Objective Increased morbidity of auto-immune disease in senescent individual might be related with the defects of apoptosis after stimulation or induction. Changes of apoptotic features in splenic T lymphocytes were studied in senescent mice. Methods Flow cytometry was used to study the rates of apoptosis by analysing sub-G 1 peak. DNA ladder was observed by agarose gel electrophoresis. Free calcium levels in both cells were detected by Fluo-3 loading. Bcl-2 protein levels were detected by Western blot. Results After co-stimulation with IL-2/ConA, flow cytometry showed that average apoptotic percentage of old T cells was 38.3% ?10.3%,significantly lower than that of young ones (58.6% ? 4.0%, P
3.Comparative study of healthy vertebral artery blood flow from plain and plateau region

Objective To conduct comparative study of healthy vertebral artery (VA) blood flow of subjects from plain and plateau region using color Doppler ultrasound (CDUS) measurement,and to investigate the influence of plateau on blood flow of VA.Methods One hundred and thirty-five subjects from plain region and eighty from high altitude region were recruited.Time averaged maximum velocity and inradium (D) of VA were measured with CDUS technology.Unilateral VA blood flow and total blood flow were calculated.Peripheral blood viscosity measurement was carried out by testing low cut,mid cut,high cut,plasma viscosity and red blood cell count (RBC),hemoglobin concentration (HGB),hematocrit (HCT).Results VA diameter,blood viscosity,RBC,HGB,and HCT of plateau group are all higher than that of the plain group (P<0.05),while the total blood flow was lower than that of plain group and the value of male was higher than that of female (P<0.05).The inradiums of right side VA were larger than that of the left side (P<0.05) in plain group,while no difference was observed in plateau group (P>0.05).Conclusions Long term (>10 years) living in plateau hypoxia (altitude>3 000 m) environment can reduce the average flow velocity and the blood flow of VA.
4.Effect of inhibition of Akt phosphorylation on apoptosis of non-small cell lung cancer H1299 cells with wild type EGFR and KRAS induced by tumor necrosis factor related apoptosis inducing ligand

Objective To explore the effect of inhibiting Akt phosphorylation on tumor necrosis factor related apoptosis inducing ligand (TRAIL)-induced apoptosis of human non-small cell lung cancer (NSCLC) H1299 cells with wild type EGFR and KRAS.Methods The TRAIL-induced apoptosis was examined by the Annexin V-FITC/PI.The expressions of TRAIL-activated Akt phosphorylation and p-Akt were measured by Western blot.After cells were treated with LY294002, an inhibitor of PI3K-Akt pathway, Annexin V-FITC/PI and Western blot were used to analyze the alteration of TRAIL-induced apoptosis and Akt phosphorylation, respectively.Results H1299 cells were not sensitive to TRAIL-induced apoptosis.When TRAIL concentration was 100 ng/ml, the apoptosis rate of the test group was significantly higher than that of the control group [(15.06±1.29) % vs (3.56±0.50) %, t =66.953, P =0.000].When TRAIL concentration was 500 ng/ml, the difference was not statistically significant compared with apoptosis rate of 100 ng/ml TRAIL group [(18.65±2.09) % vs (15.06±1.29) %, t =2.423, P =0.136].The expression level of Akt phosphorylation in H1299 cells was increased by TRAIL in a time-dependent way.When cells were pretreated with LY294002, TRAIL-induced Akt phosphorylation was suppressed to baseline level.At the same time, the apoptosis rate in LY294002-treated group was significantly higher than that in TRAIL group [(41.65±4.62) % vs (15.82±0.61) %, t =39.028, P =0.001].Conclusions TRAIL-induced Akt phosphorylation can antagonize TRAIL-induced apoptosis.Inhibition of Akt phosphorylation can significantly enhance the sensitivity of NSCLC H1299 cells with wild type EGFR and KRAS to TRAIL-induced apoptosis.
5.Expression of pyruvate kinase M2 in non-small cell lung cancer and its correlation with radiation sensitivity

Objective:To investigate the expression level of pyruvate kinase M2 (PKM2) in tissues of non-small cell lung cancer (NSCLC) patients and the correlation between PKM2 expression level and radiation sensitivity. Methods:A total of 45 NSCLC pa-tients were chosen and treated with radiotherapy for two months after surgery. The patients were classified into four groups based on the curative effect. The mRNA expression levels of PKM2 in tumor and the homologous paraneoplastic tissues of NSCLC patients were de-tected prior to radiotherapy using real time-polymerase chain reaction (RT-PCR), and the protein expressions of PKM2 in the tumor and paraneoplastic tissues of NSCLC patients were detected with Western blot analysis and immunohistochemical method. The mRNA and protein expression levels of PKM2 in the tumor tissues of different groups were detected with RT-PCR and Western blot assays. Re-sults:The effective rate of radiotherapy for 2 months is 44.8%in NSCLC patients. The expression level of PKM2 is significantly high-er in tumor tissues than in homologous paraneoplastic tissues of NSCLC patients and is negatively correlated with the curative effect of radiotherapy. Conclusion:The expression level of PKM2 is significantly higher in tumor tissues than in the paraneoplastic tissues of NSCLC patients. Patients with lower PKM2 expression level are more sensitive to radiotherapy.

