,microcirculation disturbance and retina neovascularization near ischemic areas of the retina.The exact pathogenic mechanism is still unclear.Recent studies showed that it may be related to vascular inflammation of the retina.Cytokines can cause induction of proinflammatory and adhesion molecules and thereby increase monocyte-endothelial cell adhesion,which is now accepted as the early key event in the development of diabetic retinopathy.The functions of many cytokines such as vascular endothelial growth factor,transforming growth factor-β,basic fibroblast growth factor and ideas for medicine therapeutic to diabetic retinopathy are summarized in this review.
2.Construction of training evaluation system for laboratory animal practitioners

Objective Laboratory animal practitionres need further management through technical and capacity training.Methods Relying on the Laboratory Animal Center of University and its highly qualified faculty, a high-quality training base for laboratory animal practitioners has been set up.Results Based on the relevant laws and regulations about experimental animals, appropriate training outlines, different training books and courses were prepared and organized for different trainees.Focusing on the detailed and exact experiments, training base has put emphasis on the practical ability and operational skills.Conclusions How to regulate the training for laboratory animal practitioner by means of base training is discussed and tried out in this paper.
3.In situ histological study on apoptosis of buccal carcinoma cells induced by anticancer agents

?Objective:To study apoptosis and apoptotic mechanisms of buccal carcinoma cells(BCCs) induced by anticancer agents.Methods:34 cases of buccal carcinoma were treated with pinyangmycin or pinyangmycin combined with other anticancer agents.The samples of the lesions were examined by in situ terminal deoxynucleotidyl transferase (TDT) mediated dUTP nick end labeling (TUNEL) before and after chemotherapy.Apoptotic cells were detected in parraffin embedded buccal cancer specimens from the patients.Apoptotic index (AI) were respectively quantified in prechemotherapy and postchemotherapy specimens of each patient.Fas expression was examined by SP immunohistochemical staining in each specimen.Results:The mean of AI in the samples of prechemotherapy and in those of postchemotherapy were 0.38 and 0.91 ( P
4.Histological research of calcium phosphate biomaterials promoting tendon healing in bone tunnels

Objective To investigate the effect of osteoinductive calcium phosphate ceramics (OICPC) on tendon healing in bone tunnels. Methods Anterior cruciate ligament reconstruction with semitendonous tendon was performed in the knee joints of 30 New Zealand rabbits by suspended fixation. OICPC grains were implanted into the bone tunnel of 15 rabbits which were the experimental group and the other 15 rabbits without OICPC implants severed as the control group. Specimens were collected four weeks, eight weeks and 12 weeks after surgery. Sections were stained with H. E., Sirius Red and Masson. Histological changes in the interface tissue between the bone tunnel and the tendon graft were observed, and the morphology of interface tissue was classified according to Yamakado method. Histomorphometry was done on Masson stain sections to measure new bone formation around the grafted tendon. Results In OICPC group, histological sections obtained four and eight weeks after surgery showed statistically greater new bone formation than those in the control group. Abundant penetrating Sharpey fibers appeared at eight weeks and direct connection was found at 12 weeks in the experimental group. On the other hand, Sharpey fibers appeared only at 12 weeks in the control group. Conclusion OICPC has a capability of enhancing biological fixation of a tendon to the bone tunnel.
5.Detection of soluble serum transferrin receptor of thalassemia women during pregnancy

Objective Soluble serum transferrin receptor (sTfR) is a valuable indicator to diagnose iron deficiency anemia in pregnant women whose indexes will alter because of gestation. Thus it is worth to compare and analyze the levels of sTfR among the pregnant women with thalassemia, healthy pregnant women, and the pregnant women with iron deficiency anemia. Methods ?-thalassemia and ?-thalassemia were detected employing with the percentage of hemolysis and gene assay in the pregnant women randomly selected from those aged 24~30, 20~24 weeks of gestation, and with no other complications. The ratio of OD413nm of HbA2 to OD413nm of HbA was used to distinguish from the two kinds of thalassemia. Then the genetic mutation or deletion of ?-thalassemia patients and nucleotide mutation of ?-thalassemia patients were detected with thalassemia detection kit. The value of sTfR in serum of different samples was measured with ELISA kit. Results 20 pregnant women were diagnosed as thalassemia by the percentage of hemolysis higher than 63%. Among them, 10, 9 and 1 women were judged as ?-thalassemia, ?-thalassemia and ?,?-thalassemia, respectively. 7.Expression and roles of miR-155-5p in esophageal squamous cell carcinoma

