Background: \r\n', u'Japanese encephalitis virus (JE) is the most prevalent in Asia, Pacific and in mainland northern Australia; and considered to be the leading cause to the acute encephalitis. The case- fatality and sequela rates in children stay rather high. There were some medical technologies for the JE diagnosis, of which is the application of the expensive MAC- ELISA and PEN TAX kits.\r\n', u'Objectives: \r\n', u'To evaluate and compare the light sensitivity of MAC- ELISA and PENTAX Kits in JE diagnosis.\r\n', u'Subjects and method: \r\n', u'48 pairs of sera samples obtained from the patients with clinically manifested Japanese encephalitis (JE) in 2003- 2005, in some northern provinces, they were applied and tested into MAC- ELISA Kits.\r\n', u'Results:\r\n', u'The sensitivity of MAC- ELISA and PENTAX kits detecting IgM against JE virus were 95.71%, and 98.57%, respectively. 2.Detection of IgM anti Japanese Encephalitis (JE) virus by antigen genotype 1 & genotype 3

Background: Recently Japanese Encephalitis (JE) virus type 1 has surfaced and is co-circulated with JE virus type 3 in the northern areas of Viet Nam, so a sensitivity of JE viral antigen genotype 3 to detect IgM is required. Objectives: To compare the sensitivity of JE viral antigen genotype 1 and 3 to detect IgM against the JE virus. Materials and method: 783 cerebrospinal fluid and serum samples from viral encephalitis cases from 1999-2005 were collected and examined by MAC-ELISA for JE viral antigen genotype 1 and 3. Results: The agreement on the diagnosis of these kinds of antigen was 99.7% and the sensitivity of JE viral antigen genotype 3 was higher than that of genotype 1. Thus, JE viral antigen genotype 3 could be considered as the selected antigen for JE diagnosis in Viet Nam. IgM titer determined by JE viral antigen genotype 1 was higher than that of genotype 3 in 2003 and 2005 and lower in 1999, 2000, 2001, 2002 and 2004. Conclusion: The dominant phenomenon of JE viral genotypes differing over the years might be due to the interaction of the virus and its vectors. Further study is required to clarify this observation.

3.Detection of Japanese encephalitis genotype 1 in central, southern and highland of Viet Nam

Background: In recent year, Japanese Encephalitis Virus (JEV) genotype 1 has been detected among isolates from mosquitoes and pig\u2019s blood samples in northern Viet Nam, but there has been no information on the presence of this genotype in the Central, Southern and Highland regions. Objectives: This study aims to detect the Japanese encephalitis genotype 1 in various different geographic regions of Viet Nam. Material and method: Sequence analysis\u2019s of whole E gene of 18 strains isolated from human, mosquitoes and pig\u2019s blood during 2001-2007. Results: 7 strains isolated from pig\u2019s blood and mosquito samples in the Northern, Central, Southern and Highland fell into genotype 1, but 11 others isolated from humans in the Northern and Central regions belonged to genotype 3. Conclusion: This is the first time that JEV genotype 1 was detected in the central, northern, highland Viet Nam and further studies on genotype 1 causing human diseases needs to be carried out.\r\n', u'\r\n', u'

Background: Mosquitoes and pigs play important roles in maintaining and increasing the Japanese Encephalitis (JE) virus in nature and which is then transmitted to humans. Thus, surveillance of the JE infection frequency in the pig population may predict the human JE cases. \r\n', u'Objectives: The study aimed to determine IgG antibody against the JE virus in the pig population in Hanam province \r\n', u'Subjects and methods: The study included 1791 pig serum samples collected from 3 districts of Hanam province from Apr 2006 to Mar 2007. GAC-ELISA technique was used to determine the JE virus infection in the swine population.\r\n', u'Results: The average positive rate in pig population was 34.9 % (626/1791); with the highest frequency occurring in the summer (37.7%- 84.0 %), co-incident with the JE season in Northern Vietnam. Background: IgM antibody capture ELISA (MAC-ELISA) technique has been widely applied for Japanese Encephalitis Virus (JEV) diagnosis. So far rare internationally commercial kits are available. Thus, the international evaluation of the kit is required as per the recommendation of the WHO. Objectives: To evaluate the quality of the IgM antibody capture ELISA diagnostic kit for JEV produced by the Vietnam National Institute of Hygiene and Epidemiology (NIHE). Subjects and method: In this study, NIID kit was used as control to check the kit from NIHE. Both NIHE and NIID kits were used to detect JEV IgM among 38 serum and 6 CFS samples, which belongs to 5 sample groups (JE patients group, dengue patients group, other viral encephalitis patients group, Tick Born Encephalitis (TBE) patient group and healthy JE vaccinated donors group). Results: The detection of JEV IgM by NIHE kit was concurrent with the NIID kit. There is no positive with the JE in the groups of Dengue patients, TBE, other virus encephalitis patients and JE vaccinated donors. Conclusion: MAC-ELISA kit of NIHE can be used for different diagnosis of JEV and Dengue virus (both viruses are in Flavivirus genus), as well as other viruses caused by encephalitis.

