1.Gene mutations of human immunodeficiency virus drug resistance from men who have sex with men in Shanghai,2013

Objective To investigate the gene mutations of human immunodeficiency virus (HIV)‐1 drug resistance among anti‐retrovirus (ARV ) treated‐naive men who have sex with men (MSM ) in Shanghai to provide evidence‐based data for optimized treatment .Methods All 669 treatment‐nave cases of HIV‐1 infection identified among MSM in 2013 were recruited and their plasma was collected .RNA was extracted and amplified by nest reverse transcription‐polymerase chain reaction ,and DNA was sequenced and then phylogenetically analyzed .Finally ,subtypes were identified and drug resistance was analyzed in comparison with International HIV Drug Resistance Database .Results The pol gene fragments of 645 cases were obtained .Primary drug‐resistance rate was 2 .48% (16/645) ,including mutations conferring resistance to protease inhibitor (PI) (0 .31% ,2/645) ,nucleoside reverse transcriptase inhibitors (NRTI) (0 .16% ,1/645) ,non‐nucleoside reverse transcriptase inhibitors (NNRTI) (1 .70% ,11/645) and both NRTI and NNRTI (0 .31% ,2/645) ,respectively .Mutations conferring resistance to CRF01_AE were 12 cases (2 .99% ) ,while mutations conferring resistance to CRF07_BC and CRF_01B were 0 .61%(1/163) and 4 .65% (2/43 ) including 1 case of CRF52_01B and unidentified CRF_01B , respectively . Resistance to NNRTI in B subtype were 2 .70% (1/37) .Conclusion The prevalence of HIV‐1 drug resistance‐associated mutations among MSM in Shanghai ,2013 is still low ,but resistance to NNRTI is relatively high .CRF01_AE is the major subtype of drug resistance .It is necessary to strengthen the HIV drug resistance surveillance in MSM group in Shanghai .
2.Changes of serum markers in acute liver injury induced by oral 1, 2-dichloropropane in BALB/cA-nu mice

Objective:To investigate the characteristics of inflammatory response in BALB/cA-nu mice induced by oral 1, 2-dichloropropane (1, 2-DCP) , and to provides theoretical reference for further study of subchronic, chronic toxicity and carcinogenic mechanism.Methods:From October 2018, Clean grade healthy BALB/cA-nu mice were randomly divided into 5 groups with 10 mice in each group. And 860, 1150, 1500, 1950, 2535 mg/kg 1, 2-DCP were given by gavage respectively. Meanwhile, blank group and solvent control group (corn oil) were set up. Blood samples were collected from eyeballs and liver and bile tissues were collected for histopathological examination within 24 hours after exposure. The expression of alanine aminotransferase (ALT) , total bilirubin (TBLI) , C-reactive protein (CRP) , interleukin-6 (IL-6) , tumor necrosis factor-α (TNF-α) and tumor necrosis factor-β (TNF-β) were detected by enzyme-linked immunosorbent assay.Results:With the increase of the dose of 1, 2-DCP, the number of microbubbles in liver cells and the infiltration of inflammatory cells increased gradually. No pathological changes were found in the gallbladder. Compared with the blank group and solvent control group, the content of serum ALT in each exposure group was increased, the serum levels of IL-6 and TNF-β in 860, 1150, 1950 and 2350 mg/kg exposure groups were increased, the serum TNF-α and TBLI levels in 1 950, 2535 mg/kg groups were significantly higher ( P<0.05) . The levels of ALT, TBLI and TNF-β in serum of female mice were significantly different ( P<0.05) . There were significant differences in ALT, TBLI, CRP, IL-6, TNF-α, TNF-β in serum of male mice ( P<0.05) . Conclusion:Oral 1, 2-DCP may cause acute liver injury in BALB/cA-nu mice and increase the expression of serum inflammatory factors. Moreover, the types of inflammatory factors activated in male mice are more than those in female mice.

4.Expert consensus on the clinical application strategy of NIPT2.0, a new-generation non-invasive prenatal screening technology

The new-generation non-invasive prenatal screening technology (NIPT2.0) is a new method successfully realized in recent years based on high-throughput sequencing to synchronously and accurately detect fetal chromosomal aneuploidies, microdeletion/microduplication syndromes and dominantly inherited monogenic disorders. NIPT2.0 can circumvent the shortcomings of previous non-invasive prenatal screening techniques (NIPT and NIPT Plus) including incapability to detect fetal monogenic disorders, insufficient accuracy of detection and low positive predictive values for certain chromosomal abnormalities (in particular trisomy 13, sex chromosomal abnormalities, and small-segment microdeletions and microduplication syndromes). How to apply NIPT2.0 reasonably and normatively to maximize its clinical value has become an issue which requires clarification. The Reproductive Health Branch of the Chinese Maternal and Child Health Care Association has organized experts to fully discuss and jointly drafted this consensus, which has put forwards suggestions over the clinical application strategy for NIPT2.0, including the scope of application, target disease, pre-test consultation, clinical application pathway, post-test genetic counseling and intervention, quality control and limitations, for the reference by peers, with a view to standardize its application and provide better clinical service.
5. Evaluating the level of occupational stress and its influence factors among traffic police in a district in Shanghai

Objective:
To investigate the high occupational stress and its influencing factors in traffic police in Shanghai.
Methods:
728 traffic police were selected as the study subjects, and the《Occupational Health Questionnaire》was used to investigate and evaluate their job demand-control (JDC) and effort-reward imbalance model (ERI) occupational stress situation respectively. The related influencing factors were analyzed.
Results:
The prevalence rates of high occupational stress in JDC and ERI models were 74.6% (543/728) and 51.5% (375/728) . The influencing factors of JDC were education, marriage, average weekly hours (χ2=16.82, 10.04, 18.71,
