2.Detection of ASC in spleen cells and GALT after immunization with two bivalent Shigella vaccines in different administration rout in mice

Abstract Objective:FSM-2117 and FS-5416 are two bivalent hybrid strains of S.flexneri and S.sonnei, constructed by this laboratory-The FS-5416 expresses four invassive plasmid antigens(IpaA、IpaB、IpaC and IpaD) and has a good contact heamolysis activity(CHA+ ), butFSM-2117 hasn' t. To observe the immunogenicity of Shigelk vaccines through three different mucosal administration rout, the changes of ASCin the spleen and Peyer's patch(PP) , mesenteric lymph nodes(MLN) lymphocytes are detected.Methods:BALB/c mice were divided intothree groups, 20 mice per group. Mice were immunized respectively with the Ipa+ or Ipa- vaccines(4 x 10~7 CFU) three times with an intervalof two weeks by intranasal、intragastric or intraintestinal administration routs. The spleen , PP , MLN lymphocytes were isolated of seventh dayat random after immunization. An BA-EIISASPOT was done to account the numbers of ASC.Results:The numbers of SIgA and SIgG ASC ofspleen , PP , MLN lymphocytes of intranasal and intraintestinal group were significantly increased . Significant difference were only observed inthe number of spleen lymphocytes SIgA and SIgG ASC of intranasal group between Ipa+ and Ipa- . Conclusion:Two bivalent Shigelk vaccinescan induce spleen and GALT immunity reaction by nasal or small intestinal mucosal in a low dosage compared with intragastric rout (about 1/20). Ipa can significantly increased the number of spleen lymphocytes SIgA and SIgG ASC of intranasal group.
3.Enhancing effect of DHEA ON DbcAMP-induced cell differentiation

Aim To observe whether DHEA has enhancing effect on DbcAMP -induced differentiation of NG108-15 cells, including neurite outgrowth, and study its possible mechanisms. Methods NG108-15 cells (a h ybrid cell line of mouse neuroblastoma and rat glioma) were used as a substitute for primary culture neuron in vitro. The morphology of NG108-15 cells was o bserved and neurite outgrowth was determined in an inversed microscope after treatme nt with various drugs. Gelatin-substrate gel electrophoresis was used to detect gelatinases (MMP-9 and MMP-2). Results ① DHEA and DbcAMP inhibited NG108-15 proliferation.②DHEA had enhancing effect on the promoting activity of neuronal differentiation and neurite outgrowth by DbcAMP. DbcAMP could increase neurite elongation of NG108-15 cells. Compared with this, the combined treatment with DHEA and DbcAMP significantly enhanced the neurite outgrowth of NG108-15 cells, including neurite length and numbers of cells with neurite, in a DHEA dose-dependent manner. ③ MMPs were involved in neuronal differentiation. DbcAMP induced the increase in MMP-9 and MMP-2 activities and such elevation was enhanced by DHEA in a dose-dependent manner. Conclusion DHEA enhances the effect of DbcAMP in promoting the neurite outgrowth of NG108-15 cells, which might be related to the increase in MMP-9 and MMP-2 activities.
5.Progress in ubiquitin, ubiquitin chain and protein ubiquitination.

Protein ubiquitination is one of the most important and widely exist protein post-translational modifications in eukaryotic cells, which takes the ubiquitin and ubiquitin chains as signal molecules to covalently modify other protein substrates. It plays an important roles in the control of almost all of the life processes, including gene transcription and translation, signal transduction and cell-cycle progression, besides classical 26S protesome degradation pathway. Varied modification sites in the same substrates as well as different types of ubiquitin linkages in the same modification sites contain different structural information, which conduct different signal or even determine the fate of the protein substrates in the cell. Any abnormalities in ubiquitin chain formation or its modification process may cause severe problem in maintaining the balance of intracellular environment and finally result in serious health problem of human being. In this review, we discussed the discovery, genetic characteristics and the crystal structure of the ubiquitin. We also emphasized the recent progresses of the assembly processes, structure and their biological function of ubiquitin chains. The relationship between the disregulation and related human diseases has also been discussed. These progress will shed light on the complexity of proteome, which may also provide tools in the new drug research and development processes.

6.Survey of the relationship between pathogen strains distribution and nusocomial infection

Objective To ostimate the distribution of microorganism strains isolated from hospital ward cir-curmstanee and clinical specimens, and then proceed to inquire into their interrelation to nosocomial infection, provi-ding evidence for preventing and reducing nosocomial infection. Methods Specimens were collected from hospital circumstance and each clinical laboratory, and then made a bacterial identification and pathogen strains survey. Re-sults There was a closed correlation between the microorganism strain isolated from hospital circumstance and clini-cal infection pathogen(86%). Conclusion A series of disinfection management measures should be made and for-mulated so as to reduce the hospital infection rate as far as possible.
7.Preparation and in vitro targeting of follicle stimulating hormone polypeptide modified nanoparticles

