Objective To evaluate the reliability of the white precursor cell (WPC) channel on the Sysmex XN-9000 hematology analyzer (XN-9000) in giving the abnormal white blood cell (WBC) alarm.Methods A total of 520 blood samples with abnormal WBC alarm from the Sysmex XE-2100 (XE-2100) and XN-9000 were performed the manual peripheral blood smear review,and their results were analyzed.Results The specificity of the XN-9000 WPC channel in the blast alarm was significantly higher than that of XE-2100 (x2 =6.98,P＜0.05),while the false positive rate of the XN-9000 (36％) significantly lower than that of XE-2100 (64％,x2 =6.18,P ＜ 0.05).The overall efficiency of the XN-9000 WPC channel in identifying blasts and abnormal lymphocytes (90.4％ and 94.4％) was significantly higher than that of the XE-2100 (72.2％ and 80.1％,x2 =5.6 and 6.33,P ＜ 0.05).Under the automatic reflex analysis mode of WPC channel,the specificity and overall efficiency of the WPC channel in identifying blasts and abnormal lymphoeytes of all the detected samples were significantly higher than those of the WDF channel (x2 =6.15,4.14,6.02 and 4.88,P ＜0.05).The total review rate of the XN-9000 (WDF/WPC) was significantly lower than that of the XE-2100 (x2 =4.33,P ＜ 0.05).Similarly,the review rate of the XN-9000 for the blast/abnormal lymphocyte alarm (84/260,32％) was significantly lower than that of the XE-2100 (107/260,41％) (x2 =4.38,P＜0.05).Conclusion Compared with the XE-2100,the XN-9000 WPC channel can improve the overall efficiency of the blast/abnormal lymphocyte alarm,reduce the false positive rate and the review rate,and then improve the efficiency of clinical laboratories.

Objective To evaluate the performance alarms and parameters generated by hematology analyzer during detection of circulating sezary cells of peripheral blood smears.Methods Blood samples from patients with Sezary syndrome (n=21) were studied with Sysmex XN-9000 analyzer,and compared to patients with benign or tumoral skin lesions (n=48) and patients with chronic lymphoproliferative B-cell diseases (n=55) and normal person (n=45) used as control.Results In present series,the value of structural lymphoid parameters (ly-x and ly-z) and the alarm Blast/Abn Lympho were statistically higher in Sezary cases than in control cases (U=37,40,44,55,38,41,all P＜0.05).Screening Sezary cells critical values of ly-x and ly-z determined by ROC curve were greater than or equal to 86 and was more than or equal to 68.The combination of above three indexes up (i.e.alarm Blast/Abn Lympho＞ 100,ly-x=86 and ly-z＞68) sensitivity and specificity respectively for 100 ％ and 92.1 ％,in order to get the best screening Sezary cells diagnostic efficiency.In addition,the value of ly-x was associated to the count of circulating Sezary cells and value of ly-z to the presence of large Sezary cells,both parameters described as prognostic factors.Conclusion The combination of alarm Blast/Abn Lympho and structural parameters (ly-x/ ly-z/ly-y) may allow to define rule of blood slide review to screen circulating Sezary cells.

Objective To evaluate the performance of the CellaVisionTM DM96 analyzer during detection of intracellular para-sites of peripheral blood smears.Methods This study compared the detection rates of Plasmodium and Babesia species on peripheral blood smears utilizing the CellaVision DM9 6 with the rates for a routine red blood cell morphology scan.A total of 307 slides were analyzed,consisting of 142 slides positive for Plasmodium or Babesia species and 165 negative controls. Slides were blinded,randomized,and analyzed by CellaVision 9 6 and microscopy for red cell morphology scans.The technolo-gists were blinded to prior identification.Results The examination of common technician and blood professional technician, whether using optical microscope or CellaVision DM9 6 could accurately identify all the negative control,its specificity was 100%.The sensitivity of blood professionals using CellaVision DM96 was not statistically significant in the sensitivity of the laboratory and the use of light microscopy for the detection of Plasmodium parasites (χ2=0.24,0.03,0.43,P>0.05).The higher percentage of Plasmodium in blood smear,the higher detection rate of CellaVision DM9 6 .When the blood smear Plas-modium white rate greater than 1%,only 2 contain Plasmodium falciparum samples were negative for the miscarriage of j us-tice.The blood smear percentage was higher than 2% CellaVision of Plasmodium DM96 will all the blood smear 100% cor-rect interpretation.Conclusion This study demonstrates that CellaVision had the potential to detect intracellular parasites in routine screening of blood smears resource for storing images to be used for educational purposes,including training technol-ogists in the detection of intracellular parasites.

