1.Experimental study on reconstruction of tissue-engineered uterine patch

Objective To reconstruct tissue-engineered uterine patch similar to native endometrium and myotrium. Methods The purified rabbit uterine smooth muscle cells and stromal cells were mixed with liquid type Ⅰ collagen, and laid on mold respectively. Then the collagen-embedded cells were combined to create a cell complex, epithelial cells were inoculated on top of the complex, histological and immunohistochemical staining, and scanning electron microscopy were used to analyze the engineered uterine tissue. Results The engineered uterine tissues (EUTs) were reconstructed with isolated and cultured uterine smooth muscle cells, stromal cells and epithelial cells. After cultured for 14 days, histological and immunohistochemical analyses showed that the smooth muscle cells and stromal cells in the EUTs distributed evenly in the two layers and the majority of the cells, with elongated nuclei, stretched along the stretching direction. The morphology of EUTs resembled that of the native uterine tissue. In scanning electron microscopy, some epithelial cells protruded pinopodes. The surface of epithelial cells was covered abundantly with microvilli and some cells were ciliated. Conclusion Uterine smooth muscle cells and stromal cells were mixed with Liquid type Ⅰcollagen respectively, and epithelial cells were inoculated on top of the complex for generation of EUTs similar to the native uterine tissue.
2.Vertebroplasty with vertebral pedicle screw plus calcium sulfate cement injection treats thoracolumbar vertebrae fractures:report of 28 cases

Objective To evaluate the clinical efficacy of vertebroplasty using vertebral pedicle screw technique combined with calcium sulfate cement injection in the treatment of thoracolumbar vertebrae fractures.Methods Twenty-eight patients with thoracolumbar vertebrae fractures admitted in our department in recent 2 years were enrolled in this study,including 21 cases of type A,3 cases of type B,and 4 cases of type C.All patients were fixed with vertebral pedicle screw,and then verteplasty was performed using calcium sulfate cement injection.Results All patients were followed up for an average time of 16 months.Within the follow-up,there was no complication noted,such as loosening or breakage of internal fixation,chronical lumbar back pain,and loss of effected vertebral height.Artificial bones injected into vertebrae were absorbed in about 3 months.Conclusion Vertebroplasty using vertebral pedicle screw technique combined with calcium sulfate cement injection is an effective and safe procedure for thoracolumbar vertebrae fractures,which renders it possible to bear weight early and to maintain corrected vertebral height postoperatively.

The proteins and antigens of nine strains of Toxoplasma gondii were analyzed by SDS-PAGE and immunoblotting. These strains including RH strain, Zs-2 strain, CN strain and SHI, SH2, SH3, SH4, SH5, SH8 strains, were obtained from other laboratories or isolated from deformity fetus in our laboratory. SDS-PAGE analysis revealed that these strains were very close in the major bands of proteins and differences were observed only in part. There were common components in antigens of all strains using IgG antibody prepared from high-titre rabbit antisera raised against RH strain of Toxoplasma.

5.A comparison between laparoscopic hepatectomy and open hepatectomy for the treatment of hepatocellular carcinoma

Objective To investigate the efficiency and safety of laparoscopic hepatectomy (LH) and open hepatectomy (OH) in patients with hepatocellular carcinoma (HCC). Methods A total of 45 HCC cases undergoing hepatectomy from January 2008 to December 2010 in our hospital were divided into LH group ( n =21 ) and OH group ( n =24 ).Their clinical efficiency and safety were compared and analyzed. Results The amount of intraoperative blood loss and blood transfusion in LH group were significantly higher than that of OH ( respectively x2 =3.973,x2 =4.862,all P < 0.05 ).LH is associated with less postoperative complications and duration of hospital stay,compared with OH ( respectively x2 =4.746,t =-2.717,all P <0.05).Cumulative survival rates in both groups were not statistically significant ( x2 =0.172,P > 0.05 ).However tumor-free survival rate in LH was significantly higher,superior to that of OH (x2 =4.543,P < 0.05). Conclusions LH and OH are all safe and effective hepatectomy for HCC.LH has the advantage of less complication and shorter hospital stay.
6.CPC derivedexosome protects cardiomyocytes from oxidative stress

Aim To explore the anti-apoptotic function of cardiac progenitor cells(CPCs)-derived exosome in vitro.Method CPCs were isolated from mouse heart using Magnetic Cell Sorting(MACS)system.Flow Cy-tometry(FC)determine the purity of stem cell surface antigen-1 positive(Sca-1 +)CPCs.Exosome was puri-fied from conditional medium,and confirmed by West-ern blot using CD63 as a marker,Nanoparticle Traffic-king Analysis(NTA)was used to detect the diameters and concentration of exosome.Then the cells were di-vided into control groups and CPC-exosome pre-protec-tion groups.H2 O2 was added into H9c2 cells to induce oxidative stress.Western blot was adopted to determine the expression of cleaved caspase-3.Results ① Im-munofluorescence showed that CPCs isolated by MACS were positively expressing Sca-1 protein;FC analysis showed that typical purity of Sca-1 +CPCs from the first
preparations was more than 95%.② WB demonstrated that CD63 of exosome isolated from CCMwas positively expressed,and NTA results showed that the diameters of exosome were (82.33 ±3.06)nm(n =3).Micro-scope detected PKH-26 labeled exosome appeared in the cytoplasma of H9c2 cells.③ Western blot showed the CPC-exosome pre-protection groups significantly down-regulated the levels of cleaved caspase-3 com-pared to the control groups(P <0.05).Conclusion CPC can secrete exosome which carries many important cargos,which can effectively gather in H9c2 cells. CPC-exosome can protect H9c2 cells from the oxidative stress induced by H2 O2 .Our results highlight a new perspective strategy for cardiac disease.
7.Stellate ganglion block treatment of OSAS combined curative effect observation of patients with high blood pressure

