1.Protective effects of artemisinin challenged with CpG DNA in mice

AIM: To investigate the protective effects of artemisinin on mice and its inhibition effects on the release of pro-inflammatory cytokines challenged with CpG DNA. METHODS: A total of 60 mice of Kunming species were randomly divided into six groups. Animals received an intraperitoneal injection of D-galactosamine (D-Gal, 600 mg?kg -1) 1 hour prior to intravenous injection of CpG DNA. CpG DNA group received CpG DNA at 4 mg?kg -1 via caudal vein, artemisinin group were orally administered artemisinin at 200 mg?kg -1, CpG DNA plus artemisinin group first received artemisinin at 50, 100, and 200 mg?kg -1, respectively, then received CpG DNA at 4 mg?kg -1, and the control group received the saline only. The mortality was observed within seven days after injection. RAW 264.7 cell lines were cultivated in vitro and stimulated by CpG DNA to release TNF-? and IL-6, then various concentrations of artemisinin were administrated to observe its dose-dependent inhibitory effect, and artemisinin were also added at different time to observe the time-dependent effect. Contents of cytokines in culture supernatant were detected by ELISA. 5.Preventive and therapeutic effects of Yishen Huanji Decoction on kidney injury in rats induced by simulation of military overtraining

OBJECTIVE: To observe the preventive and therapeutic effects of Yishen Huanji Decoction (YSHJD), a compound traditional Chinese herbal medicine, on military overtraining-induced kidney injury in a rat model. METHODS: Thirty SD rats were randomly divided into normal control group, untreated group and YSHJD-treated group. The military overtraining-induced kidney injury in rats was established by forcing to run on the treadmill for 8 weeks. The rats in YSHJD-treated group were administered with YSHJD at the same time. The 24-hour urines were collected every weekend for detecting the contents of urinary sediment, 24-hour urine total protein, 24-hour urine albumin and activity of N-acetyl-beta-D-glucosaminidase (NAG). The blood and renal tissues were collected after 8-week training, and the levels of serum urea nitrogen (BUN) and creatinine (SCr) were detected. Angiotensin II (Ang II) was detected by radioimmunoassay and activity of Na(+), K(+)-ATPase in kidney was analyzed by chemical colorimetric method. RESULTS: Compared with the normal control group, after 8-week training, the contents of 24-hour urine protein, activities of NAG in urine, and the levels of BUN and SCr in rats in the untreated group and YSHJD-treated group were obviously increased (P<0.05), and these parameters of the YSHJD-treated group were lower than those of the untreated group (P<0.05). The contents of Ang II in blood plasma and kidney of rats in the untreated group and YSHJD-treated group were higher than those of the normal control group (P<0.05), and had no statistical difference between the untreated group and YSHJD-treated group. Compared with the normal control group, the activity of Na(+), K(+)-ATPase in kidney of rats in the untreated group was obviously decreased (P<0.05), and had no statistical difference between the normal control group and YSHJD-treated group. CONCLUSION: YSHJD can protect against military overtraining-induced kidney injury in rats by decreasing the contents of 24-hour urine protein, BUN and SCr, and the activity of NAG, and increasing the activity of Na(+), K(+)-ATPase.

7.Protective effect of Losartan and vitamin E against military overtraining-induced kidney injury in a rats model

Objective To study the preventive and therapeutic effects of losartan and vitamin E on kidney injury induced by overtraining in a rat model. Methods Forty SD male rats were randomly divided into four groups (10 each): control group, model group, losartan group and vitamin E group. Rats in control group were fed with conventional diet. Rats in other three groups were forced to run on the treadmill for 8 weeks, once a day for 20 minutes and 5 days for each week, till exhaustion to induce the kidney injury. Rats in losartan group and vitamin E group were gavaged either with losartan or vitamin E everyday. Urine was collected at the end af each week, urine protein and N-acety-D-glucosaminidase (NAG) were detormined, and the serum urea nitrogen, creatinine (SCr), angiotensin Ⅱ, Na+-K+-ATP-ase activity in kidney were analyzed after 8 weeks' training. Results After 8 weeks' training, the urine protein, NAG in urine, blood urea nitrogen (BUN), SCr were obviously increased in model group, losartan group and vitamin E group as compared with that in control group, while those in model group were higher than that in losartan group and vitamin E group (P0.05). 9.Association between Angle class II division 2 malocclusion and congenital tooth anomalies

Objective: To study the relationship between Angle Class II division 2 malocclusion and congenital anomalies of dentition, Methods: 100 untreated patients (aged 12~42 years old) with Class II division 2 malocclusion were investigated by history,general examination,radiograph and tooth model. The data of congenital tooth anomalies were analysed.Results: Congenital tooth anomaly was found in 50 of the patients. Upper lateral incisor enamel hypoplasia was found in 7 patients, small size of maxillary lateral incisor in 28, canine impaction in 4, missing of lower incisor in 9, transposition in 2 and anterior tooth Bolton ratio discrepancies in 30 of the patients. Conclusion: Angle Class II division 2 malocclusion is closely related to congenital tooth anomalies. The main factors of anterior tooth Bolton discrepancies may be the small size of maxillary lateral incisor and absence of lower incisor.
10.Impact of injury severity on transplantation of embryonic neural stem cells following traumatic brain injury in rats

Objective To explore the influence of injury severity on transplantation of embryonic neural stem cells (NSCs) after traumatic brain injury (TBI).Methods The NSCs were isolated from the hippocampus of fetal rats aged at from 12-14 days.The cells were cultured and proliferated in the serum-free medium and identified in vitro.The animals received transplants in the bilateral hippocampal areas at day 3 following mild or moderate TBI separately.Conventional histology,TUNEL and immunohistology were examined to detect BrdU,NSE,GFAP,GalC,NGF and BDNF at day 14 post-implantation.Results BrdU-labeled positive cells in the bilateral hippocampus in the mild TBI group were more than those in the moderate TBI group at day 14 post-implantation.Significant differentiation of the astrocytes recognized as GFAP positive cells in the bilateral hippocampus was found at day 14 post-implantation.The expression of NGF and BDNF proteins was increased following TBI,the most evident in the mild TBI group.Conclusion The influence of injury severity on transplantation may be associated with the change of the microenvironment after TBI.
