1.Effects of turmeric volatile oil on the morphology of human lung adenocarcinoma A549 cells

Objective To investigate the effects of turmeric volatile oil extracted from curcuma on human lung adenocarcinoma A549 cell line. Methods The morphological changes of human lung adenocarcinoma A549 cells treated by different concentrations of turmeric volatile oil were observed by inverted microscopy, Giemsa staining, and Acridine orange staining. Results ① The inverted microscopy revealed that after treatment of A549 cells with different concentrations of turmeric volatile oil for 24-72 h or specified concentration of turmeric volatile oil for 1-7 d, the lung carcinoma cells were distributed sporadically, cells were scarce, and the majority of cells were in round or oval shapes, indicating that the cell proliferation was inhibited. These changes were associated with the concentration of turmeric volatile oil. ② Giemsa staining indicated that the apoptotic rates of A549 cells were 34% and 42% respectively after turmeric volatile oil treatment at the concentrations of 0.8 ?l/ml and 1.0 ?l/ml. Objective To introduce a new method to open bite.Methods Compound resin materials were used to make a bonding type deep-bite opening device on the lingual side of anterior teeth in 21 patients.Results 21 patients with deep overbite treatment by this method achieved satisfactory results.Average time in Ⅱ°deep bite pa tients was(3.5 ± 1.4)months,and(5.4 ± 1.2)months in Ⅲ°deep bite patients.Conclusion This device is favorable for rapidly opening deep-bite and is simple and effective.
3.Determination of Content of Astragaloside in Bushenbanlong Tablet by HPLC-ELSD

Objective To establish the quality control method for Bushenbanlong Tablet. Methods The content of astragaloside was determined by HPLC-ELSD. Results The linearity range was 1.44~4.32 ?g with a correlation coefficient r=0.999 9, and the average recovery was 99.69% with RSD=1.16%. Conclusion The method was simple, convient, quick and sensitive. It provide an effective method for quality control of Bushenbanlong Tablet.
4.The effects of scopolamine on Na~+-K~+-ATPase activity during cerebral postischemic reperfusion in rabbits

0 05).CONCLUSION:Na +-K +-ATPase is not sensitive to ischemic damage,but sensitive to reperfusion damage.Like hypothermia,scopolamine can protect Na +-K +-ATPase activity in the cerebral cortex during postischemic reperfusion,but hypothermia has no cooperating effect on scopolamine.
5.Construction and identification of CA916798 eukaryotic expression vector for RNA interference

Objective To construct an eukaryotic expression vector encoding an shRNA targeting CA916798. Methods According to the CA916798 cDNA sequence in GenBank, 2 pairs of oligo nucleotides were designed and synthesized. After primer annealing, they were inserted into plasmid pGenSil-l to construct the shRNA eukaryotic expression vector. The recombinant plasmid were transformed into DH5?, and the positive strain were identified by enzyme digestion and sequence analysis. The recombinant vector were transfected into A549/CDDP cells with Lipofectamine 2000. Drug sensitivity and proliferation of the transfected cells were measured by MTT test. Results The pRNAi-CA916798 shRNA of recombinant plasmid was constructed successfully. Objective To observe the inhibitory effect of xiaoaiping injection and octreotide on H22 tumor-bearing mice and find the best drug concentration,then to explore its mechanism.Methods Establish a mouse H22 subcutaneous tumor model.After tumor the experiment animals were divided into normal control group,model group,Xiaoaiping low,medium and high dose group,octreotide group,and the group of XAP low,medium and high dose groups were combined with OCT.Calculate the tumor's volume and draw the tumor growth curve.Intraperitoneal injection for 14 days,Inhibitory rate was calculated; To observe its pathological changes by light microscope; The ratio.of CD3 + NK1.1-T cells,CD3-NK1.1 + NKcells,CD3 + NK1.1 + NK-Tcells in peripheral blood were measured by flow cytometry.Results Compared with the control group,H22 liver cancer in different treatment group had a certain inhibition effect on growth,The inhibitory effect of the combination group was better than single-agent group,High-dose Xiaoaiping + octreotide was best,Tumor model group compared with normal control group,The ratio of T cells,NK cells and NKT cells was significantly lower(P <0.05) ; T cells,NK cells and NKT cells after treatment in each group had some enhancement,High-dose Xiaoaiping + octreotide was the most obvious,the ratio of T cells,NK cells and NKT cells of the combination group was significantly more than the single-drug group and the same concentration of octreotide monotherapy Xiaoaiping group(P < 0.05).Conclusion High-dose Xiaoaiping + octreotide is the best drug for the inhibitory drug concentration.The inhibition of tumor growth may pass to improve the tumorbearing mice with immune status and enhance the body's anti-tumor capacity.
7.Experimental study on the inhibitory effects of verapamil on the proliferation of meningiomas cells.

In order to investigate the effects of verapamil on the proliferation of meningiomas cells in vitro and in vivo, the cultured meningiomas cells were cultured with verapamil at different concentrations for 24 h and the inhibitory effects of verapamil on cell proliferation were observed by MTT method. The meningiomas model was established by implanting the newly removed tumor fragments into the nude mice subcutaneously. The nude mice with tumors were divided into two groups: verapamil-treated group and control group. Tumor volumes were measured and after 12 weeks the tumors were taken out and examined histologically. The expression of proliferating cell nuclear antigen (PCNA) in the tumors was detected by using immunohistochemistry. It was found that verapamil could inhibit the growth of cultured meningiomas cells in a concentration-dependant manner. The inhibitory effect could be observed in the concentration of 1 micromol/L verapamil and the most obvious effects appeared in the concentration of 100 micromol/L. Tumor volume in the verapamiltreated group was obviously smaller than that in the control group (211.40+/-5.50 vs 163.94+/-3.62, P<0.01) and the expression of PCNA was also lower (1.52+/-0.24 vs 2.86+/-0.53, P<0.05). Tumor inhibition rate was about 22.45%. It was suggested that verapamil could inhibit the proliferation and growth of meningiomas cells in vitro and in vivo.

10.Anatomic basis of sensation restoration of distally based island flap pedicled with nutrient vessels of superficial peroneal nerve

Objective To provide anatomic basis for sensation restoratiou of distally based island flap pedicled with nutrient vessels of superficial peroneal nerve by use of lateral sural cutaneous nerve. Methods The origin, course and distribution rule of lateral part sensory nerve of leg were dissected and observed in 40 antisepticized adult cadaveric limbs. Results ①Lateral sural cutaneous nerve originated from common peroneal nerve 7cm above apex of fibular head, descended short distance along common peroneal nerve, then passed through pepliteal fascia to facies lateralis cruris,along the way it sent out 1-3 terminal branches, which distributed over the skin of Ⅰ , Ⅱ area in the posterior lateral leg. ②Superficial peroneal nerve originated from common peroneal nerve 1.9cm below apex of fibular head, descended forward between peroneus longus and fibula, then descended between peroneus longus and peroneus brevis, and sent out branches to the two muscles. The nerve bole (pure sensory nerve branch) descended straight between peroneus brevis and anterior cmral intermuscular septum, at the juncture between Ⅱ、Ⅲ area in facies lateralis eruris, passed through deep fascia to subcutaneous tissue, then sent out medial dorsal cutaneous nerve of foot and intermediate dorsal cutaneous nerve of foot, which distributed over the skin of dorsum of foot and Ⅲ area in facies lateralis cmris. Conclusion It may be available in sensation restoration of distally based island flap pedicled with nutrient vessels of superficial peroneal nerve through anastomosis of lateral sural cutaneous nerve bole with sensory nervous ramification of recipient site.
