1.A expectation for breakthrough of glaucoma-related genetic study

Glaucoma,especially common primary open-angle glaucoma and primary angle-closure glaucoma,shows high genetic heterogeneity. The causing-disease genes known are difficult to explain some glaucomatous cases,and the study on the susceptibial gene of glaucoma do not achieve new breakthroughs. Combined with the newest progression in genetic study of glaucoma,some views were proposed in this paper in order to better understand the updating study of the pathogenesis mechanism.
2.The function of Hoxa - 10 gene in the reproductive system

Hoxa gene plays an essential role in the generation and development of the human female reproductive system. This gene is expressed in the Mullerian duct and the adult female reproductive system. Expression of Hoxa10 dramatically increased during the midsecretory phase of the menstrual cycle, corresponding to the time of implantation. Female mice lacking Hoxa10 have a uterine factor defect that results in death of the preimplantation embryo and failure of implantation. These results suggest Hoxa10 may have an important function in implantation. Hoxa10 - deficient males exhibit cryptorchidism that lead to male infertility.
3.Expression of HOXA10 gene in human endometrium and its relationship with unexplained infertility

Objective To study the expression of HOXA10 gene in the endometrium of normal fertile women and patients with unexplained infertility during different phases of menstrual cycle. Methods Endometrium samples were obtained by curettage in 52 normal fertile women and 38 patients with unexplained infertility during different phases of menstrual cycle, HOXA10 mRNA expression were detected by in situ hybridization and reverse polymerase chain reaction (RT-PCR). Results (1) HOXA10 mRNA were detected in the glandular and stromal cells of endometrium of fertile women during the menstrual cycle. By in situ hybridization (positive unite, PU), HOXA10 mRNA levels were significantly higher in the mid-secretory phase [glandular cells (G) 5.69?0.57,stromal cells (S) 7.48?0.67] and late-secretory phase(G 5.99?0.40,S 7.98?1.08) than those in late proliferative phase (G 3.35?0.20,S 3.20?0.37) and early secretory phase (G 3.07?0.26,S 3.18?0.27)(P
4.Determination of Menthol and Methyl Salicylate in Shangtongning Adhesive Plaster by Capillary Gas Chromatography

Objective To establish a capillary gas chromatography method for the determination of menthol and methyl salicylate in Shangtongning adhesive plaster. Methods The internal standard substance of n-Pentadecane was employed. The GC system consisted of a DB-WAX capillary column (30 m?0.32 mm, 0.25 ?m), nitrogen as the carrier gas, column temperature at 130 ℃, FID as the detector at 200 ℃, and with a split ratio of 12∶1. Results The contents of menthol and methyl salicylate in 6 lots of samples were determined and the constituents concerned were separated well under the chromatographic condition. The average recovery of menthol and methyl salicylate were 101.52% (RSD=2.66%) and 100.34% (RSD=3.38%), respectively. Conclusion The method was sensitive, accurate, specific and suitable for the quality control of the preparation.

6.Cycloplegic effects of 0 . 5 g/L tropicamide in various low myopia age groups

AlM: To investigated the refraction and cycloplegic effects of 0. 5g/L tropicamide eye drops in various low myopia age groups, and provide clinical guidelines for proper usage of cyclopegic agent in different population.METHODS:Three low myopia age groups (n=45) were selected, non-cycloplegic and cycloplegic auto-refraction using “Shin-Nippon” auto-refractor were performed at various time point for refractive error. The subjective accommodation amplitudes were also collected at each time point.
RESULTS: The accommodation amplitude showed a similar but significant decline between 20~60min after the first cyclopegic drops in all three age groups. However, the decline of accommodation amplitudes were not statistically correlated with the results of “Shin-Nippon”auto-refraction results at different time points.
CONCLUSlON: The refraction time should be arranged according to the cycloplegic effects of tropicamide. Open-field auto-refractor could reduce the usage of cycloglegice drugs in low myopia groups, even for the youngest.
7.Investigation of catheter-related thrombosis around the port in patients with breast cancer after chemotherapy

