1.Determination of eugenol and methylsalicylate in Suhong Tongluo Tincture by GC

AIM:To establish the method for quality control of Suhong Tongluo Tincture(eugenol and methylsalicylate). METHODS: Gas phase chromatogram approach was used to determine the content of eugenol and(methylsalicylate.) The chromatogram condition consisted of 10% PEG-20M,carrier was N2(99.999%),temperature of the entrance of the column was at 230(?C),temperature of the detector was at 250(?C);column temperature was at 70(?C)(4 min),3(?C)/min→90(?C)(4 min),then 13(?C)/min→200(?C)(3 min);0.5 ?L Sample quantity. RESULTS: The linear range of methylsalicylate was 2.41-14.21 ?g,r=0.999 7,the average recovery was (99.76%),RSD=(2.64%(n=6).) The linear range of eugenol was 2.43-14.34 ?g,r=0.999 96,the average recovery was (99.33%),RSD=2.47%(n=6). CONCLUSION: This method is simple,quick,accurate and reliable for the quality control of Suhong Tongluo Tincture.
2.Orthogonal Design on the Extraction Process for Capsule of Buyang Huanwu

Objective To choose the optimum extraction process for capsule of Buyang Huanwu.Methods With the orthogonal method,the main factors effecting the water-extraction process,such as the quantity of water,the time,pH and degree for decoction,were reviewed according the content of total glycoside and Astragalus Glycoside A.Results The optimum condition of water-extraction was extracting with 8 times amount of water for the first time and 4 times amount for the second and third time,1 hour for each time,pH was 12.Conclution The experimental results provide the basis for the water extraction process for capsule of Buyang Huanwu.
3.Research on degradation and biocompatibility in vitro and in vivo of a woven chitosan fiber

Objective To investigate the characteristics, degradation and biocompatibility of a woven chitosan fiber (CF). Methods Scanning electron microscopic observation, deteronination of density, degree of swelling and tensile strength were performed to characterize the CF. In vitro degradation was observed by immersing CF in buffered aqueous solution of pH 6 containing lysozyme at 37 ℃, while in vivo degradation was studied after intramuscular implantation of CF in rats. Furthermore, rat mesenchymal stem cells were directly cultured on CF to estimate its toxicity. Results The diameter of CF was about 2000?63?m, with a sleek surface. The density and rate of swelling were 1.093?0.27g/cm3 and 2.30?0.21, respectively. The tensile strength was 80.18?1.56MPa. Specific viscosity, measured after the addition of lysozyme to the solution, decreased dramatically within 2 hours compared with that of the control mixture without enzyme. There was obvious inflammatory reaction after intramuscular implantation in the first week after surgery, bat inflammation subsided gradually. Biodegradation was not obvious within 4 weeks, but majority of it was absorbed in twelve weeks. Cells grew well on it with normal morphology, and no inhibition of cell proliferation could be observed. Conclusion Woven CF has a good tensile strength and fairly good biodegradation and biocompatibility, and it can be used in bone and cartilage tissue engineering.
4.Survey of the KAP and its related factors on AIDS among the migrant miners in a nonferrous metal company

Objective To investigate the status and influencing factors of knowledge,attitude and practice(KAP) about AIDS among the migrant miners in a nonferrous metal company of Anhui Province and to provide evidence for constituting strategies and measures for AIDS health education and behavioural intervention.Methods 303 migrant miners in a nonferrous metal company were surveyed.Univariate analysis,Logistic regression and correlation analysis were used to explore their influencing factors.Results The general rate of awareness of AIDS was 87.9%,and the main factor affecting miners knowledge level of AIDS was their educational level.Only 55.1% of migrant miners thought people with AIDS should not be isolated;50.8% of them would like to contact people with HIV;38.0% did not object to extramarital sex.There were 32(10.6%) migrant miners admitting having had extramarital sex in the recent three months,but 11(34.4%) admitted never having used condoms,only 7(21.9%) having used each time.Multivariate analysis identified the influencing factors for condom utilization including age and attitude to people with HIV/AIDS.Conclusions ①The migrant miners have sufficient knowledge about AIDS.②The traditional sexual beliefs are changing,and an increasing number of people are tolerant to extramarital sex.Consequently,it is likely that extramarital sex might rise.③The promotion of condom use should be strengthened.
5.The suppressive effects of IFN-? on human gastric carcinoma cell line BGC-823

AIM:To observe the inhibitory effect of interferon-?(IFN-?)on the growth invasiveness and metastasis of human gastric carcinoma cell line BGC-823,and mechanism of its action.METHODS:We detected the influence of IFN-? on the proliferative ability of BGC-823 in cell culture system,the cell vitality with the MTT colorimetric assay,and the cell cycle with flow cytometer(FCM).The regulatory functions of IFN-? to the expression of E-cadherin and matrix metalloproteinase-2(MMP-2)in tumor cells were estimated by immunohistochemical analysis(S-P).The ultrastructural changes of the junction among the tumor cells were observed under electron microscope.RESULTS:IFN-? can significantly inhibit the growth of human gastric carcinoma cell line BGC-823 in a dose-dependent manner.When the concentration of IFN-? was ≥106 U/L,the cell proliferation can be effectively suppressed,the suppression rate was ≥12.2%,and the blockage appeared at the phase of G_1-S of the cell cycle.Under the induction of IFN-?,the expression level of the cell E-cadherin increased while the MMP-2 decreased.The changes on ultrastructure of the cells showed the increased adhesive junctions and the relative compact structure.CONCLUSION:IFN-? can suppress the growth of human gastric carcinoma cell line BGC-823 through its influence on cell cycle.IFN-? can regulate the expression of E-cadherin and MMP-2,make the cell junction closely,so that it has the potential on restricting the invasion and metastasis of gastric carcinoma cells.
6.Effect of laparoscopic cholecystectomy on systemic stress response

