1.The effect of cyclindependent kinase 2 On herpes simplex virus replication

Objective To explore the effect of cyclindependent kinase(CDK)2 on the replication of herpes simplex virus(HSV).Methods Dominant-negative CDK2 expressing cell line HT29Tet-On dn-CDl(2 was used as host cells to determine the role of CDK2 in HSV rephcation.HT29Tet-0n dn-CDK2 cells were infected with HSV.The activity of CDK2 Was determined by the immunoprecipitate and autoradiograph before and after HSV infection.The growth curve of HSV Was conducted.Results The CDK2 activity was induced after infection for 6 h and reached the hishest level at 9 h.HSV replicated rapidly and productively after the CDK2 was induced.Coatiusion CDK2 is required for the replication of HSV,and HSV replicates in host cells by inducing the CDK2 activity.

4.Assessment of the difference of myocardium strain between myocadial infarction and ventricular aneurysm by two-dimensional speckle-tracking imaging in rabbits

Objective To evaluate the difference of myocardium strain between myocadial infarction and ventricular aneurysm (VA) by two-dimensional speckle-tracking imaging in rabbits.Methods 26 healthy New Zealand white rabbits were enrolled in this study,6 were put into the control group and 20 in the experimental group.models of myocardial infarction concomitant with VA were prepared by the ligation of the middle segment of left anterior descending artery and left circumflex artery.At 4 weeks after the procedure,according to the pathological inspect and echocardiography for whether VA formed,the experimental animals were divided into the VA group and the myocadial infarction group.Echocardiography was performed to measure the LV cavity size and cardiac function.Radial strain rate (SrR) and circumferential strain rate(SrC) indexes were measured by 2D-STI software for each animal at the level of apex in left ventricular short-axis view,and then calculated the strain rate decrease percentage in each segment.Results 1)Compared with the control group,SrR-S,SrR-E,SrR A,SrC-S,SrC-E,SrC-A in all segments were reduced significantly in the VA group and the myocadial infarction group (P < 0.05),the VA group was the most significant.2)Compared with the myocadial infarction group,the strain rate decrease percentage of SrR-S,SrR-E,SrR-A,SrC-S,SrC-E,SrC-A in all segment had significant in the VA group.Comparison between the each section,the strain rate decrease percentage of SrR-Santerior wall,SrR-Sateral wall,SrC-Santerior wall,SrC-Slateral wall were the most significant.3)There were the tighter relationship between the strain rate decrease percentage of SrR-Santerior wall,SrC-Santerior wall,SrR-Slateral wall,SrC-Slateral wall and left ventricular ejection fraction (r =0.82,0.72,0.75,0.71,repectively,all P < 0.05).4) The ROC curve analysis showed that when the strain rate decrease percentage was 60% as the critical point,the strain rate decrease percentage of SrC-Santerior wall,SrC-Slateral wall,SrR-Santerior wall,SrR-Slateral wall had a sensitivity of 83.30 %,84.40%,83.30%,67.70%% and a specificity was 73.60%,78.20%%,69.70%,83.30% for prognosis ventricular aneurysm,repectively.Conclusions After ventricular aneurysm formation,anterior and lateral wall myocardial strain were give priority to the myocardial mechanics reconstruction,thus systolic myocardial strain rate decrease percentage can be important indicator of infarction myocardial developing into ventricular aneurysm.
5.Effect of glucagon-like peptide-1 receptor agonists on growth and energy metabolism in human medullary thyroid cancer cells

Background and purpose:This study aimed to investigate the effect of glucagon-like peptide-1 receptor agonists on proliferation, secretion of calcitonin and energy metabolism of medullary thyroid cancer (MTC) cell.Methods:The MTC cell line (TT) was culturedin vitro. After treatment with exenatide and liraglutide (0, 1, 10 and 100 nmol/L) for 24, 48 and 72 h, the proliferation of TT was analyzed by CCK-8 kit, the calcitonin was measured by calcitonin assay kits, and the energy metabolism of TT was measured by Seahorse XF instrument.Results:When compared with control group, neither exenatide nor liraglutide had effects on proliferation of TT (P>0.05); the calcitonin levels did not change signiifcantly after treatment with GLP-1 receptor agonists (P>0.05). Exenatide and liraglutide did not alter glycolysis and mitochondrial respiration in TT cells in a dose- and time-dependent manner.Conclusion:GLP-1 receptor agonists have no effect on the development of TT. Further collection of the safety data of exenatide and liraglutide on thyroid is still needed.
6.The application of TIPSS in portal vein cancerous thrombosis complicated with portal hypertension

