1.Observation on the subclasses of IgG and subsets of T cell in 120 patients with inflammatory polyps of large intestine

Objective To study the corelation between patient with inflammatory polyp of large intestine and the changes of immunologic function. Methods Polyps of large intestine were extirpated from 120 patients with fiberoptic colonoscope and the pathological changes were studies. IgG subclass and T cell subsets in the peripheral blood were determined by ELISA and IFA methods. Similar parameters were determined in 30 cases of chronic colitis as control. Results CD8 in patients were lower than that of the control and CD4/CD8 were increased compared with those in control. Conclusion Disorder of humoral and cellular immunity might play an important role on the pathogenesis of polyp of large intestine.
2.Correlation of chronic colitis and fungus allergy in 246 patients

Objective To investigate the relationship between allergic reaction and fungus sensitinogen in patients with chronic colitis. Methods Skin test with 3 groups of fungus allergen (6 types), eosinophil detection in serum and intestinal secretion and serum specific IgE (SIgE) test were tested in 246 patients with chronic colitis. The other 61 patients with chronic diarrhea were served as control. Results Fungus positive was found in 110 patients (43.09%) in dermal fungus allergen test, but only 8.20% in control (P<0.01). In patients with chronic colitis, the positive rates of eosinophil in intestinal secretion and specific IgE induced by fungus allergen were 29.35% and 41.60% respectively. Conclusion It is indicated that some patients with chronic colitis have correlation with allergic reaction of digestive system induced by food with fungus allergen.

4.Therapeutic Observation of JIN's Three-needle Therapy in Improving Intelligence in Mental Retardation as An Assistant Treatment

Objective To explore the effect of Jin's three-needle therapy, as an assistant treatment, in improving the intelligence of children with mental retardation (MR).Method Seventy-six MR patients were divided into a control group and an observationgroup, 38 cases in each group.The control group was treated with rehabilitation, while the observation group with JIN's three needle therapy plus the rehabilitation training.Prof. JIN Rui's 10-item observation index was adopted to evaluate the memory ofthe patients before and after the treatment; Gesell Developmental Schedule (GDS) was used to measure the intelligence before and after the treatment. The clinical efficacies in the two groups were statistically analyzed.Result After the treatment, Prof.JIN Rui's 10-item index scores dropped obviously and the GDS scores increased significantly in both groups, while the decrease and increase of scores in the observation group were more significant than those in the control group (P<0.05). The total effective rate was 94.7% in the observation group, significantly higher than 71.1% in the control group (P<0.05).Conclusion As an assistant treatment, JIN's three-needle therapy can help rehabilitation training to improve the intelligence of MR patients.

7.Clinical Efficacy of Loratadine Combined with Pidotimod in the Treatment of Children with Recurrent Re-spiratory Tract Infection and Effect on Immune Function

Objective:To study the clinical efficacy of loratadine combined with pidotimod in the treatment of children with repeat-ed respiratory tract infection and the effect of the combination on immune function. Methods:Totally 78 cases of patients with repeated respiratory tract infection were randomly divided into the observation group ( n=39 ) and the control group ( n =39 ) . Both of the groups were treated with the routine correct treatment, and the control group was treated with pidotimod and the observation group was treated with loratadine combined with pidotimod with the treatment course of 8 weeks. The clinical efficacy, clinical symptoms, signs improvements, changes of immune function markers level and incidence of adverse drug reactions between the two groups were observed and compared. Results:The total effective rate of the observation group was significantly higher than that of the control group ( P<0. 05);the disappearance time of pulmonary rales and cough, fading time of tonsil swelling and fever clearing time of the observation group were significantly shorter than those of the control group(P<0. 05). After the treatment, the levels of IgG, IgM and IgA in both groups were significantly lower than those before the treatment (P <0.05); the levels of CD3 +, CD4 + and CD4 + /CD8 + in both groups were significantly higher than those before the treatment (P<0. 05);the levels of immune indices in the observation group were significantly better than those in the control group(P<0. 05). Comparing with the incidence of adverse drug reactions in the control group, no significant difference was shown in the observation group (P>0. 05). Conclusion:The clinical efficacy of loratadine com-bined with pidotimod in the treatment of children with repeated respiratory tract infection is effective, which can improve the clinical symptom and signs and enhance the immune function of children significantly.
8.The correlation study between the manifestation of MRI and pathology of early cerebral infarction after treatment with AQP4 mRNA silence