Objective Studies show that miRNA plays an important role in regulating the development, metastasis, inva-sion, and angiogenesis of carcinoma.This study aimed to investigate the expression of miR-155-5p in esophageal squamous cell carci-noma ( ESCC) and its role in the occurrence and progression of ESCC. Methods We collected 20 surgical specimens of ESCC and another 20 from the adjacent normal tissue at least 5 cm from the lesion and determined the expression of miR-155-5p in the tissues by qPCR.We transfected the miR-155-5p mimic, inhibitor and negative control plasmid into the Eca109 cells, measured the proliferation and apoptosis of the cells by CCK8 assay and flow cytometry, and examined their migration using the scratch test. Results The ex-pression of miR-155-5p was significantly up-regulated in the ESCC tissue as compared with that in the normal tissue (529.42% vs 100%, P<0.05).The scratch test showed a markedly increased migration of the cancer cells when miR-155-5p was overexpressed. No remarkable differences were observed in the apoptosis rates of the cells transfected with the miR-155-5p mimic ([5.43 ± 3.09]%), inhibitor ([5.28 ±1.98]%), and negative control plasmid ([5.67 ±1.99]%) (P<0.05). Conclusion The ex-pression of miR-155-5p is significantly up-regulated in the ESCC tissue, which may act as a potential maker for the diagnosis of ESCC. The up-regulated expression of miR-155-5p does not significantly in-fluence the proliferation and apoptosis but enhances the migration of Eca109 cells, which may be related to early metastasis of ESCC.
8.Death among children under five years of age in Huzhou City from 2012 to 2021

Objective:
To analyze the death among children under 5 years of age in Huzhou City, Zhejiang Province from 2012 and 2021, so as to provide insights into reduction of mortality among children.
Methods:
The mortality surveillance data among children under 5 years of age in Huzhou City from 2012 to 2021 were collected from Children Death Report Cards and Surveillance Report among Children under 5 Years of Age, including gender, place of residence, date of death and death diagnosis. The trends in mortality and cause of death were analyzed among children under 5 years of age in Huzhou City from 2012 to 2021.
Results:
A total of 1 262 deaths occurred among children under 5 years of age in Huzhou City from 2012 to 2021, with mean annual mortality of 4.39‰, and the mortality appeared a tendency towards a decline (χ2trend=132.695, P<0.001). A total of 899 infants died, with mean annual mortality of 3.13‰, and 363 children at ages of 1 to <5 years died, with mean annual mortality of 1.26‰. The mortality appeared a tendency towards a decline among both infants (χ2trend=117.778, P<0.001) and children at ages of 1 to <5 years (χ2trend=19.201, P<0.001). A total of 724 local children died, with mean annual mortality of 3.33‰, and there were 538 deaths among floating children, with mean annual mortality of 7.65‰. The mortality appeared a tendency towards a decline among both local (χ2trend=43.728, P<0.001) and floating children (χ2trend=94.038, P<0.001). The five most common causes of death included preterm birth or low birth weight (207 deaths, 16.40%), drowning (155 deaths, 12.28%), accidental asphyxia (138 deaths, 10.94%), other congenital abnormalities (126 deaths, 9.98%), and congenital heart diseases (113 deaths, 8.95%).
Conclusions
The mortality appeared a tendency towards a decline among children under 5 years of age in Huzhou City from 2012 to 2021, and preterm birth or low birth weight was the predominant cause of death.
9.Comparison of the efficacy of supraclavicular approach and infraclavicular approach in ultrasound-guided brachial plexus block

Objective To compare the efficacy of supraclavicular approach and infraclavicular approach in ultrasound-guided brachial plexus block(BPB).Methods One hundred and twenty patients,ASA Ⅰ-Ⅲ,aged 18-80 yr,scheduled for upper extremity surgeries of both gender,were randomized into two groups(n =60):supraclavicular group(group SCB)and infraclavicular group(group ICB).The anesthetic mixture consisted of ropivacaine 0.375% and lidocaine 1% in equal volumes with epinephrine 1∶200 000,the total dose was 0.5 ml/kg.The block performance time,duration of anesthesia and success of anesthesia(surgery was accomplished without supplementary block)were recorded.A blinded observer assessed pinprick sensory block in the seven distal nerve territories(axillary,radial,musculocutaneous,median,ulnar,medial antebrachial and medial brachial cutaneous nerves)every 5 min up to 30 min after injection.Success of nerve block,side effects and complications were recorded during and after operalion.Results Group ICB was superior in success rate of anesthesia and nerve block of ulnar,medial antebrachial and medial brachial cutaneous nerves,the rate of parasthesia was lower and the block performance time was longer compared with group SCB(P < 0.05).No major complications occurred in both groups.Conclusion Under ultrasound guidance,infraclavicular BPB is superior to supraclavicular approach.
10.Effect and mechanism of chlorogenic acid in allergic rhinitis mice

Objective To study the effect and mechanism of chlorogenic acid in allergic rhinitis(AR) mice.Methods The model of AR mice was induced by intraperitoneal injection of adjuvant suspension on every other day for 7 times, and then by nasal drip of 10% OVA for 10 days.After administration with 50, 100, 200 mg/kg of chlorogenic acid for 30 min, AR mice were attacked locally for 10 days.Behavior analysis were observed and the effect of chlorogenic acid on the contents of cytokines, IgE and histamine by ELISA in AR mice were determined.Results Chlorogenic acid decreased the number of nose-scratching events in 10min, the weight of nosal draingage, the index of spleen, the content of IL-4 in nasal lavage fluid ( NLF) and the contents of cytokines, IgE and histamine in serum; while increased the index of thymus, the content of IFN-γin NLF, the contents of IFN-γin serum and the rate of IFN-γ/IL-4.The data showed a statistically significant up-regulation of IFN-γmRNA expressions and down-regulation of IL-4 mRNA, IL-5 mRNA, IL-10 mRNA expressions after adiminstration of chlorogenic acid.Conclusion Chlorogenic acid might exert the effect in AR mice through regulating the balance of Th1 and Th2.