Objective To observe the stellate ganglion block (SGB) on obstructive sleep apnea syndrome (OSAS) combined the curative effect of sleep respiration and blood pressure control in patients with hypertension.Methods Incorporating meets the criteria for the OSAS patients with high blood pressure in hospital order randomly assigned into normal group and experimental group and routine group was given antihypertension drugs,adjustment in lifestyle,continuous positive airway pressure (CPAP) treatment,the experimental group on the basis of conventional treatment at the same time give SGB to intervene.Using t test on admission and intervention were compared after a period of treatment in patients with sleep apnea and blood pressure control,using 2 test comparison blood pressure control rates of two groups patients.Results Compared with normal group,the experimental group after intervention in a course of apnea hypoventilation index (AHI),SaO2 and 24 h mean arterial pressure were obviously improved,the difference was statistically significant (P<0.05).Conclusion SGB as a new treatment method,not only can improve clinical symptoms in patients with OSAS,but also make the patients get better control of blood pressure.
8.Drug Resistance and Homology Analysis of Multi-drug Resistant Acinetobacter Baumannii in Orthopaedic Hospital

Objective To investigate the drug resistance and homology status of Multi-drug Resistant AcinetobacterBauman-nii (MDR-AB)in orthopaedic hospital.Methods 34 strains MDR-AB were isolated from 2016.1~2016.7 for DNA extrac-tion and were typed by repetitive extragenic palindromie-polyrnerase chain reaction (REP-PCR).Results The resistance rate of MDR-AB were ≥70% to 15 of 17 antimicrobials,except to cotrimoxazole (14%)and levofloxacin (61%),34 strains of MDR-AB were divided into three types by REP-PCR including typeⅠ,typeⅡ and typeⅢ.Conclusion Drug resistance of MDR-AB was severe and mainly composed of the same genotype (typeⅠ).Rational use of antimicrobial and regular mo-nitoring of drug resistance is necessary to reduce the nosocomial transmission.
9.Large diameter esophagogastric internal fistula combined with fundoplication for treatment of achalasia

Objective To explore the clinical the efficacy of large diameter esophagogastric internal fistula combined with fundoplication and modified Heller surgery for treatment of achalasia.Methods From June 2008 to March 2014,18 patients diagnosed as achalasia were se-lected into this suty,and they were divided into the observation group (8 cases)and the control group (10 cases).Patients of the observation group were received large diameter esophagogastric internal fistula combined with fundoplication while patients of the control group received modified Heller surgery.Compared the surgical curative effect and complications between the two groups.Results All the 18 patients suc-cessfully completed the arranged surgical treatment.The imaging examination 3 months after the operation showed that the contrast agent suc-cessfully passed through the esophagus,cardia and the anastomosis,and then went into the gastric lumen.The total effective rate was 100% in the observation group 12 months after operation,while it was 60% in the control group,and the difference between the two groups was statisti-cally significant (P =0.014).The incidence of complications in the observation group was lower than that of the control group,the difference was statistically significant (P =0.026).Conclusion Large diameter esophagogastric internal fistula combined with fundoplication for treat-ment of achalasia is safer,and there is a possibiltiy to cure the achalasia.
10.Endoscopic treatment for acute appendicitis

Objective To study the effect of endoscopic treatment for acute appendicitis (AA)without perforation or gangrene.Methods A total of 94 patients with AA were randomly divided into operation group (n =45 ) to receive appendectomy,control group (n =15 ) to accept conventional medicine of metronidazole and Cefoxitin and colonoscopy group (n =34) to undergo conventional medicine plus endoscopic treatment.The time for alleviation of abdominal pain,duration and mean cost of the hospitalization,and the recurrence rate in one year were compared.Results Compared to operation group,colonoscopy group was superior in the duration [ (2.77 ± 0.27) d vs.(6.65 ± 1.68) d ] and mean cost of hospitalization [ ( 1011.35 ± 22.12) yuan vs.(4023.37 ± 32.02 ) yuan ] ( P < 0.05 and P < 0.01,respectively).There were no significant differences in the time for alleviation of abdominal pain or the recurrence rate in one year between 2 groups.Colonoscopy group was superior to control group in all the indices (P < 0.05 ).Conclusion Endoscopic treatment for AA without perforation and gangrene is effective and safe,which can be considered as the first-line treatment.
