1.EFFECT OF GYNOSTEMMA PENTAPHYLLUM ON THE LIFE SPAN OF FRUIT FLY

The influence of Gynostemma pentaphyllum extract ( G. P ) on the life span of fruit fly was observed. G. P (0.5% and 1.0%) significantly increased mean life span by 11.8% and 12.2%, respectiyely, when fed to male adults.At 0.25% concentraction, the female adults were found to yield an increase of 24.1% in mean life span and the male an increase of 18.5%.Rearing flics on 0. 5% G. P during both the developmental and adult stages proved to be even more effective than just feeding during the adult stage. The mean life span of male and female adults were increased by 53.3% and 46.1% , respectively, but at the higher concen-tration of 1.0%, the increase was 14.4% and 9.7%, respectively,G. P obviously improved the growth of larvae when given aconcentration of 0.5%,No marked change was observed in the copulatory rate of fruitflies after the treatment of 0.5% G.P.
2.Advances on pharmacotherapy for erectile dysfunction

It is reported that plenty of men,especially the aged,have been bothering in penile erectile dysfunction(ED).There are series of treatments for ED.Oral erectogenic agents have become the first-line treatment options,including drugs that act on the central and peripheral nervous systems,testosterone,traditional Chinese medicine and so on.Sidenafil,apomorphine and yohimbine are the only three oral therapies currently available for the treatment of this disorder.The second therapy includes intracavernosum injection,transurethral medications,transdermal medications etc.Intracavernosum injection of PGE 1 is an effective approach at the present.With the rapid understanding of erectile physiology and etiology of ED and the introduction of new effective oral agents,the future of pharmacotherapy for ED will extend.In this review,we have highlighted the advances in current treatments and therapeutic approaches in male erectile dysfunction.
3.Radio frequency ablation combined with chemotherapy in the treatment of clinical research of lung tumors

Objective:To research on the radio frequency ablation (RFA) combined with the clinical effect of chemotherapy in patients with advanced non small cell lung cancer and lung metastasis. Methods:From 2011 May to 2012 in our hospital 06 months 60 cases of metastatic non-small cell lung cancer and lung cancer patients, divided into radio frequency ablation(RFA) combined with chemotherapy group of 30 cases and a simple chemotherapy group 30 cases, referred to as the treatment group and the control group. To observe the change of tumor volume in patients with poor survival rate, years, tumor recurrence rate, and postoperative complications. Results:Three months after treatment in treatment group tumor recession(CR) in 5 cases(16.7%), part of the recession(PRa) 18(60%), partial mild recession(PRb) 6(20%), no degradation(NR), 1(3.3%), tumor size change significantly better than the control group. The treatment group patients 12 months survival rate was 86.7%, higher than that of the control group(70%). The treatment group of 12 months of tumor recurrence rate of 6.7%, lower than that of the control group(26.7%). No obvious adverse complications of the treatment group of 30 patients after operation. Conclusion:Radiofrequency hyperthermia ablation combined with chemotherapy in treatment of advanced non small cell lung cancer and metastatic lung cancer clinical effect is obvious.
4.The infectivity of the recombinant vaccinia virus expressing human interleukin-2 and its effect on MKN45 gastric cancer cells in vitro

Objective To study the effect of the recombinant vaccinia virus expressing human interleukin 2 on MKN45 gastric cancer cells in vitro. Methods Recombinant vaccinia virus expressing hIL 2 (VMJ601hIL 2) was constructed by homologous recombination using molecular virology, and VMJ601hIL 2 was detected by DNA hybridization technique and the recombinant gene product was analyzed by SDS PAGE. In addition, MKN45 gastric cancer cells was infected by VMJ601hIL 2 and the effect of VMJ601hIL 2 on the gastric cancer cells was evaluated in vitro. Results hIL 2 protein with biological activity can be secreted by MKN45 gastric cancer cells after heavily infected by VMJ601hIL 2. Conclusions It is one of the crucial steps that VMJ601hIL 2 has been constructed and identified since it forms essential prerequisite for further in vivo gene therapy of gastric cancer.
5.Clinical value of laparoscopic surgery by pull through technique for anterior resection of rectum in patient with rectal cancer

