1.The changes of calmodulin and cAMP in rabbit myocardium during ischemia and reperfusion
Chinese Journal of Pathophysiology 1986;0(01):-
This study was performed on rabbits to examine the changes of calmodulin (CAM) and cAMP in the myocardium during ischemia and reperfusion by radioimmunoassay method. The result showed the levels of CaM and cAMP increased at 20 min of ischemia. Although these two parmeters were unchanged at 40 min of ischemia as compared with the changes at 20 min, they were changed during reperfusion at different times of ischemia. At 20 min ischemia following 10 min reperfusion, the content of CaM decreased, the content of cAMP increased. On the basis of 40 min ischemia, reperfusion induced opposite changes: CaM increased, cAMP decreased. The possible mechanism of these changes was discussed.
2.Study on the effect of costunolide on sensitivity of chronic myeloid leukemia K562/ADR cells to doxorubicin via p38-MAPK pathway
Journal of Leukemia & Lymphoma 2021;30(1):23-26
Objective:To explore the effect of costunolide on sensitivity of chronic myeloid leukemia cell line K562/ADR to doxorubicin and its mechanism.Methods:K562/ADR cells in the logarithmic phase were used, and the cells were treated with different concentrations of costunolide, doxorubicin or costunolide combined with doxorubicin for 72 h. The cell proliferation was detected by CCK-8 method, the cell proliferation rate was calculated, and the half inhibitory concentration (IC 50) of the two drugs was obtained. The cells were treated with 10 μmol/L costunolide, 10 μmol/L doxorubicin or costunolide combined with doxorubicin for 48 h, the apoptotic rate was detected by flow cytometry, and the expression level of p38-MAPK pathway related proteins was detected by Western blot. Results:The cell proliferation rate in the costunolide combined with doxorubicin group was lower than that in the corresponding concentration of the two drugs alone groups, and the differences were statistically significant both ( P < 0.05). The IC 50 of doxorubicin for K562/ADR cells was (13.50±0.86) μmol/L, and costunolide was (7.30±0.55) μmol/L ( t = 7.044, P = 0.002). The results of flow cytometry showed that the apoptosis rate of K562/ADR cells in the 10 μmol/L costunolide combined with 10 μmol/L doxorubicin group was higher than that of the blank control group, costunolide alone group and doxorubicin alone group, and the differences were statistically significant [(19.68±3.21)% vs. (2.96±0.87)%, (9.34±2.89)%, (9.18±2.13)%, all P<0.01]. Compared with the 10 μmol/L costunolide alone group and the 10 μmol/L doxorubicin alone group, the expression of the apoptosis inhibitor protein bcl-2 in the two-drug combination group was down-regulated, and the expressions of bad, p-p38, cleaved-caspase-3 and cleaved-PARP proteins were up-regulated. Conclusion:Costunolide can enhance the inhibitory effect of doxorubicin on the proliferation of K562/ADR cells and promote doxorubicin-induced apoptosis, which may reverse the drug resistance of K562/ADR cells by regulating the p38-MAPK pathway.
4.Effects of electroacupuncture on chemotherapy-induced nausea and vomiting and its mechanism.
Chinese Acupuncture & Moxibustion 2014;34(11):1061-1064
OBJECTIVETo observe the effects of electroacupuncture at Neiguan (PC 6) and Jianshi (PC 5) com- bined with granisetron on nausea and vomiting as well as serum levels of 5-hydroxy tryptamine (5-HT) and dopamine (DA) in chemotherapy patients.
METHODSSeventy-two chemotherapy patients who met the inclusive criteria were randomly divided into an observation group (38 cases) and a control group (34 cases). The patients in the observation group were treated with electroacupuncture at Neiguan (PC 6) and Jianshi (PC 5) combined with granisetron injection; the patients in the control group were treated with electroacupuncture at sham-point 1 and sham- point 2, and the rest treatment was identical to the observation group. After 3-day treatment, the clinical indices including times of vomiting and severity of nausea, etc. were observed in the two groups.
RESULTS(1) At the acute stage within 24 h, the times of vomiting in the two groups were not significantly different (P>0.05); at the delay stage, the severity of nausea was significantly higher in the control group (P<0.05). (2) As for the comprehensive efficacy and rate of complete control, there was no obvious difference between the two groups at the acute stage (P>0.05); while in the delay stage, the rate of complete control in the observation group was superior to that in the control group (P<0.01). (3) Compared before and after the treatment, serum levels of 5-HT and DA were evidently reduced in the observation group (both P<0.05), while those in the control group were not significantly changed after treatment (both P>0.05).
CONCLUSIONElectroacupuncture at Neiguan (PC 6) and Jianshi (PC 5) combined with granisetron could obviously relieve the nausea in chemotherapy patients at the delay stage. The effect mechanism is possibly related to reducing the content of 5-HIT nndl nA