Objective·To investigate the incidence, risk factors and treatment of the catheter-related thrombosis (CRT) in breast cancer patients after implantation of totally implantable venous access port (TIVAP) in chemotherapy. Methods·A total of 190 cases after implantation of TIVAP were investigated. Color Doppler ultrasound was used to monitor the neck blood vessels to find whether there was CRT before chemotherapy and before taking out the port. The incidence of CRT, occurrence time, risk factors and treatment efficacy were observed. Results·There were 112 (58.9%) cases with CRT and 108 (56.8%) patients with asymptomatic thrombosis, and only 4 cases had symptomatic thrombosis, the incidence of which was 2.1%. Most thrombosis developed on the 21th day after catheterization, and the patients over the age of 60, with clinical stage Ⅲ - Ⅳ and chemotherapy regimens TEC (docetaxel combined pirubicin and cyclophosphamide) were the risk factors for thrombosis. All the patients with asymptomatic thrombosis accepted anticoagulant treatment with low molecular heparin, earthworms enzyme or aspirin, respectively, but there was no significant difference in efficacy in the three groups (P=0.743). Conclusion·Port catheter related symptomatic thrombosis incidence is low but the incidence of symptomatic thrombosis is high in the breast cancer patients after chemotherapy. Age, tumor stage and TEC chemotherapy regimens are the risk factors for catheter-related thrombosis.
8.Effect of doxycycline on matrix metalloproteinase activity and vascular remodeling in balloon-injured rat carotid arteries

AIM: To examine the inhibition of matrix metalloproteinase (MMP) activity by doxycycline (Doxy) and its effect on vascular smooth muscle cells (SMCs) proliferation, neointimal hyperplasia and vascular remodeling. METHODS: The model of rat common carotid artery injury was established by balloon-dilatation. Doxy was administered to the animals of treatment group at dose of 30 mg?kg -1?d -1. The activity of MMPs in the tissue of injured carotid arteries was measured by gelatin zymography. The thickness and area of neointimal, lumen area and the proliferation of SMCs were measured by histological and morphometric analysis. RESULTS: 1. 9.Expression of serum inflammatory factors in patients with glaucoma at different stages and its clinical significance

AIM: To analyze the change of serum inflammatory factors in glaucoma patients at different stages and its clinical significance. ·METHODS:Totally 70 cases of 128 eyes with primary open-angle glaucoma in our hospital from January 2014 to August 2016 were selected. According to the mean defect of visual field, they were divided into light ( observation Group 1 ) , moderate ( observation Group 2 ) and heavy group(observation Group 3). Another 65 cases of 130 eyes with cataract were taken as the control group in our hospital. The observation and expression of serum cytokines in these patients with glaucoma were taken. ·RESULTS:There was no significant difference in serum IL-2 and IFN- γlevels between the two groups (P>0. 05). The sIL-2R and IL-4 levels in the glaucoma group were higher than those in the control group, and the levels of IL-6 and IL-12 were lower than those of the control group (P<0. 05). The IOP and proportion of myopia in the glaucoma group was higher than that in the control group, the difference was not statistically significant ( P<0. 05) . The ratio of moderate myopia to severe myopia in each group was not statistically significant (P>0. 05). To observe the low myopia ratio in Group 3 of patients, it was less than observation Group 1 and observation Group2 ( P > 0. 05 ). There was no statistically significant difference between observation Group 1 and 2 of patients on IL-2, sIL-2R, IL-4, IL-6 and IFN-γlevels (P>0. 05). The level of sIL-2R in the Group 3 was higher than that in the Group 1, and the level of IL-12 was lower than that in the Group 1 and in the Group 2 (P<0. 05). There was no significant difference in IL-2, IL-4, IL-6 and IFN-γlevels between the glaucoma patients (P>0. 05). The IOP level and the proportion of myopia in the Group 3 were higher than those in the Group 1 and the Group 2 were observed, the difference was statistically significant (P<0. 05). The intraocular pressure was higher in Group 2 than in group 1 (P<0. 05). There was no statistically significant difference on proportion of myopia between observation Group 1 and observation Group 2 (P>0. 05). ·CONCLUSION: The levels of serum IL-12, sIL-2R and intraocular pressure in patients with primary open-angle glaucoma fluctuated significantly at different stages of the nerve injury, indicating that the immune response and intraocular pressure were involved in the process of optic nerve damage.
10.Diffusion tensor imaging(DTI) and its application in visual development and amblyopia research

Diffusion tensor imaging(DTI) is an efficient and noninvasive tool to investigate the characteristics and functions of white matter fiber tracts,and it can be used to study the relationship between the structure and function in the nervous system,the plasticity and damage repair of visual development,abnormalities of the white matters that relate to the visual pathways.Recently,the quantification of white matter fiber tracts from the lateral geniculate bodies to the occipital lobe visual cortex using DTI offers morphological information for the function and structure of the white matters during visual development.Some studies attempt to compare the DTI outcomes and tractography between amblyopic children and normal children so as to better display the variations of optic radiation in children with amblyopia,and to evaluate the relationship between structure and function of the white matters.DTI becomes an important supplementary research approach in these fields.This paper reviews the application of DTI in visual development and amblyopic eyes.