Objective To compare the clinical values of laparoscopic surgery and laparoscopic surgery by pull through technique for anterior of rectum in patients with rectal cancer. Methods From Jan, 2013 to Jan, 2014, 120 patients with low rectal cancer were prospectively collected. The patients were randomly divided into study group (n= 60) and control group (n= 60). Patients in the study group were treated with laparoscopic surgery by pull through technique for anterior resection of rectum, while patients in the control group were treated with laparoscopic surgery. The primary outcomes were intraoperative situations, postoperative complications and recovery. Results When compared with the control group, patients in the study group got a significantly shorter operative duration [(132.32±14.92) vs (154.73±17.65) min, P=0.000];a signiifcantly lower postoperative drainage volume [(299.93±56.49) vs (365.24±68.94) ml, P= 0.000]; a significantly shorter gastrointestinal function recovery time [(57.42±5.84) vs (61.85±7.40) h, P=0.002]; and a signiifcantly less of length of hospital stay [(12.54±2.76) vs (14.75±2.10) d, P= 0.000]. There was no significant difference between the two groups in the amount of bleeding, the number of lymph node dissection, pneumonia, lower extremity arteriovenous thrombosis, incision infection, anastomotic leakage, intestinal obstruction, 2-year recurrence rate and mortality rate (P> 0.05). Conclusion laparoscopic surgery by pull through technique for anterior resection of rectum can accelerate postoperative recovery.
6.Preliminary study on the value of laparoscopic ultrasound in urinary laparoscopic surgery

Objective To evaluate the clinical value of laparoscopic uItrasonography ( LUS) in u-rinary laparoscopic surgery .Methods In 6 cases of urological laparoscopic surgery , laparoscopic ultra-sound examination were taken , including 3 cases of simple renal cysts , 1 case of adrenal pheochromocyto-ma, 2 cases of hamartoma , were excised under the LUS guidance and the results were compared with the traditional simple laparoscopic surgery .Results With laparoscopic uItrasonography guidance , the bounda-ry and blood supply of cyst or tumor could be shown more clearly compared with trans -abdominal ultrasound . We could differ the tumors from dilated pelvis and renal calices , So the damage to the collecting system and renal blood vessels were avoided .With LUS guidance , tumors were excised more completely , so that re-mained tumor tissues would be avoided and normal renal tissues would be protected .Conclusion LUS may provide more precise surgical information and may play an important role guiding laparoscopic operation in urinary surgery .
7.Factors influencing the treatment effect of cancer pain in patients with moderate and severe chronic pain

Objective:This study aimed to analyze the factors affecting the outcome of cancer pain in patients with moderate and severe chronic cancer pain for clinical decision making. Methods: Data were collected from 426 cancer patients with moderate and severe chronic cancer pain, and the factors affecting pain treatment were analyzed. Results:A total of 85.6%of patients had good pain control in 3 days (NRS≤3). Multivariate logistic regression models showed that the pain of patients with bone metastases (P=0.026), breakthrough pain after stable pain control (P<0.001), and high MEDD (P<0.001) was poorly controlled. Moreover, age, sex, TNM stage, cause of pain, and medication ladder were not risk factors of pain control (P>0.05). Opioid combination with NSAIDs contributed to easier pain control (P=0.024). Digestive system tumors, pain intensity, limb pain, neuropathic pain, use of transdermal fentanyl matrix patch, multiple metastases in stage-IV patients were suggested to be risk factors of pain control in univariate logistic regression models (P<0.05). Conclusion: Bone metastases, breakthrough pain after pain relief, and high dose of MEDD were independent risk factors. Opioid combination with NSAIDs was a protective factor of pain control.
8.Construction of Dickkopf1-expressing recombinant adenovirus and its effect on ?-catenin expression in A375 cells

