1.Design and Verification of Lung Diffusion Function Detection System.

A lung diffusion function detection system is designed. Firstly, the controllable collection of air, test gas source and calibration gas source was based on single-breath method measurement principle. Secondly, pulmonary diffusing capacity for carbon monoxide (DlCO) was calculated by gas concentration measured by the non-dispersive infrared sensor to measure, the gas flow measured by the differential pressure sensor, and the temperature, humidity and atmospheric pressure sensors to test and evaluate the quantitative detection and evaluation of lung diffusion function. Moreover, a preliminary verification of the lung diffusion function detection system was implemented, and the results showed that the error of the lung carbon monoxide diffusion and the alveolar volume did not exceed 5%. Therefore, the system has high accuracy and is of great value for early screening and accurate assessment of COPD.

2.Clinical research of approach selection of extraction of maxillary embedded mesiodens

Objective :
To explore the related factors for the unilateral flap and bilateral flap by changing the original operation plan in the extraction of maxillary impacted mesiodens.
Methods :
81 patients with impacted mesiodens in the middle of the maxillary were retrospectively analyzed. The primary outcome variables were planned surgery (unilateral flap)and unplanned surgery (bilateral flap) . The secondary outcome variables consisted of operation time and postoperative swelling. The predictive variables were as follows:the differential value of the shortest distance from the supernumerary tooth to the labial and palatal bone plates , which was divided into≥1. 5 mm group and < 1. 5 mm group ; the ratio of the distance from the adjacent tooth apex to the nasal floor, compared to the length of the supernumerary teeth , was recorded as≥1 and < 1 . A statistical software SPSS 20 was used to complete the statistical analysis.
Results :
When the differential value was less than 1. 5 mm , the possibility of unplanned surgery increased , and the probability of planned surgery was 0. 085 times than that of unplanned surgery. With age growing each 1 ⁃year, the probability of planned surgery gradually decreased , HR = 0. 745. The postoperative swelling of the palatal approach was only 0. 374 times than that of the labial approach. With age increasing , the operation time increased gradually , B = 1. 213. The ratio of the distance from the adjacent tooth apex to the nasal floor to the length of the supernumerary teeth did not affect the change of the surgical plan during the operation.
Conclusion
The shortest distance difference between the supernumerary teeth and the labial and palatal bone plates can be used as a reference for the selection of surgical approach for the extraction of maxillary impacted mesiodens.
3.Analysis of donation related vasovagal reactions in Hefei

【Objective】 To explore the influencing factors and characteristics of blood donation-related vasovagal reactions in whole blood donation in Hefei city, and to put forward specific prevention measures. 【Methods】 Whole blood donors with vasovagal reactions related to blood donation in Hefei area from January 2021 to December 2021 were selected as the study subjects, and the related factors such as gender, age, blood donation month, initial or multiple blood donation, blood donation volume, blood donation location, and organization type of blood donation were retrospectively analyzed. 【Results】 The incidence of blood donation-related vasovagal reactions was 10.46‰ in the whole blood donation in Hefei in 2021, among which 71.89% occurred during blood collection. There were statistically significant differences in blood donation-related vasovagal reactions by blood donation age, blood donation history, blood collection month, organization type of blood donation and blood donation volume(P<0.05), but not by gender and location of blood collection(P>0.05). After balancing the age factors, there was no statistically significant difference in the incidence of DRVR between donors aged 18~22 and 23~29 years by gender(P>0.05), while there were significant differences in blood donors aged 30~39, 40~49, and 50~60 years by gender(P<0.05). Among blood donors of different ages, there were statistically different incidences of DRVR among first-time and repeated donors (P<0.05). 【Conclusion】 In the process of whole blood donation, attention should be paid to blood donors first-time, young (18~30 years old) and student, which can help prevent the occurrence of blood donation-related vasovagal reactions and better retain blood donors.
4. The value of the prominent hypointense vessels sign in acute ischemic stroke

Susceptibility-weighted imaging (SWI) is a tool that uses the intrinsic nature of local magnetic fields to enhance image contrast in order to improve the visibility of various susceptibility sources. SWI has blood oxygen levels dependent effect and is sensitive to the change of the cerebral oxygen saturation. This imaging method is applied to various diseases with abnormal deoxyhemoglobin concentration, such as ischemic stroke and cerebral arteriovenous malformation. Patients with acute ischemic stroke have elevated levels of deoxygenated hemoglobin in the affected area, so the ischemic area can show abnormal venous imaging on SWI images. SWI could recognize penumbra and guide the management of patients with acute stroke. Besides, SWI also could evaluate the severity of symptoms, predict prognosis and future surviving state. This paper reviews the research progress of the prominent hypointense vessels sign and its application in acute ischemic stroke.
5.Three⁃dimensional cephalometric measurement of craniofacial hard tissues with normal occlusion in Anhui area

