1.The Prevalence of Type 2 Diabetes Mellitus and its Risk Factors in the Elderly in Gushan Town,Fuzhou City

Objective To know the prevalence of type 2 diabetes and its risk factors in the elderly in Gushan Town, Fuzhou City. Methods From July to November of 2007, 4653 elderly permanent residents from 14 administrative units were selected using random cluster sampling. Results The prevalence of elderly type 2 diabetes was 11.2%. Non-conditional logistic regression analysis showed that age, occupation, monthly income (OR=1.42, 95%CI: 1.13~1.77), family history of diabetes (OR=6.49, 95%CI: 4.62~9.10), concen- tric obesity (OR=1.47, 95%CI: 1.15~1.88) were independent risk factors for diabetes. Conclusion The type 2 diabetes prevalence rate of the elderly was higher, and the health education and interventions should be strengthened to the people who have family history of diabetes and concentric obesity.
2.Effects of epidural and general anesthesia on stress response in gynecological laparoscopic operation

0.05). In the general anesthesia group, glucose level at 10 minutes after pneumoperitoneum was significantly lower than that before anesthesia (P0.05). Conclusion Both epidural and general anesthesia can effectively inhibit the patients' stress response in laparoscopic gynecological operation.

4.MR and DWI Imaging of Central Pontine Myelinolysis after Liver Transplantation

Objective To study the clinical and MRI features of central pontine myelinolysis after liver transplantation.Methods 3 cases of orthotopic liver transplantation patients,displaying unconsciousness,drowsiness,reduced muscle strength of both lower extremities 3 to 21 days after the operation respectively,went through MRI scan in 2 days.Results MRI showed that symmetric long T1 and T2 signals,high-signal-intensity DWI and low-signal-intensity ADC were found in central pontine region.The cerebral cortex was involved in 2 cases,and abscess in basal ganglia appeared in 1 case.MRI examination showed that 2 cases had decreased lesion range,clear boundary and equal-signal-intensity DWI 35 days and 2 months respectively after.Conclusion MRI is valuable for the diagnosis of the patient with central pontine myelinolysis after liver transplantation.
5.Re-infection of hepatitis B virus after liver transplantation

Objective To determine the nucleic acid content of hepatitis B virus with different clinical types of hepatits B before、after liver transplantation patients and explore the approach for detection of hepatitis B.Methods The HBV DNA content in before the liver transplantation, 7 day, 30 day, 90 day after the liver transplantation was detected with fluorescent quantitative PCR and HBV M with ELISA in 275 liver transplantation patients.Results HBsAg、HBcAb was positive of 275 patients;HBV DNA was positive in all HBsAg(+)/HBeAg(+)/HBcAb(+) specimens;79( 73.8 % )out of the 107 HBsAg(+)/HBeAb(+)/HBcAb(+) specimens; 48( 67.6 % )out of the 71 HBsAg(+)/HBcAb(+) specimens in before the liver transplantation.HBV DNA was negative in out of the 275 HBsAg(-) in 7 day after the liver transplantation; 48( 69.6 % ) out of the 69 HBsAg(+) in 30 day after the liver transplantation;104( 75.9 % ) out of the 137 HBsAg(+) in 90 day after the liver transplantation.Conclusions The recurrent hepatitis B were higher in the different clinical types of hepatitis B after liver transplantation,Detection of HBV DNA with fluorescent quantitative PCR can accurately reflect the state of HBV infection and HBV replication and HBV DNA was complementarity with HBV M,it can be used to guide clinical diagnosis with recurrent hepatitis B.

7.Preparation and Preliminary Application of Monoclonal Antibody Against Carcinoembryonic Antigen Glypican-3

Objective To prepare monoclonal antibody specifically against carcinoembryonic antigen glypican-3 (GPC3) and its preliminary application. Methods GPC3 was cloned with PCR to pET16b vector and expressed in E. co-liBL21. Spleen cells were obtained from Balb/c mice embedding immunized with purified antigen intraperitoneally, and fused with Sp2/0 cells. Hybridoma cells were screened by indirect ELISA, and identified by Western blot assay using puri-fied protein after the cell fusion. The indirect immunofluorescence method was used to detect the GPC3 expression in HepG2 cell line. Results The prokaryotic expression vector of GPC3 was successfully constructed, and GPC3 was stably expressed in E. coliBL21. A mouse hybridoma cell line secreting monoclonal antibody to GPC3 was obtained. Western blot analysis showed that monoclonal antibody specifically recognized recombinant protein. Monoclonal antibody could be used to detect GPC3 protein expression in HepG2 cell line by indirect immunofluorescence. Conclusion The monoclonal antibody against GPC3 was successfully obtained.