Objective To investigate the systemic stress responses of laparoscopic cholecystectomy(LC). Methods Seventy patients with cholecystolithiasis were randomly divided into two groups: LC and open cholecystectomy(OC). Peripheral blood samples were obtained during the preoperative day and 1, 3, 5, 7 day after operation. Serum cytokines levels (TNF-α, IL-1β and IL-6) and C-reactive protein(CRP) were measured. Results Changes of TNF-α and IL-1β before or after surgery in both groups showed no significant difference(P>0.05). Serum concentrations of IL-6 and CRP after surgery in both groups were significantly higher than those before surgery(P<0.05); but there were no significant difference between the two groups(P>0.05). Conclusions The systemic stress responses of LC have no significant difference comparing with OC.

8.Development of a nested PCR assay for detection of Helicobacter bilis

In this study ,the objective is to establish a nested‐PCR assay for the detection of H .bilis with high sensitivity and specificity .The nested primers were designed based on sequences of 16S rRNA gene of seventeen subtypes of H .bilis .Af‐ter optimizing reaction condition ,the sensitivity and specificity of the assay were examined via the detection of feces simulated samples ,mice infection model samples and clinic patients’ samples .The detection sensitivity of H .bilis strain for feces simu‐lated samples was 10 CFU/100 μL .H .bilis was successfully detected in the liver ,caecum and feces of experimentally infected mice .Moreover ,H .bilis was successfully detected in the bile ,cholecyst mucous membrane and feces samples from two of ten patients with cholelithiasis .Due to the PCR assay’s high sensitivity and specificity ,the method may be used to detect the infec‐tion of H .bilis .
9.Optimization of water decoction process for Guiqi Yiqi Yangxue Mixture by orthogonal test

Objective To establish ultra-high performance liquid chromatography-evaporative light scattering detector (UPLC-ELSD) method to determine the content of astragaloside in Guiqi Yiqi Yangxue Mixture and optimize the best water decoction process of the mixture.Methods The UPLC-ELSD conditions to determine the content of astragaloside in the mixture were as follow.The method was carried out on a Waters Acquity UPLC BEH C18 column (2.1 mm ×50 mm,1.7 μm) by using acetonitrile-water (35∶65) as mobile phase, and the flow rate was 0.3 mL/min and the column temperature 30℃.The parameter of ELSD:the drift tube temperature was set at 60℃, and the gas flow rate of N2 2.0 L/min.The content of paeoniflorin in the mixture was detected by previously established HPLC method.The orthogonal test was designed to optimize the water decoction process of the amount of water (A), decoction time (B) and decoction times (C), and the extract yield and the contents of astragaloside and paeoniflorin were evaluated as the comprehensive markers for the purpose.Results The UPLC-ELSD determination of astragaloside had a good linear relationship (r =0.9999), its mean recovery was 103.0%, and it had a good precision, specificity and repeatability.The orthogonal experimental results showed that decoction times (C) >the amount of water (A) >decoction time (B) were major factors affecting the decoction process in order and the ANOVA result showed that decoction times has significant effect on the decoction process (P<0.05).The optimal process of Guiqi Yiqi Yangxue Mixture was adding eight-fold water each time to decoct twice for 1.5 h totally.Conclusion This optimized water decoction process is feasible and stable, and suitable for industrialized production.
10.Detection and Genotyping of Treponema pallidum by a Nested PCR

Objective To develop a nested PCR for the detection of early syphilis and genotyping of Treponema pallidum (TP), and to investigate the distribution of genotypes of TP in Guangzhou. Methods Specimens were consecutively collected from genital ulcers of patients with suspected chancre during 2002-2004, and were detected by dark-field microscopy and nested PCR. The acidic repeat protein (arp) gene and the T. pallidum repeat (tpr) gene family were amplified with the positive specimens above. The number of repeats presented in the arp gene and the restriction fragment length polymorphism by Mse I in the tpr gene were analyzed by electrophoresis. The strains were genotyped according to Pillay's criteria. Results Out of 62 patients with suspected chancre, 33 cases (53.2%) were positive by dark-field microscopy and 54 cases (87.1%) by nested PCR. Of 47 TP-positive specimens genotyped by arp gene, 36 (76.6%) were type 14, while of 49 cases genotyped by tpr gene 39 (79.6%) were type d. By combining genotypes of arp and tpr genes, 7 genotypes were found, including 14d (31, 66.0%), 13d (5, 10.6%), 14b (4, 8.5%), 12b (3, 6.4%), 12d (2, 4.3%), 15d(l, 2.2%) and 14i (1, 2.2%). Conclusions Nested PCR shows a high sensitivity in early detection of TP. Genotype 14d seems the predominant type of TP in Guangzhou.