Objective This study investigated magnetic resonance imaging (MRI)manifestations and pathological changes after aquaporin-4 (AQP4)gene silencing in treatment of early cerebral infarction.Methods Ninety adult Wistar rats were randomly divided into con-trol group,infarction group and treatment group.All the above groups were assigned to five sub-groups according to the time point:0.5,1,2,4 and 6 hours,respectively (n= 6 for each group).For the right middle cerebral artery occlusion (MCAO-infarction group),the rats were separately injected with short hairpin RNA-AQP4 liposome (shRNA-AQP4+MCAO-control group)and in-terference agents (siRNA-AQP4 + MCAO-interference group).The rats in all groups were examined with T2 weighted image (T2 WI)and diffusion-weighted imaging (DWI).The rs-T2 WI,rs-DWI and relative apparent diffusion coefficient (ADC)of the big-gest high-signal-intensity layer on T2 WI and DWI were measured.Corresponding brain tissue was collected for pathological observa-tion.The data were analyzed using SPSS 13.0 software.Results Identical changes were detected between control and infarction groups.The high-signal intensity was found on DWI at 0.5 hour and on T2 WI at 2 hours after MCAO.rs-DWI and rs-T2 WI in-creased with time.The value of relative ADC decreased quickly within 2 hours after MCAO,and cytotoxic edema was significantly aggravated.The value of relative ADC increased slowly at 4 and 6 hours,but angioedema was detected during this period.There was significant change in relative ADC between the treatment and control groups at 0.5 to 4 hours (P < 0.01)and cytotoxic edema was significantly lightened.But there was no significant difference in relative ADC between treatment and control groups at 6 hours (P >0.05).However,angioedema was visible in this group at 6 hours.Above results confirmed that during early cerebral in-farction,the differential regions of DWI and T2 WI high-signal area were considered as cytotoxic edema,which was the pathologic basis of the decreased ADC values.AQP4 gene silencing could effectively reduce cytotoxic edema.Conclusion DWI and ADC value could reveal timely and exactly therapeutic outcomes of cytotoxic edema.The high-signal intensity on T2 WI represents angioedema, and AQP4 gene silencing has no obvious change.
9.Determining the ideal decellularization protocol applied for preparing for liver biological scaffold

Objective To develop an appropriate and simple decellularization protocol for preparing decellularized liver bio-logical scaffold(DLBS) .Methods Through immune-ofluorescence analysis ,scanning electron microscope ,quantification of Glyco-samin-oglycans(GAGs) and DNA ,we evaluated the effectiveness of low concentrations of SDS(0 .25% ,0 .50% ) and 1% Triton X-100 in the preservation of extracellular matrix of rat livers .Then ,we assessed Cytotoxicity of DLBS treated with three methods by MTT assay .Results Residual DNA after SDS(0 .25% ,0 .50% ) treatments were below 50 ng/mg dry weight ,which were less than Triton X-100 treatments .The content of GAGs in the 0 .25% SDS and 1% Triton X-100-treated scaffolds were higher than other detergent-treated scaffolds .The result of MTT assay showed that the liver scaffolds treated with three methods were not cytotoxic to proliferation of C3A cells .Conclusion The protocol containing 0 .25% SDS and a perfusion rate of 5 mL/min could be a rapid and effective decellularization protocol .DLBS constructed using this protocol could be an ideal material for preparing a transplant-able organ .
10.Correlation research of diabetes distress, self-perceived burden and social support in patients with middle-and-high risk of diabetic foot

Objective To investigate the status of diabetes distress in patients with middle-andhigh risk of diabetic foot and explore the impact of self-perceived burden and social support on diabetes distress.Methods Totally 205 patients with middle-and-high risk of diabetic foot were investigated with Diabetes Distress Scale,Self-perceived Burden Scale and Social Support Scale.Results The total average score of diabetes distress was (2.59±0.57),the diabetes distress level was correlated with self-perceived burden,dimensions of social support (r=0.481~0.653) and were influenced by gender,vocation,objective support,use of support.Conclusions Diabetes distress is widespread in patients with middleand-high risk of diabetic foot and was influenced by many factors,these provide the basis for taking effective interventions to reduce diabetes distress.
