2.The Study on the Relationship between Hepatic Resection Hepatic Regeneration and Hepatic Function by CT and Hepatic Function Test

Objective To study the relationship between hepatic resection,hepatic regeneration and hepatic function.Methods Totally 8 cases undergone partial hepatic resection were undergone CT scan pre-and pastoperation.The volumes of the whole liver and the lesions were measured on CT images,and the hepatic resectable rate was calculated before operation.The volume of liver was measured again after operation,and the hepatic regenerative rate at different time was also calculated after operation.The hepatic function was tested before and after operation.Results The negative relation was showed between hepatic resectable rate and hepatic regenerative rate.Conclusion There is postiive relation between hepatic resectable rate and the function of liver.
3.Detection of DNA Damage of p53 Induced by Organic Concentrate of Drinking Water Using Terminal Linker,Dependent PCR

Objective To detect DNA lesions induced by complex pollutants existing in environment using the technique of randomized terminal linker,dependent PCR (RDPCR). Methods SD rats were treated with organic concentrate by injection i.p., DNA were extracted from rat's liver, kidney, lung and leucocytes, then RDPCR was used to detect DNA damage of exon 7 of p53 gene. Results Organic concentrate of drinking water could make rat's genomic DNA broken. Hybridization bands were found in liver and kidney tissues after RDPCR, none in blood and lung tissues. Conclusion Organic concentrate of drinking water can cause the DNA lesions of exon 7 of p53 gene and the major target organs are liver and kidney. The technique of RDPCR can be used to detect DNA damage induced by complex environmental pollutants.
4.Literature Analysis of 31 Cases of Drug-induced Lupus Induced by Infliximab

OBJECTIVE:To analyze the general patterns and characteristics of drug-induced lupus (DIL) induced by inflix-imab,and to provide reference for the safety of drug use. METHODS:Usinginfiximab,lupusas searching words,inflix-imab-induced DIL literatures included in CJFD,VIP and PubMed from Jan. 2002 to Jun. 2016 were retrieved and analyzed. RE-SULTS:Totally 30 effective literatures were retrieved,involving 31 DIL patients,there were 8 male cases (25.81%) and 23 fe-male cases(74.19%);17 patients aged more than 50(54.84%). 20 cases occurred within 5-24 weeks(64.51%). 16 patients re-ceived infliximab alone (51.61%). Main clinical manifestations were skin rash or photosensitivie enteritig (15 cases),joint pain (16 cases,multiple arthritis and synovitis)and fever(12 cases). 5 patients suffered from 3 above symptoms simultaneously. Labo-ratory examination mainly manifested as antinuclear antibody,double stranded DNA antibody and anti-histone antibody positive. Thirty patients with DIL received drug withdrawal or steroid hormones treatment,among which the symptoms of 26 patients disap-peared after drug withdrawal or treatment within 6 months,and 1 patient deteriorated. CONCLUSIONS:DIL-induced by inflix-imab invovle patients'age and gender,disease types,family history and other factors. Clinicians should be aware of rules and char-acteristics of DIL induced by infliximab,and tighten drug use monitoring to reduce ADR.

6. Research on the mechanism of dioscin-containing Serum against apoptosis of cardiomyocytes induced by hydrogen peroxide

OBJECTIVE: To discuss the mechanism of dioscin against apoptosis of cardiomyocytes induced by hydrogen peroxide through Serum Pharmacologic Method. METHODS: The drug-containing serum [0.6 g (crude drug) · kg-1] was prepared by serum pharmacologic method. Cardiomyocytes from neonatal SD rats were cultured in Dulbecco Modified Eagle Medium (DMEM). The primary cultured cardiomyocytes were injured by hydrogen peroxide before giving the drug-containing serum with different concentrations. The cardiomyocyte viability was determined by MTT method. Morphological changes of cardiomyocytes were observed by fluorescence microscope after treating with the drug-containing serum at different concentrations. The anti-apoptotic effect of dioscin was indicated by detecting the activity of caspase-3. The protein expression of Bcl-2 and Bax were semi-quantified by Western-blot method after treating with the drug-containing serum. RESULTS: The cardiomyocyte viability was elevated (P < 0.01) after being treated with different concentrates of drug-containing serum (0.4, 0.8, 1.2 g (crude drug) · kg-1, prepared from the original drug-containing serum). Typical apoptotic features of the cardiomyocytes such as membrane blebbing, cell shrinkage and detachment, and nucleus condensation and fragmentation were dose-dependently improved after being treated with drug-containing serum. The activity of caspase-3 decreased with the increasing concentration of drug. Western blot approach showed that the Bcl-2 level increased meanwhile the Bax decreased. CONCLUSION: Dioscin could increase the viability of the primary cultured cardiomyocytes in hydrogen peroxide induced injury. It could also decrease the activity of caspase-3, improve nucleus condensation and fragmentation, increase Bcl-2 level and decrease the Bax level. It is considered that the central nervous system (CNS) is an immunologically privileged organ, but its immunological privilege is not complete, and immunological rejection may also occurred after tissue transplantation. Neural stem cell (NSC) transplantation has developed a brand-new approach for the treatment of various CNS diseases. Despite the low immunogenicity of NSC, there are also troubles of immunological rejection. This article reviews the immunological characteristics of CNS, the mechanisms of immunological response and immunological rejection in CNS, as well as the problems of immunological rejection of NSC transplantation.
9.Influence of estrogen on the proliferation of MCF7, a human breast carcinoma cell line

Purpose: To study the influence of estrogen on the proliferation of human carcinoma cells. Methods: Treated with different concentrations of estrogen, the influence of estrogen on MCF7 was detected by MTT test. Several proteins related with cell cycle, tumor suppressor, proliferation and apoptosis were detected by the method of Western blot. Results: MTT test showed that estrogen could promote the proliferation of MCF7. Cyclin Dl was increased at the same time. But the protein level of PTEN, a tumor suppressor gene, was not changed by estrogen. Furthermore, as an anti -apoptotic protein, PKB protein level was also unchanged. Conclusions: Estrogen can promote the proliferation of human carcinoma cells by certain pathways including cyclin Dl, but not including PKB.
10.MLVA based Mycobacterium tuberculosis clinical isolates genotyping analysis f rom HIV co-infection patients in Dali area,China

A new method based on the multiple locus variable number tandem repeat analysis (MLVA) was applied for the genotyping of combined HIV Mycobacterium tuberculosis in Dali to investigate the genotyping and distribution pattern of Myco‐bacterium tuberculosis clinical isolates with MLVA .Mycobacterium tuberculosis clinical isolates were selected from Dali area , and the polymorphism of VNTR locus was tested with PCR .The clustering of genotype was analyzed by BioNumerics (6 .6) . Result showed that 15 VNTR loci of 61 combined HIV Mycobacterium tuberculosis clinical isolates were analyzed respectively . There were obvious polymorphisms of VNTRs .The discrimination power of these loci appeared different from each other ,with the biggest Hunter‐Gaston index (0 .839) loci was MIRU26 ,and the smallest one (0 .341) loci was MIRU4 .The clustering of genotype showed that these strains could be categorized into 5 gene clusters and 61 genotype ,the proportions of cluster Ⅰ was the biggest one ,51 .6% were cluster Ⅰ which including 32 strains .The standard strain H37Rv was belongs to cluster Ⅱ .Its indicated that there are obvious polymorphisms of VNTRs of combined HIV Mycobacterium tuberculosis clinical isolates in Da‐li .The main genotype was Beijing family genotype .