OBJECTIVE:To provide information for carrying out pharmaceutical care for future emergency relief work. METHODS: Based on our own experiences,the pharmaceutical care-related work involved in the medical relief in "May?12" Wenchuan earthquake was investigated and analyzed summarily. RESULTS: Scientific emergency plan could create order for the busy relief work,and creative work could make up the deficiency of the plan. It is advisable to attach great importance to the role of the clinical pharmacists in the earthquake relief work. Good command and powerful measures contribute to the smooth proceeding of emergency medical rescue. CONCLUSION: The pharmaceutical care plays an important role in the emergency medical rescue.

Objective To study the seasonal change of antibodies levels in the serum of asthma patients and Der P1 concentration in the dust samples collected from their bedrooms. Methods 36 asthma patients who were allergic to mites were treated as the patients group,21 male and 15 female,aged 43 years of average age. 20 healthy individuals were treated as the control group,11 male and 9 female,with the same age as the patients group. In September and December in 2005,and Match and June in 2006,the dust samples in the bedrooms of patients and the venous blood were collected,the samples of the control were collected only in September. Der P1 concentrations in dust samples and the levels of total IgE,mite specific IgE (s-IgE),s-IgG1,s-IgG2,s-IgG3 and s-IgG4 were detected by ELISA assay. Results In autumn,the levels of total IgE,s-IgE,s-IgG1,s-IgG2,s-IgG4 and the Der P 1 concentrations in patients were significant higher compared with the control group (P

Objective To identify the nanobaeteria in prostate fluid of patients with CPPS.Methods Expressed prostatic secretion(EPS)and urine specimens were collected by Meares-Stamey way from CPPS patients(n=100)and normal controls(n=100).The specimens were cultured and nanobacteria was identified by indirect immunofluoreseenee staining with rnonoelonal antibody.The morphological features were observed by using transmission electron microscopy(TEM). Results The positive rate of nanobaeteria in the EPS cdture of CPPS patients and controls were 43% and 5% respectively,with significant statistical difference(X2=39.58,P＜0.01).By TEM,the sizes of NB ranged from 100 to 500 nm and appeared eoccoid-ccccobacillary shape. Conclusion Nanobaeteria infection may exist in EPS of CPPS patients.

Objective To find out the satisfaction changes among young medical workers and explore the influence of the healthcare reform on such people.Methods A survey was conducted by means of random sampling among 716 medical workers under 45 years old from 6 hospitals,and combined with the survey data in 2009,young medical workers′satisfaction changes and the influencing factors were analyzed.Results Young medical workers′satisfaction is generally low,with declining satisfaction of social respect as well;satisfaction of medical workers in community medical institutions and secondary medical institutions was found declining dramatically.Conclusions Healthcare reform should pay more attention to these young people in terms of their own satisfaction,especially for those working at primary healthcare institutions.

Based on a definition of the concept and its connotation,and summary of the evolution and practice of global budget in China,this paper focused on analyzing the existing problems,namely a defective mechanism in the determination and adj ustment of the total budget,unreasonable settlement, absence of a regulatory mechanism and poor linkage of this system with other payment modes reform.Thus the authors proposed such policy recommendations as designated management of those covered by health insurance,and experiments with the global budget mode for regional groups,scientific estimation and adj ustment of the total budget,reasonable setting of settlement standards and rules,and enhanced supervision and appraisal,as well as various payment system reforms based on global budget.

Objective To probe the gene expression of normal epithelial cell specific-1 (NES1) in normal liver cells and liver cancer cell lines , and investigate the gene methylation status and its impacts on gene expression and cell biology .Methods The expression level of NES1 mRNA was detected in HepG2 and L02 cells by RT-PCR and RT-QPCR, and the level of gene methylation was examined by MSP .We de-tected cell viability by MTT , NES1 mRNA by RT-QPCR and cyclinD1, P21 and P53 level by Western blot after treating cells with 5-Aza-CdR.Results Compared with L02, NES1 mRNA expression in HepG2 was significantly reduced, and the level of NES1 exon 3 CpG island methylation in HepG2 cells was much higher than that in L02 cells.After demethylation , NES1 mRNA expression and protein level of p 21 and p53 in HepG2 cells were up-regulated , while the cell viability and the level of CyclinD 1 were decreased .Conclu-sions In hepatocellular carcinoma , low expression of NES1 mRNA is related to the gene exon 3 CpG island methylation .NES1 exon 3 methylation may be one of the molecular mechanisms for reducing NES 1 mRNA level, and 5-aza-dC could inhibit cell proliferation by inducing cell cycle arrest in HepG 2 cells.