7.Results of medical arthropod surveys in the natural conserve zone- Can Gio mangrove forests, Ho Chi Minh city

Background: Can Gio mangrove forest is bio-diversified area. There is few studies on medical arthropod in Can Gio mangrove forest until now. Objective: To study bio-diversified feature and identify species with disease transmitting role. Subject and Method: In 2007, medical arthropod surveys were conducted at 3 sites located in the natural conserve zone- Can Gio mangrove forest. A cross-sectional study was used. Results and Conclusions: A total of samples of 6178 individuals of medical arthropod belonging to 65 species have been collected, including 3 species of flea (Siphonaptera), 1 species of ticks (Ixodidae), 6 species of chiggers (Trombiculidae), 6 species of mites (Gamasoidea), 25 species of flies (Muscoidea) and 24 species of mosquitoes (Culicidae). They belong to 37 genus, 17 families, 3 orders (Siphonaptera, Acarina and Diptera), and 2 classes (Insecta and Arachnida). 20 species having epidemiological role were found in Can Gio mangrove forest. The rate of Anopheles epiroticus biting human at night indoor, outdoor and daytime indoor is rather high, especially at the end of dry season at centre of the forest.

8.Effects of Compounds from Physalis angulata on Fatty Acid Synthesis and Glucose Metabolism in HepG2 Cells via the AMP-activated Protein Kinase Pathway

The ability of the total extract from Physalis angulata; three fractions after partitioning with n-hexane, ethyl acetate (TBE), and water; and four withanolides (compounds 1 – 4) to phosphorylate 5'-adenosine monophosphate-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in HepG2 cells was evaluated. The TBE fraction (50 μg/mL) activated p-ACC and p-AMPK expression most strongly. Compounds 1 – 4 (10 μM) upregulated p-ACC expression at different levels. Compound 4 induced the most significant changes in p-AMPK expression, followed by 1 and 2. Sterol regulatory element-binding proteins (SREBPs) play a functional role in the transcriptional regulation of the lipogenic pathway, including fatty acid synthase (FAS) and ACC. The effects of compounds 2 and 4 (10 μM) on FAS and SREBP-1c expression under high glucose conditions (30 mM) in HepG2 cells were evaluated further. Both dose-dependently inhibited FAS and SREBP-1c expression as well as lipid accumulation (1 – 10 μM) were compared to high-concentration glucose control, which upregulated FAS and SREBP-1c. These results suggest that compounds 2 and 4 upregulate AMPK, suppress FAS and SREBP-1c, and have potential effects on glucose and lipid metabolism.

Background: Human albumin was produced and used in many countries. Cohn's technique had been used to precipitate albumin from human plasma. This technique was easy and cheap and the quality of the product was good. In Vietnam, human albumin had to import, but the prices was very expensive. Vietnam was having good plasma in large quantity and high quality. That\u2019s why research on production plasma albumin was essential.\r\n", u"Objectives: This study aimed at using Cohn's technique improved by Drohan and Van - Aken to produce standard albumin from human plasma. Subjects and method: Human plasma detected VIII-factor was used for present study. Plasma \ufffd?albumin was precipitated by ethanol at low temperature and pH. The collected albumins have been liophilizated and storage at 40C. The quality and quantity of Albumin was evaluated by quantitative analysis and protein \ufffd?electrophoresis. Results: The 418g of albumin powder was produced from 16 liters of plasma detected F \ufffd?VIII. The quality of this albumin come up to standard (>95%) and quantity of albumin collected from one liter of this plasma was 26g. Introduction: The need for gama \u2013 globulin, especially gama - globulin - anti \u2013 HBs, is huge in Vietnam. A number of patients cannot to afford use them due to the high price as they are imported. Meanwhile, Vietnam has high quality input sources for producing gama \u2013 globulin. \r\n', u'Objectives: To study the production process of gama - Globulin from Human Plasma and Standard gama - Globulin with Rich of Anti \u2013 HBs. \r\n', u'Subjects and method: 168 samples of human plasma from voluntary blood donors, which had been screened with for transfusion transmittable infections (TTLs), were chosen as plasma with rich of anti - HBs. The plasma with anti - HBsAg was precipitated with ethanol 25%, pH 6,9 to gain gama - globulin with rich anti - HBs, which was dried by Dutch Ly - 3 - TTE machine. Activation of anti - HBs gama - globulin was identified by a standard method of the degree of antibody specific for anti - HBsAg kit of BIORAD, \r\n', u'Results: The purity of the gama - globulin achieved was 93%, which was almost equal with the results of some foreign researchers (7.8), the activation of anti - HBs was 1:128 dilution degree. The productivity of gama - globulin gaining from 1 litter plasma was 6.0 gram.