Objective:
To establish a three⁃dimensional cephalometric standardized value database for Anhui province , to study the craniofacial characteristics of normal adult population in Anhui , China and to provide effective
information for making orthodontic and orthognathic surgery treatment plans of the craniofacial deformity.
Methods:
160 normal occlusal volunteers were selected according to the inclusive and exclusive criteria and were photographed by Cone Beam CT (CBCT) . The coordinate system was established for the 3D model reconstructed with Dolphin based on CBCT data , and 38 hard tissue landmarks were located. 40 lines and 20 angles were defined and measured , and measurements were analyzed with SPSS 17. 0 , to evaluate craniofacial features of the overall , midface , maxilla , mandible , alveolar bone , and dentition of the standard population.
Results:
The 3D cephalometric measurements of craniofacial hard tissues of normal adults in Anhui were obtained. The symmetry of craniofacial hard tissues was good both in males and females with the maximum asymmetry ratio of 2. 2% and the minimum of 0. 01% . The gender comparisons showed that most of lines measured of males were larger than those of females (P< 0. 01) , while there was no significant difference in the measurements of the midline deviation of upper and lower mandible , the relative deviation of the maxillary and mandible and the protrusion of upper and lower incisor. Additionally , there was no statistical significance in angles measured between genders , except for the occlusal plane angle. The results also showed that Chinese had unique facial features based on the measured data , compared with other ethnic populations.
Conclusion
The normal occlusal and maxillofacial hard tissue of Anhui Han has good symmetry. The gender obviously influences line measurements , with generally the measurements of males larger than females , but it has no effect on most angle measurements.
6.Influence of obesity and gender on the dynamic and static balance in children aged 8-10 years

Objective:
To explore characteristics of dynamic and static balance of children aged 8 to 10 years, and to provide a reference for prevention of injuries caused by physical activities among obese children and the choice of facilities for physical activities.
Methods:
Totally 100 obese children and 100 normal children were selected as the subjects by one legged jumps from 5 primary schools in economic and technological development district of Hefei, the proportion of male and female children was 1∶1 in each group. IIM-BAL-100 balance tester was used to assess the static balance ability under double feet standing with eyes closed and right foot standing with eyes opened. The dynamic balance of double feet standing with eyes opened was measured by the Balance check dynamic balance tester. Two factor analysis of variance (ANOVA) was used to examine the effect of obesity and gender on the dynamic and the static balance.
Results:
In the static balance ability, when standing with both eyes closed, there was no significant difference in all static balance values between groups, genders and the interaction between the two factors( F=2.33, 0.42 ,0.76, P >0.05). When standing on one foot with eyes opened, there was significant difference in the static equilibrium index between the groups and the gender( F=2.72, 3.07, P <0.05). In terms of dynamic balance ability, all the dynamic balance indexes had statistically significant differences among the groups( F=43.67, P <0.01).
Conclusion
Obesity can significantly reduce the ability of single leg static balance and dynamic balance in 8-10 year old children. Sex can significantly affect the single foot static balance ability of 8-10 year old children, but it has little effect on the dynamic balance ability of 8-10 year old children.
7.Efficacy of low-frequency neuromuscular electrical stimulation in the treatment of penile hypersensitive premature ejaculation

【Objective】 To investigate the efficacy of low-frequency neuromuscular electrical stimulation in the treatment of penile hypersensitive premature ejaculation. 【Methods】 A total of 66 patients treated during Nov.2021 and Aug.2022 were randomly divided into electrical stimulation group (n=22), local anesthesia group (n=21), and combined therapy group (n=23). The electrical stimulation group received low-frequency neuromuscular electrical stimulation, 5 times a week;the local anesthesia group used compound lidocaine cream 30 minutes before sexual intercourse;the combined therapy group received both treatments. After 3-month treatment, the latency of dorsal nerve somatosensory evoked potential (DNSEP), glans penis somatosensory evoked potential (GPSEP), intravaginal ejaculation latency time (IELT), premature ejaculation diagnostic tool score (PEDT), and spouse sexual satisfaction score were collected. 【Results】 After treatment, IELT, PEDT, spouse’s sexual life satisfaction score, DNSEP and GPSEP of the three groups were significantly improved (P<0.05). The combined therapy group outperformed the other two groups in all parameters (P<0.05). There were no statistically significant differences between the electrical stimulation group and local anesthesia group (P>0.05). 【Conclusion】 Low-frequency neuromuscular electrical stimulation is effective in the treatment of penile hypersensitive premature ejaculation, and the combination of local anesthetics is more effective, which is worthy of clinical application and promotion.
8.Construction of eukaryotic expression of mouse derived pcDNA3. 1 ⁃3 × Flag⁃c⁃NUP85 plasmid and its partial function research