Objective To investigate the MR postprocessing workstation on PC level.To discuss the hardware enviroment suitble for the lab MR postprocessing and the correspongding clinical diagnostic value. Methods MR diffuison-weighted imaging and routine MRI were performed in 6 normal adults.The ADC and AI parameter images were rebuilt by using Perfect-Tech (version 2.0) software on the market saled PC computer. Results The process and analysis could be performed sucessfully on the the market saled PC computer. On the ADCav images,the boundary between the grey matter(GM)and white matter(WM) was blurring and showed the similar intensity.The anistropy in white matter was averaged out on the ADCav images.The ADC value was (7.59?1.12)?10 -4 mm 2/s in WM and (10.50?4.34)?10 -4 mm 2/s in GM. On the AI images,there was sharp boundary between GM and WM,with hypointensity in GM and hyperintensity in WM. The anistropy contrast was enhanced on AI images. AI was 0.20?0.09 in WM and 0.09?0.07 in GM with statistically significant difference between them(t=4.13,P
7.Robotic and computer assisted orthopaedic surgery

This paper introduces the applications of Computer Assisted Orthopaedic Surgery (CAOS), including pre-operative planner, surgical simulator, intra-operative navigation systems and medical robotics. Basic principals of surgical navigation are also discussed. In view of the high precision requirement for navigation assisted procedures, there exists a definite need for development of medical robotics to carry out these procedures. We have a brief review on the different robot systems and present the surgical navigation arm developed in the Chinese University of Hong Kong. The laboratory test results of navigation guided distal targeting of intramedullary nail with or without surgical navigation arm showed the better repeatability and precision when the robot arm was used. So far the surgical navigation arm has been successfully applied in clinical practice, including distal targeting of intramedullary nailing; percutaneous sacro-iliac screw and percutaneous trans-iliac screw in pelvic fractures and hip screw fixation for fracture of femoral neck. To conclude, medical robotics is an essential component of the future CAOS.
8.S632A3 promotes LPS-induced IFN-beta production through inhibiting the activation of GSK-3beta.

LPS stimulation of macrophages production of IFN-beta plays a key role in innate immunity defending the microbial invasion. In this study, the effect of S632A3 promoting LPS-induced IFN-beta production and the underlying mechanism were investigated, mRNA level was measured by real-time PCR, cytokine production was determined by ELISA, GSK-3beta activity was investigated by kinase assay, protein phosphorylation and expression were evaluated by Western blotting. The results revealed that S632A3 significantly augmented IFN-beta production by LPS-stimulated macrophages. S632A3 inhibition of the activation of GSK-3beta, reduced the threonine 239 phosphorylation of transcription factor c-Jun but increased the total level of c-Jun in LPS-stimulated macrophages. Moreover, small interfering RNA-mediated knockdown of c-Jun level abrogated the ability of S632A3 to augment IFN-beta. The study thus demonstrates S632A3 being a new anti-inflammation lead compound and provides a molecular mechanism by which S632A3 promoted LPS-induced IFN-beta production in macrophages through inhibiting the activation of GSK-3beta.
9.Haploidentical hematopoietic stem cell transplantation for leukemia in 23 cases

BACKGROUND:Al ogeneic hematopoietic stem cel transplantation (HSCT) is one of the effective methods in the treatment of leukemia. The haploidentical HSCT is an option for the patients who need a HSCT without a human leukocyte antigen (HLA)-matched donor. OBJECTIVE:To study the clinical efficacy of HLA-haploidentical HSCT on leukemia and its complications. METHODS:A total of 23 patients (4 cases of acute lymphoblastic leukemia, 12 of acute myelogenous leukemia, and 7 of chronic granulocytic leukemia) who had been treated with HLA-haploidentical HSCT from November 2007 to March 2015 were enrol ed. Conditioning regimen I was set as cyclohexyl nitrosourea+cytarabine+busulfan+cyclophosphamide; regimen II as cyclophosphamide+total body irradiation;regimen III as fludarabine+cytarabine+busulfan+cyclophosphamide; and regimen IV as busulfan+cyclophosphamide. Cyclosporin A, mycophenlate mofetil, antithymocyte globulin and methotrexate were used to prevent graft-versus-host disease (GVHD). Hematopoietic remodeling, complications and prognosis were observed in al patients undergoing HLA-haploidentical HSCT. RESULTS AND CONCLUSION:Of the 23 patients, 22 achieved reconstitution of the granulocyte series, and 19 achieved reconstruction of the megakaryocyte series. Additional y, there were 7 cases of acute GVHD and 4 of chronic GVHD. Transplant-related mortality was 22%(5/23) within 100 days post transplantation including graft failure, acute GVHD, intracranial hemorrhage and disseminated infections. There were 14 cases of disease-free survival from 100 days to 24 months post transplantation, 2 cases of death due to GVHD and fungal infection, or recurrence and chronic GVHD, and 2 cases of recurrence under treatment. These findings indicate that HLA-haploidentical HSCT is an effective approach for the treatment of patients with leukemia, which is worth further investigation in clinical practice.
10.Chemical constituents from the bioactive extract of Chuanminshen violaceum

AIM: To study the chemical components from the bioactive extract of Chuanminshen violaceum Sheh et Shan in Sichuan province. METHODS: The isolation and purification of this extract were conducted by means of silica gels column chromatography.The structures of the compounds were elucidated based on their physical,chemical features and spectrum data. RESULTS: 8 compounds were isolated from the extract.They were elucidated as hexadecanoic Acid(A),2,6-dimethoxybenzoic acid(B),succinic acid (C),combination of sigmasterol and sitosterol(1(∶)1)(D),chrysophanol(E),8-hydroxy-5-methoxypsoralen(F),4-hydroxy-3-methoxybenzoic acid(G) and stigmasteryl 3-O-?-D-glucopyranoside(H). CONCLUSION: Compounds B,C,E,G were isolated from Chuanminshen violaceum for the first time.