Objective To prepare follicle stimulating hormone (FSH) polypeptide modified nanoparticles (NP) in order to achieve specific ovarian tumor targeting. Methods Expression of FSH receptor protein in human liver cancer and ovarian cancer cell lines BEL-7402, SKOV-3 and Caov-3 was detected by immunocytochemistry. The polypeptide fragment of FSH β 81 -95 amino acids (FSHL81-95)was synthesized and covalently coupled to NP. The specific binding of FSHL81-95 and FSHL81-95-NP was examined by fluorescence microscopy and flow cytometry. Results BEL-7402 and SKOV-3 cells were negative for FSH receptor staining, while Caov-3 celia were positive. The diameters of NP were about 100 nm, with a Zeta potential of -25 mV or so. Caov-3 cells showed a more specific interaction with FSHL81-95-NP than SKOV-3 cells (4. 17 ± 0. 86 and 2. 30 ± 0. 21 ; P < 0. 05). The uptake of FSHL81-95-NP was more than NP in Caov-3 cells (4. 17 ± 0. 86 and 0. 41 ± 0. 32 ; P < 0. 05 ). FSHL81-95-NP showed a selective targeting at Caov-3 cells compared with control NP. Conclusion FSH polypeptide modified NP could selectively target ovarian cancer cells expressing FSH receptor, which might contribute to specific endocytosis mediated by FSH receptor.
8.Characteristics and establishment of ischemic tolerance rat models with hypoxic preconditioning

BACKGROUND: We can investigate mechanism of endogenous neuroprotection in rat cerebral hypoxic tolerance trial. OBJECTIVE: To observe the characteristics of cerebral hypoxic tolerance in rat models with cerebral hypoxic preconditioning. DESIGN: Randomized controlled observation. SETTING: Department of Neurology, China-Japanese Friendship Hospital, Jilin University. MATERIALS: The experiment was performed in the Basic Animal Experimental Center, China-Japan Friendship Hospital, Jilin University from April 2003 to April 2004. Inbred line healthy Wistar rats, of either sex, with the body mass of 200-300 g, were randomly assigned into normal control group (n=6), sham operation group (n=6), ischemic control group (n=20), hypoxic preconditioning (3 hours, 8% O2 and 92% N2) plus ischemic group (n=60) (according to different hypoxic phases, there were 5 time phases: 30 minutes, 1, 3, 5 and 6 hours with 12 rats in each time phase), hypoxic preconditioning group (n=18) [according to different hypoxic phases, there were 3 time phases: 1, 3 and 5 hours with 6 rats in each time phase, 3 rats received TTC staining and 3 rats received hematoxylin and eosin (HE) staining]. METHODS: ①Hypoxic preconditioning: Firstly, natrica calx was put into closed glass container to absorb CO2 and O2, secondly, mixed gas of 8% O2 and 92% N2 was input, and then animals were put into the container, 3 rats each time. Temperature and humidity were kept steadily. ②Permanent ischemic middle cerebral artery rat models were established. ③The models were determined with a series in procedures: neurological score, infarcted volume evaluation, pathological sample preparation, immunohistochemical staining, imaging analysis and so on. ④The data were compared in groups with variance analysis.MAIN OUTCOME MEASURES: Changes in cerebral infarcted volume, neurological score and pathological morphology in rats of experimental group and control group. RESULTS: Neurological score in the hypoxic preconditioning (8% O2, at hours 1, 3 and 5) plus ischemic group was lower than in the ischemic control group(P<0.01). Neurological score at minute 30 and hour 6 after hypoxia (8% O2) had insignificant difference in the ischemic control group. Mean cerebral infarcted volume ratio in the hypoxic preconditioning (8% O2, at hours 1, 3 and 5) plus ischemic group was lower than in the ischemic control group(P<0.01). Mean cerebral infarcted volume ratio after hypoxia (8% O2, at minute 30 and hour 6) had insignificant difference with ischemic control group (P>0.05). CONCLUSION: Hypoxic preconditioning in rats can effectively release nerve injury induced by focal cerebral ischemia, suggesting that it has protective effect on brain. The procedure of establishing cerebral ischemic tolerance models with hypoxic preconditioning, which is simple and stable, with little injury on experimental animals, is a useful tool for studying cerebral ischemic tolerance.
9.Effect of ischemia/reperfusion on expression of endothelin in the rat ventral prostate and preventive measure

AIM:Effect of ischemia/reperfusion on expression of endothelin-1(ET-1) in the rat prostate and preventive measure were studied. METHODS:The abdominal aorta of rat was clipped briefly and repeatedly so as to treat the prostate with ischemia/reperfusion and expression of ET-1 mRNA in the ventral prostate was determined by RT-PCR.RESULTS:Expression of ET-1 mRNA in the ventral prostate was significantly increased at 1 h and 3 h after 90 min repeated ischemia/reperfusion (P<0.05), and was not significantly changed after previous treatment of Dizocilpine maleate (MK-801) (P>0.05). CONCLUSIONS:Expression of ET-1 in the prostate can be affected by repeated brief ischemia/reperfusion and it may play a role in the development of benign prostatic hyperplasia. Ischemia-reperfusion-induced ET-1 expression in the prostate of rats can be inhibited by prectreatment of MK-801.
10.Reliability and validity of the Chinese version Successful Aging Inventory

Objective To evaluate the reliability and validity of the Chinese-version Successful Aging Inventory (C-SAI).Methods The C-SAI was translated according to the Brislin translation model,and its reliability and validity was tested in 181 old adults.Results The content validity index for the scale (S-CVI/Ave) was 0.975.Five factors were extracted by principal components analysis which contributed 58.035% to the variance.The Cronbach α and split-half reliability was respectively 0.832 and 0.871 for the total scale.Conclusions The C-SAI has good psychometric quality and can be used as a measurement tool for the successful aging.