Objective To construct a recombinant adenovirus vector expressing Dickkopf1 (DKK1) (Ad-DKK1) and study the expression of ?-catenin in melanoma cells A375 after Ad-DKK1 infection. Methods DKK1 gene was amplified by PCR and subcloned to shuttle plasmid Adeasy by molecular cloning and adenovirus recombination technique,then converted in BJAdeasy cells. The recombinant adenovirus plasmid was generated. After cut by PacⅠ enzyme,the recombinant adenovirus plasmid Ad-DKK1 was packaged and amplified in HEK293 cells. Ad-DKK1 effect group,Ad-SimDKK1 effect group,Ad-GFP effect group,Ad-RFP effect group and blank group were used to infect the A375 cells in multiplicity of infection (MOI) 80,and cell morphology was observed. The expression of DKK1 and ?-catenin in A375 cells was detected by RT-PCR. Cell vitality was detected by MTT assay. Results Ad-DKK1 was proved to be recombined successfully by identification with restriction enzyme and sequencing. The infection was efficient (80%) in A375 cells. Objective To study the pharmacokinetics of ferulic acid (FA) in plasma of the healthy female volunteers after oral administration of Shenghua decoction.Methods Using p-hydroxybenzaldehyde as the internal standard, the plasma concentration of FA was determined by RP-HPLC. Plasma samples were extracted and treated with boiling water and 3P97 programme was used to calculate the pharmacokinetic parameters. Results The main pharmacokinetic parameters of FA were as follows: T1/2?=18.72 min ,T1/2?=79.21 min,T1/2 Ka=11.19 min,AUC=18004.87 ?g?min?L-1,CL=0.17L?min-1?kg-1,Cmax=206.30 ?g?L-1, Tpeak=22.78 min.Conclusion After oral administration of Shenghua decoction, FA could be absorbed and eliminated rapidly and pharmacokinetics of FA conforms to a two-compartment open model.
10.Expressions and Clinical Significance of Runx3,Smad4,Cdk2 and p21 in Gastric Cancer

Background:Tissue microarray has been increasingly used in research of malignancies. It has been revealed that TGF-β signaling pathway contributes to the tumorigenesis and progress of malignancies. Aims:To determine the expressions of Runx3,Smad4,Cdk2 and p21,the key molecules in TGF-β signaling pathway by tissue microarray,and investigate their correlations with clinicopathological features and prognosis of gastric cancer. Methods:A total of 378 paraffin embedded tissue blocks,including 130 gastric cancer tissue and 248 para-cancer tissue from 130 patients undergoing radical resection of gastric cancer were obtained. Tissue microarray and immunohistochemistry were used to determine the expressions of Runx3,Smad4,Cdk2 and p21. Results:The aberrant expression rates of Runx3,Smad4, Cdk21 and p21 in gastric cancer tissue were significantly higher than those in para-cancer tissue(67. 7% ,35. 4% , 63. 8% and 70. 0% vs. 14. 1% ,12. 5% ,18. 1% and 37. 1% ,P < 0. 05,respectively). Aberrant expression of Runx3 was closely correlated with histological grade and lymph node metastasis of gastric cancer( P < 0. 05),while aberrant expressions of Smad4 and p21 were correlated with histological grade only(P < 0. 05);aberrant expression of Cdk2 was correlated with histological grade,lymph node metastasis and TNM staging(P < 0. 05). Pairwise correlations were seen among aberrant expressions of Runx3,Smad4 and p21 in gastric cancer,while Cdk2 was correlated with Runx3 only. Kaplan-Meier survival curve showed that 5-year survival rates in Runx3,Smad4 and p21 aberrant expression groups were significantly lower than those in normal expression groups(P <0. 05). Furthermore,Cox proportional hazard model indicated that Runx3 and Smad4 were independent prognostic factors for gastric cancer. Conclusions:Runx3,Smad4,Cdk2 and p21 might play pivotal roles in tumorigenesis and progression of gastric cancer. As interactions occurred among these four proteins,whether Runx3 and Smad4 could be used for predicting the prognosis of gastric cancer needs to be further studied.