Objective To discuss the technical skills and the contraindication of transjugular intrahepatic portosystemic shunt stent (TIPSS) in portal vein cancercous thrombosis (PVCT) complicated with portal hypertension. Methods There were 16 cases of PVCT with portal hypertension, and average age of 53.6 yr. There were 9 cases with complete occlusion of portal vein trunk and 7 cases with incomplete thrombosis. There were 5 cases with cavernous transformation of the portal vein(CTPV). 1 case of simple upper gastro intestinal tract (GIT) massive bleeding, 4 with refractory ascites and 11 with upper GIT massive bleeding and refractory ascites. Results The procedure of TIPS was successful in 11 cases, the successful rate reached about 68.8%. The mean portal vein pressure was reduced from 4.9kPa to 2.4kPa with average 2.5kPa reduction. Ascites decreased, bleeding stopped and the clinical symptoms disappeared. The average survival period was 136 days. The procedure failed in 5 cases. Conclusions TIPSS is an effective method to control the bleeding and ascites caused by PVCT. The PV cavernous transformation was the contraindication of TIPSS.

Myocardial hypertrophy is the complication of many cardiovascular diseases that induce cardiac remodeling.The molecular mechanism of cardiac remodeling involves abnormal changes in various transmembrane ionic currents in the heart.Recent studies suggest the potential involvement of volume-regulated Cl-current(ICl.Vol)in cardiac hypertrophy.Although the molecular basis of ICl.Vol remains to be elucidated,recent progress is reviewed in the potential role of ICl.Vol in cardiac remodeling.

Objective To evaluate the rapid diagnosis of bacterial and (or) fungal endophthalmitis by multiplex polymerase chain reaction (MPCR). Methods MPCR was performed to detect the DNA segment of bacteria and (or) fungi from standard strains and 41 samples of intraocular fluid or vitreous from 38 patients (3 with double eyes and 35 with single), and the results were compared with the cultured bacteria and fungi. 9.Inhibitory effect of polyphyllin D on the proliferation of human chronic myelogenous leukemia cell line K562 and its mechanism

Objective To explore the inhibitory effect of polyphyllin D on the proliferation of human chronic myelogenous leukemia (CML) cell line K562 and its mechanism. Methods K562 cells were treated with various concentrations of polyphyllin D (0, 0.1, 0.2, 0.4, 0.8, 1.2, 2.4 μmol/L) at 24 h, and cell viability was assessed by CCK-8 assay. Flow cytometry was used to detect the effect of polyphyllin D on the apoptosis, and the cell cycle arrest of K562 cells. The relative proteins were analyzed by using Western blot. Results The polyphyllin D could significantly inhibit the proliferation of K562 cells, and the effective inhibitory concentration (IC50) was (0.9 ± 0.1) μmol/L at 24 h. The results of flow cytometry showed that after treatment with 0.9 μmol/L polyphyllin D at 12 h and 24 h, the apoptotic rate of the cells [(11.46 ±1.51) %, (28.87 ± 2.35) %] were significantly higher than that of the control group [(2.05±0.45) %], and the difference was statistically significant (F= 38.637, P< 0.05). The expressions of bcl-2, CDK1, CyclinB1 and bcr-abl fusion protein were down-regulated by polyphyllin D, and the expressions of Bax, cytochrome C, activated caspase-3 and p21 were up-regulated (all P<0.05). In addition, polyphyllin D could arrest cell-cycle at G2/M phase (F=42.355, P<0.05). Conclusion Polyphyllin D can significantly inhibit the proliferation of human CML cell line K562, and its mechanism could play a role by inducing apoptosis and promoting cell cycle arrest.
10.Influence of clinic design of the prostheses with ERA attachment for distal extension defect: A three-dimensional finite dement analysis

Objective:To evaluate the stress distribution and displacement pattern in the restoration of Stern-ERA attachment mandible removable partial denture (RPD) with different designs through three-dimensional (3D) finite element (FE) analysis.Methods:3D-FE models were established based on human CT data and manufacturer's catalog.Vertical and 45° lingual oblique forces of 100 N were applied on the artificial teeth of the RPD.Three ERA attachment partial dentures with different bracing arms were designed,namely the basic design,rest bracing arm design and cross-arch bracing arm design.The von Mises stresses and displacements of abutment teeth and mucosa under denture base were analysed.The displacement of denture bases was examined.Results:Under the vertical and oblique loading,the maximum yon Mises stress was found at the distal shoulder on the distal abutment.The maximun displacement was observed at the buccal and distal occlusal surfaces of the distal abutment.The maximum displacement on the abutment was found to be the highest in rest bracing arm design group (14.91 μm and 63.09 μm),followed by the basic design group (9.38 μm and 50.56 μm) and the cross-arch bracing arm design group exhibited the smallest displacement (8.85 μm and 47.31 μm).Conclusion:The basic design Stern_ERA attachment for unilateral mandible distal extension defect appears to be more reasonable compared with rest arm and crossing-arch arm design from biomechanical point of view.