Objective:
To construct a mouse derived pcDNA3. 1 ⁃3 × Flag⁃c⁃NUP85 expression plasmid and observe its effect on expression of inflammation factors in LPS⁃induced RAW264. 7 cells , as well as on the proliferation and apoptosis of RAW264. 7 cells.
Methods:
The NUP85 gene was amplified by PCR to construct pcDNA3. 1 ⁃3 × Flag-c⁃NUP85 eukaryotic expression plasmid. The pcDNA3. 1 ⁃3 × Flag⁃c vector was divided with enzymes. The purified PCR product was ligated with the vector, and the ligated product was transformed into bacterial competent cells. After identification by enzyme digestion , sequencing and analysis were performed. Then , it was transfected into RAW264. 7 cells , and the blank plasmid without NUP85 gene was set as the control group. The effect on cell proliferation and apoptosis were detected by CCK⁃8 assay and flow cytometry , and the expression of inflammatory cytokines such as tumor necrosis factor⁃α (TNF⁃α ) and interleukin⁃6 (IL⁃6) in LPS⁃induced RAW264. 7 cells was detected by Western blot and ELISA.
Results:
Enzyme digestion identification and Western blot results showed that pcDNA3. 1 ⁃3 × Flag⁃c⁃NUP85 eukaryotic expression plasmid was successfully constructed and expressed. The results of CCK⁃8 assay showed that the cell survival rate of NUP85 overexpression group was significantly lower than that of control group after 24 h[(0. 55 ± 0. 03) vs (0. 67 ± 0. 05) , F = 30. 98 , P < 0. 05 ] . The results of flow cytometry showed that the cell apoptosis rate of NUP85 overexpression group was higher than that of control group[( 15. 78 ±1. 05)% vs ( 13. 40 ± 0. 47)% , F = 75. 38 , P < 0. 05] . The results of Western blot and ELISA showed that after transfection of pcDNA3. 1 ⁃3 × Flag⁃c⁃NUP85 , the expression of TNF⁃α and IL⁃6 in RAW264. 7 cells were higher than those in the control group ,with statistical significance (P < 0. 05) .
Conclusion
NUP85 can inhibit the proliferation and promote apoptosis in LPS⁃stimulated RAW264. 7 cells , and NUP85 can promote the expression of inflammatory cytokines IL⁃6 and TNF⁃α in LPS⁃stimulated RAW264. 7 cells.
9.Characteristic comparison of mouse primary macrophages cultured in L929 cell conditioned medium.

The purpose of this study is to provide a culture for mouse bone marrow-derived macrophages (BMDM) and peritoneal macrophages (PM) and to characterize their molecular and cellular biology. The cell number and purity from the primary culture were assessed by cell counter and flow cytometry, respectively. Morphological features were evaluated by inverted microscope. Phagocytosis by macrophages was detected by the neutral red dye uptake assay. Phenotypic markers were analyzed by real-time fluorescent quantitative PCR. Our results show that the cell number was much higher from culture of BMDM than PM, while there was no significant difference regarding the percentage of F4/80+CD11b+ cells (98.30%±0.53% vs. 94.83%±1.42%; P>0.05). The proliferation rate of BMDM was significantly higher than PM in the presence of L929 cell conditioned medium, by using CCK-8 assay. However, PM appeared to adhere to the flask wall and extend earlier than BMDM. The phagocytosis capability of un-stimulated BMDM was significantly higher than PM, as well as lipopolysaccharide (LPS)-stimulated BMDM, except the BMDM stimulated by low dose LPS (0.1 μg/mL). Furthermore, Tnfα expression was significantly higher in un-stimulated BMDM than PM, while Arg1 and Ym1 mRNA expression were significantly lower than PM. The expression difference was persistent if stimulated by LPS+IFN-γ or IL-4. Our data indicate that bone marrow can get larger amounts of macrophages than peritoneal cavity. However, it should be aware that the molecular and cellular characteristics were different between these two culture systems.

10.Mechanism of Danzhi Jiangtang Capsules on improving liver injury in hyperlipidemia rats based on MAPK pathway.

This study aimed to investigate the protective effect and preliminary mechanism of Danzhi Jiangtang Capsules( DJC) on liver of hyperlipidemic rats. The hyperlipidemia models were successfully made by high-fat diet for 12 weeks in male SD rats,and then divided into model control group and DJC treatment groups( 500 and 1 000 mg·kg~(-1)·d-1) via gavage administration for additional 8 weeks.The levels of serum lipid and liver metabolism indices were detected; HE and oil red O staining were used to observe the pathological changes of liver. Expression levels of extracellular regulated protein kinase 1/2( ERK1/2),c-Jun N-terminal kinase( JNK),and p38 mitogen-activated protein kinase( p38 MAPK) were detected by real-time polymerase chain reaction( RT-PCR). Expression of MCP-1,phosphorylated ERK( p-ERK),phosphorylated JNK( p-JNK),and phosphorylated p38 MAPK( p-p38) were analyzed by immunohistochemistry and Western blot. The results showed that DJC decreased body weight and serum levels of total cholesterol( TC),triglyceride( TG),alanine aminotransferase( ALT),aspartate aminotransferase( AST),increased serum high-density lipoprotein cholesterol( HDL-C) level,ameliorate injury and lipid deposition in the liver induced by the high-fat diet,decreased mRNA expression of ERK1/2,JNK and p-38 MAPK as well as protein expression of p-ERK,p-JNK,p-p38,and MCP-1,somewhat showing a dose-dependent effect. Therefore,DJC has an obvious protective effect on liver of hyperlipidemic rats with certain dose-dependent effect,and the mechanism may be related with inhibiting MAPK pathways and inflammation.
