1.Influence of different disinfection solution on infection and pain of puncture hole in patients with open fracture of both tibia and fibula treated with external fixation

Objective To discuss influence of different disinfection solution on infection and pain of puncture hole in patients with open fracture of both tibia and fibula treated with external fixation.Methods 41 patients were divided into group A(21 cases)and group B(20 cases).Group A adopted 75% ethanol to sterilize the puncture holes.While the group B used 5%iodide instead.The incidence of infection and pain of puncture holes was observed separately at the 1st week,6th week after operation and removing of fixation pin.Results The incidence of infection of puncture holes was not apparently different from each other(P>0.05)at any other time points.The incidence of paint in group B was lower than group A at the 1st week and 6th week after operation(P<0.05).The incidence of pain was higher in patients with puncture hole infection than those without puncture hole infection(P<0.01).Conclusion No difference was seen in puncture hole infection with either 75% ethanol or 5% iodide.Infection of puncture hole was correlated with pain only at the 1st week after operation.Pain could be alleviated with application of 5% iodide.
2.Study on the mechanism of SNMT on acute liver injury in mice by NF-κB signaling pathway﹡

Objective To explore whether glycyrrhizin has protective effect on acute liver injury in mice by NF‐κB signaling pathway .Methods Totally 200 Kunming mice which the body weight were about 29 -30 g were chose and divided into 4 groups randomly ,50 cases in each group .The first group was the control group;the second group was acute liver injury group induced by CCl4 ;the third group was injected with Stronger Neo‐Minophagen C(SNMC) injection on the basis of second groups;the fourth group was treated with NF‐κB inhibitor (proDTC) on the basis of the third group .After 1 ,3 ,5 d treatment ,the serum expressing levels of ALT ,AST ,total bilirubin ,albumin of mice and prothrombin time were detected ,the Child‐Pugh score was calculated ,and the pathological observation was performed .Results The results showed that ,compared to the first group ,after CCl4 treatment ,the expressing levels of AST ,ALT ,total bilirubin and albumin ,the PT and Child Pugh score all were significantly increased(P<0 .05) , which indicated that the acute liver injury model induced by CCl4 was successfully established .After the treatment of SNMT (CCl4 +SNMT) in mice ,although the level of each index did not return to normal level ,with the extension of treatment time ,the level of each index was also significantly reduced (P<0 .05) .At the same time ,this effect could be reversed by NF‐κB inhibitor proDTC .Conclusion SNMT plays a significantly protective role in acute liver injury via regulating NF‐κB signaling pathway .

4.IFN-αcould induce the expression of SAMHD1 by down-regulating miR-181a

Objective To investigate whether the expression of sterile alpha motif and histidine/aspartic acid domain containing protein 1 ( SAMHD1 ) could be induced by IFN-α and mediated by microRNA-181a (miR-181a). Methods THP-1 and Jurkat cells were treated with different doses of IFN-α(200 IU/ml and 1 000 IU/ml) for 24 h. The expression of miR-181a and SAMHD1 at mRNA level were de-tected by quantitative PCR. Western blot assay was performed to measure the expression of SAMHD1 at pro-tein level. THP-1 and Jurkat cells were transfected with p-181a, an over-expression vector of miR-181a, and then treated with 200 IU/ml of IFN-α. Changes in the expression of SAMHD1 in those cells were analyzed. Results IFN-α significantly enhanced the expression of SAMHD1 at mRNA and protein levels, but inhibi-ted the expression of miR-181a, especially in Jurkat cells. The expression of SAMHD induced by IFN-αwas inhibited in cells over-expressing miR-181a. Conclusion This study suggests that IFN-α could induce the expression of SAMHD1 by down-regulating the level of miR-181a.

Objective: To analyze the status quo of assanation and ethics courses and their social requirement, thus to explore a scientific mode for the course setting; Method: The course setting status quo of assanation and ethics is analyzed among 18 colleges in which the nursing speciality is set up. A questionnaire was also conducted to investigate students' inclination towards politics-sorted courses, and relevant data were analyzed. Results: The establishing rate of assanation courses in the higher professional colleges is higher than that in the comprehensive universities. The most favorable subjects among freshmen are the Chinese national political theories, and assanation & ethics, while the counterparts among seniors are assanation & ethics, law basis, and philosophy. Objective To investigate the effect of different HAART regimens on liver function,viral load and CD4+T cells of HIV infected children with HCV co-infection.Methods A total of 40 patients were divided into 4 groups:13 cases in AZT+3TC+NVP group(group A),16 cases in AZT+3TC+EFV group(group B),5 cases in D4T+3TC+EFV group(group C)and 6 cases in D4T+3TC+NVP group(group D).Liver function,viral load and CD4+T cells of patients in 4 groups were compared between before and after HAART.Results Eight patients(20%)had abnormal liver function before HAART treatment,and all of them had 1 level of liver damage.After receiving HAART,levels of ALT and AST were increased in group A and B(P<0.01);AST levels were increased in group C(P<0.01).CD4+T counts were all increased and HIV viral loads were decreased in 4 groups(P<0.01).There was no significant difference in RNA HCV load before and after HAART treatment(P>0.05).Conclusion Different HAART regimens can increase the liver damage in HIV infected children co-infected with HCV,but have little effect on the treatment efficiency of HAART regimens.
8.Strategy for soluble expression of phage-displayed scFv antibody specific for zebrafish vitellogenin.

We developed a new method for soluble expression of phage-displayed scFv antibody specific for zebrafish vitellogenin. The scFv antibody F5 could bind zebrafish vitellogenin specifically in phage-displayed form but not soluble form. The gene of scFv antibody F5 was cloned into vector pET 32a and transferred into Escherichia coli ori DE3. With inducible expression, soluble scFv antibody 32a-F5 was obtained successfully and could also specifically bind to zebrafish vitellogenin. The insoluble expression of phage-displayed scFv antibody was a common problem in the practical use of phage display. This study offered a feasible way to express soluble scFv antibodies with biological activity.