5.Induce and differenctiation of marrow mesenchymal stem cell into photoreceptor-like cell in vitro and microenvironment
Chinese Journal of Experimental Ophthalmology 2013;(7):659-663
Background Marrow mesenchymal stem cells (MSCs) has been successful induced to differentiate into corneal cells, retinal ganglion cells (RGCs) and retinal neuron-like cells in recent years, but there are few studies about MSCs induced into photoreceptor cells and their microenvironment. Objective The aim of this study was to explore the induce and differentiation of bone marrow mesenchymal stem cells (BMSCs) into photoreceptor-like cells in vitro and microenvironment. Methods The second generation of human BMSCs strain and RPE cells strain were cultured and passaged, respectively, and the fourth generation of BMSCs and the third generation of RPE cells were used in the experiment.BMSCs were cocultured using the mesenchymal stem cells medium (MSCM) contained 20 μg/L basic fibroblast growth factor (bFGF),20 μg/L epithelial growth factor (EGF)and 20 μg/L brain derived neurotrophic factor (BDNF) with RPE cells to induce the differentiation of BMSCs in the induced group, and BMSCs were cultured in MSCM only in the control group. The morphology of induced and differentiated cells were observed under the inverted microscope.Inmmunocytochemistry was used in induced for 3-,5-,7-day cells to detect the expression rate of rhodopsin protein for the identification of phenotype of the differentiated cells.RT-PCR was used in induced for 5-,7-day cells to detect the expressions of rhodopsin mRNA and recoverin mRNA.Results Cultured BMSCs grew well with the spindle shape, and passaged RPE cells showed the uniform size and polygon shape with the abundant pigment in the cells. Some induced cells appeared to be the neuron-like cells with round shape and long prominence and the secondary reticular branches. The expression rates of rhodopsinin the cells were (5.83±0.29)%,(20.36±0.32)% and (29.80±2.30)% in the third, fifth and seventh day after induce, which were significantly higher than (0.71 ±0.35) %,(2.56±0.24) % and (2.32±0.42) % of control cells (t3 d =41.510,t5d =107.290,t7 d =30.036,P<0.01).The grey scales of rhodopsin mRNA and recoverin mRNA were significantly elevated in the induced and differentiated cells compared with control cells in the fifth and seventh day (rhodopsin mRNA:t5 d =103.506,t7 d =122.584,P<0.01 ; recoverin mRNA:t5 d =106.674,t7 d =189.992,P<0.01).Conclusions BMSCs can be successfully induced to differentiate into photoreceptor cells after cocultured by conditioned medium with RPE cells.

China Journal of Chinese Materia Medica 2014;39(1):40-43
This study was conducted to investigate the physicochemical properties of Polyporus umbellatus sclerotial exudate. Morphological characteristics of the sclerotia and its exudate were observed during different stages of sclerotial formation. The pH of the exudate was detected at different time during cultivation. A phenol-sulfuric acid method was employed to determine the polysaccharide content of P. umbellatus sclerotial exudate during cultivating time. Additionally, the protein content was measured by means of BCA protein assay. Furthermore, CAT content was detected using ultraviolet absorption method. That the protein content of the exudate and CAT specific activity rose gradually during the passage of the cultivating time indicated a high level of oxidative stress during P. umbellatus sclerotial exudate formation. The results showed that the pH of the exudate increased gradually and then dropped down during sclerotial formation. That the pH of the exudate maintained the acidity state during the cultivation indirectly indicated that acidic environment would help sclerotial formation. The exudate produced gradually and was absorbed by the sclerotia itself.

10.Communication between clinicians and laboratorians in semen analysis.
National Journal of Andrology 2015;21(5):387-390
By direct communication between clinicians and laboratorians, obstacles between clinical work and laboratory diagnosis were unequivocally found at an academic conference. Many clinical physicians feel disappointed and frustrated at the laboratory diagnosis by semen analysis and frankly express their expectations, as to recognize the role and significance of laboratory diagnosis, understand the value of routine semen analysis, work out the puzzles in the analysis of sperm morphology, fulfil the requirements for the identification of cells in semen, and obtain responses from and initiate discussions among laboratorians about the relevant questions. Both laboratorians and clinicians are appealing for strengthened management and earlier establishment of national standards and operating specifications for laboratory diagnosis by semen analysis.
