An isocratic high-performance liquid-chromatographic method is-described for the simultaneously determining carbamazepine and its biologically active metabolite, carbamazepine-10, 11-epoxide. Serum samples containing an internal standard (10-methoxycarba-mazepine) are extracted with dichlor-methane and 4 mol ? L-1 sodium hydroxide. The organic extract are evaporated at ambient temperature under a stream of nitrogen. The residues are reconstituted in mobile phase and injected on to a re-versed-phase C18 Column (150?4. 6mm I. D). The mobile phase consists of ace-tontrile/methanol/water (50/210/260 by vol) . The UV detector is operated at 214nm. The elution times of all com-pounds are within 7 min.The least detectable concentrations of carbamazepine and carbamazepine-10, 11-epoxide are 0. 08mg ? L-1 and 0.1mg ? L-1, respectively. The average absolute recovery for carbamazepine is 96.0% and for carbamazepine-10,11-epoxide is 97.3%. Within-run CV is 3. 3%~5.7%. and between-run CV is 4.3%~9.0%. The standard curve is linear at least within 40mg ? L-1 for carbamazepine and within 20mg ? L-1 for carbamazepine-10, 11-epoxide.

Objective To discuss the variety of blood zinc,copper,iron,calcium level in children of lead poisoning Methods The stratified - random sampling method was used. Five hundred and forty children were selected in the urban areas, the rural areas and the islands. We examined blood lead level and other elements levels of these children, and analyzed the changes of these elements levels on condition of lead poisoning. Objective To study the effect of hypothyroidism on oxidative stress status in developing rat brain and to further explore the mechanism of impaired brain development caused by hypothyroidism. Methods Perinatal hypothyroidism was induced by administering propylthiouracil(PTU) solution to the dams by gavage.The oxidative stress indexes were measured in brain homogenate of normal and hypothyroid pups which were sacrificed on the 21st d after birth. Results As compared to the control,the following indexes were found to be increased in the hypothyroid group: protein carbonyl contents,thiobarbital acid reactive substances,reduced glutathione,total antioxidative capacity,activities of superoxide dismutase and glutathione peroxidase(P0.05). Conclusion Hypothyroidism during rat brain development may cause oxidative stress,which may be related to the brain damage caused by hypothyroidism.

Objective To inhibit the expression of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in human kidney tubular epithelial cell(HKC) with specific small interference RNA(siRNA),then to observe whether the expression of intercellular adhesion molecule-1(ICAM-1) on HKC could be influenced.Methods HKCs were transfected with MMP-2,MMP-9 siRNA or irrespective siRNA respectively,untransfected HKC was considered as blank control.Four groups of HKCs were stimulated with 100nmol/L phorbol myristate acetate(PMA) for 18h.Total RNA or protein of cells were extracted with Trizol and RIPA cell lysate respectively,the mRNA expression levels of MMP-2,MMP-9 and ICAM-1 were detected by real time PCR,the protein expression levels of above indexes were detected by western blotting and immunofluorescence combined with confocal laser microscopy.Results The specific siRNA could inhibit the expression of MMP-2 and MMP-9 in HKCs effectively.Immunofluorescence combined with confocal assay demonstrated that while HKCs was stimulated with PMA,the expression of ICAM-1 was up-regulated,which was significantly up-regulated in the group transfected with MMP-9 siRNA(P0.05).Conclusions The inhibitory expression of MMP-9 of HKC may up-regulate the expression of ICAM-1,implying that,besides taking part in the degradation of extracellular matrix,MMP-9 may throw an influence on the proceeding of renal fibrosis by inflammatory pathway.

BACKGROUND: Transformation growth factor β1 (TGF-β1) can promote bone marrow mesenchymal stem cells (BMSCs) migration and proliferation, but the underlying mechanisms remain unclear.OBJECTIVE: To observe the invasiveness of TGF-β1 on BMSCs cultured in vitro, and to investigate regulatory effect on Snail and matrix metalloproteinase 2 (MMP-2) expression.METHODS: Rat BMSCs were isolated and cultured with density gradient centrifugalization and adherence method. The influence of different concentrations of TGF-β1 on the BMSC migration was detected using the modified Transwell chambers. Small interfering RNA for Snail gene was synthesized and transfected into BMSCs by liposomel before TGF-β1 was treated, and the expression of Snail and MMP-2 before and after transfection were measured by western blot assay.RESULTS AND CONCLUSION: The exogenous TGF-β1 can induce a dose-dependent increase in cell migration, which peaked at 2 μg/L. The expression levels of Snail mRNA and MMP-2 mRNA were significantly increased after 2 μg/L TGF-β1 treatment. Snail gene can effectively inhibit the expression of MMP-2 promoted by TGF-β1. Experimental findings indicate that TGF-β1 could increase the MMP-2 expression and then promote the BMSCs migration through the upregulation of the Snail expression.

Objective To explore the culturing strategies,curiculum provision,courses conferring methods,teaching effects as well as the associated managerial evaluations on the basis of Sino-French cooperation on medical education with the hope of summarizing helpful suggestions to Sino-Foreign cooperation on medical education. Methods The achievements of our Sino-French cooperation on medical education were analyzed and compared in the teaching models,culturing strategies along with courses conferring processes among seven-year medical students from both English-teaching and French-teaching classes. Results Our Sino-French cooperation on medical education was featured in its distinct culturing purposes and effective teaching model.Its scientifically formulated culturing strategy found its full expression in French-teaching atmosphere.The Sino-French cooperation on medical education was consistently welcomed and favorably recommended by both faculties and students. Conclusion The Sino-French cooperation on medical education has not only gained precious experience in culturing the cutting-edge medical talents with the international visions but also conduced to fulfill the goal to establish a modernized and internationalized medical school.
