1.Effects of turmeric volatile oil on the morphology of human lung adenocarcinoma A549 cells

Objective To investigate the effects of turmeric volatile oil extracted from curcuma on human lung adenocarcinoma A549 cell line. Methods The morphological changes of human lung adenocarcinoma A549 cells treated by different concentrations of turmeric volatile oil were observed by inverted microscopy, Giemsa staining, and Acridine orange staining. Results ① The inverted microscopy revealed that after treatment of A549 cells with different concentrations of turmeric volatile oil for 24-72 h or specified concentration of turmeric volatile oil for 1-7 d, the lung carcinoma cells were distributed sporadically, cells were scarce, and the majority of cells were in round or oval shapes, indicating that the cell proliferation was inhibited. These changes were associated with the concentration of turmeric volatile oil. ② Giemsa staining indicated that the apoptotic rates of A549 cells were 34% and 42% respectively after turmeric volatile oil treatment at the concentrations of 0.8 ?l/ml and 1.0 ?l/ml. AIM: To explore the protective effects and mechanism of astragalus injection on asthmatic rats. METHODS: OVA was injected intraperitoneally and inhaled to produce the asthmatic rat models. Forty rats were randomly divided into five groups: control group, asthma group and astragalus groups of high, medium and low dose. The concentration of IL-5 in BALF and the expression of IL-5 mRNA, phospho-p38 MAPK in lung tissue were respectively measured by ELISA, in situ hybridization staining and Western blot. Objective To construct an eukaryotic expression vector encoding an shRNA targeting CA916798. Methods According to the CA916798 cDNA sequence in GenBank, 2 pairs of oligo nucleotides were designed and synthesized. After primer annealing, they were inserted into plasmid pGenSil-l to construct the shRNA eukaryotic expression vector. The recombinant plasmid were transformed into DH5?, and the positive strain were identified by enzyme digestion and sequence analysis. The recombinant vector were transfected into A549/CDDP cells with Lipofectamine 2000. Drug sensitivity and proliferation of the transfected cells were measured by MTT test. Results The pRNAi-CA916798 shRNA of recombinant plasmid was constructed successfully. 7.The loss of heterozygosity on chromosome 3 of plasma free DNA of small cell lung cancer

Objective To study the diagnostic and monitoring values of the loss of heteroxygosity(LOH) of 3p loci in free plasma DNA of small cell lung cancer. Methods PCR silver staining was used for the detection of LOH of three microsatellite sites in the 3p loci of plasma free DNA and tissue of SCLC. Results In 33 cases of tissue, the incidence rate of LOH was 54.5%(18/33), but the positive rate of LOH in plasma free DNA was 42.4%(14/33). The correlation between them was 78.8%(14/18). Conclusion The plasma free DNA in patients with lung cancer is primarily originated from the tumor tissue. LOH of plasma free DNA may be valuable molecular markers in the diagnosis of SCLC.
8.The significance of microsatellite analysis of plasma free DNA in diagnosis of lung cancer

Objective To study the diagnostic value of LOH and MSI of free DNA 3p microsatellite point in plasma of lung cancer patients. Methods Using PCR and sliver dye techniques, LOH and MSI of three microsatellite sites of plasma DNA on chromosome 3p in resected fresh tissue from 37 patients and whole blood sample from 94 patients were detected. Results The plasma free DNA concentration of most lung cancer patients was above microgramme level. Above 70% of tissues and plasma were positive. No significant difference was found at different stages and in different pathologic types. The result of patients with lymph node metastasis was significantly different from that of the patients without lymph node metastasis. Conclusion Plasma DNA of lung cancer patients is a good medium for gene diagnosis and may be used widely in the future.
9.GP combination therapy for treatment of 50 patients with advanced non-small cell lung cancer

The major non hematologic toxicity was nausea or vomiting. Conclusion The GP combination chemotherapy is an active regimen for the advanced NSCLC with tolerable toxicity.

The proteins and antigens of nine strains of Toxoplasma gondii were analyzed by SDS-PAGE and immunoblotting. These strains including RH strain, Zs-2 strain, CN strain and SHI, SH2, SH3, SH4, SH5, SH8 strains, were obtained from other laboratories or isolated from deformity fetus in our laboratory. SDS-PAGE analysis revealed that these strains were very close in the major bands of proteins and differences were observed only in part. There were common components in antigens of all strains using IgG antibody prepared from high-titre rabbit antisera raised against RH strain of Toxoplasma.
