1.Protective effect of tanshinones against ischemia injury in cultured primary cortex neurons

AIM To investigate protective effects and mechanism of tanshinones on ischemia-like injury models. METHODS Six ischemia models including hypoxia, hypoglucose, oxidant injury, caffeine injury, nitric oxide neurotoxicity and excitatory amino acid injury were used to assay the anti-ischemic roles of tanshinones in cultured primary cortex neurons. The changes of injuried cortex neurons were observed by the way of morphological examination, and live neurons of crystal violet staining were measured according to absorbent index. RESULTS It was found that tanshinones possessed obvious protective effects on primary neurons in injury models by the way of morphological examination. Crystal violet staining also indicated that tanshinones increased number of live neurons in injury models significantly. The protective effects of tanshinones on models of oxidant injury, caffeine injury and NMDA injury were superior to other injury models. CONCLUSIONS 83.0 μmol*L-1 tanshinones protected rat cortex cells from all injury models effectively in vitro.

Aim To investigate protective effects and mechanism of TPG on ischemia_like injury models. Methods Six ischemia models including hypoxia, hypoglucose, oxidant injury, calcium overload, nitric oxide neurotoxicity and excitatory amino acid injury were used to assay the anti_ischemic roles of TPG in cultured primary cortex neurons. Results It was found that TPG possessed obvious protective effects on primary neurons in injury models by the way of morphological examination. Crystal violet staining also indicated that TPG increased number of life neurons in injury models significantly. Couclusions 50~200 ?g?ml-1 TPG protected rat cortex cells from all injury models effectively in vitro.
4.Alteration of ion channel currents in ventricular myocytes of the rabbit 24 hours after acute myocardial infarction

BACKGROUND:After acute myocardial infarction(AMI),there is still surviving myocardium in and around the infarcted area,which plays an important role in the occurrence of arrhythmia. OBJECTIVE:To study the alterations of the activities of Na+ channel current(INa),L-calcium current(ICa-L),transient outward K+ current(Ito) and inward rectifying K+ current(IK1) in the cardiomyocytes in the infarcted area after AMI. DESIGN: A randomized controlled study. SETTING:Department of Cardiology,Bethune International Peace Hospital. PARTICIPANTS:The experiment was finished in the Central Laboratory of the Department of Cardiology,Bethune International Peace Hospital from January to June 2003.Twenty New Zealand pure big-ear rabbits were randomly divided into AMI group(n=10) and control group(n=10). INTERVENTIONS:Rabbit AMI models were established by ligation of the left anterior descending coronary artery.The ventricular myocytes were separated with the method of enzymatic dissociation technique,and the changes of the ion currents were recorded with the whole cell patch-clamp techniques. MAIN OUTCOME MEASURES:The changes of INa,ICa-L,Ito and IK1 in the cardiomyocytes taken from the infarcted area of epicardium 24 hours after AMI in both the AMI and control groups. RESULTS:Twenty-four hours after AMI,the peak current densities of INa,ICa-L and IK1 in the AMI group [(28.48± 3.53) pA/pF,n=16;(3.91± 0.95) pA/pF,n=12;(26.93 ± 3.48) pA/pF,n=16]were all significantly reduced as compared with those in the control group [(45.50± 5.33) pA/pF,n=12;(5.58± 1.53) pA/pF,n=10;(34.12± 4.21) pA/pF,n=10] (t=3.026,P< 0.01;t=2.985,P< 0.01;t=2.706,P< 0.05).There was no significant difference in the Ito density between the AMI group and control group (P >0.05). CONCLUSION:The reduce of INa,ICa-L and IK1 caused by AMI can result in the decrease of myocardial conduction velocity,the shortening of action potential-time,abnormal repolarization,which is possibly the ionic mechanism for the reentrant ventricular arrhythmia after AMI.
5.Infection situations of fungi and trichomonas and changes of cell counts in patients with non-bacterial vaginosis

Objective To explore the infection situation of fungi and trichomonas and the changes of pus cell and epithelial cell counts in patients with non-bacterial vaginosis in Lanzhou .Methods The pus and epithelial cell counts of vaginal secretion samples from patients with vaginitis were detected .Fungi and trichomoniasis were checked under high power microscope ,and the samples were defined positive if fungal spores or pseudohyphae were found .Results In the 4 404 cases of patients with vaginitis ,the total rate of fungi and trichomoniasis infection was 28 .63% (1 261/4 404) .The infection rate for fungi was 25 .39% ,and for trichomonad was 3 .25% ,respectively .The dual infection rate of fungi and trichomonad was 0 .54% .Compared with ≤20 age group ,the counts of pus cells and epithelial cells in other age groups were significantly different(P<0 .05) .Conclusion Fungi were the primary in-fection etiology of non-bacterial vaginosis in Lanzhou .
6.DETERMINATION OF HBV-DNA BY MOLECULAR HYBRIDIZATION IN SERA OF 186 CHRONIC ASYMPTOMATIC HBsAg CARRIERS

32P-HBV-DNA was used as a probe to determine HBV-DNA by molecular hybridization in the sera of 186 chronic asymptomatic HBsAg carriers (ASC). The result of HBV-DNA determination correlated closely with that of the HBeAg/anti-HBe system.The viral DNA was detected in 51(82.2%)of 62 HBeAg (+) cases but only in 1 of 103 anti-HBe ( + ) cases. The prevalence of HBV-DNA was directly proportional to the P/N ratio of RIA for HBeAg. When the P/N was above 8.1. its detection rate was 95.0%; when P/N below 5.0. it was only 12.5%. HBV-DNA determined by molecular hybridization was deemed to be a more direct proof of viral replication in ASC.
7.Transsphenoidal microsurgical treatment of male prolactinoma and sexual disfuction

Objective To explore the features of transsphenoidal microsurgical treatment of male pro- lactinoma and to evaluate the sexual function before and after transsphenoidal microsurgery.Methods A se- ries of 23 cases male prolactinoma were analysed combined with retrospective and prospective study.9 casese with sexual disfunction had finished the International Index of Erectile Function with 15-item questionnnaire (ⅡEF-15) before and after the surgery.Results 8/10 cases treated with dopaminergic agonist combined with surgery achieved complete remission,while only 4/13 cases achieved complete remission with surgery on- ly(x~2=5.490,P=0.036).Aecoding toⅡEF-15,sexual function improved after surgery in 9 cases(paired- sample) t test,P<0.05).Conclusion Transsphenoidal microsurgical treatment of prolactinoma may im- prove the male sexual functions.Dopaminergic agonist combined transsphenoidal microsurgery may achieve a better outcome.
8.Study on the validation for reference intervals of adult blood cell analysis

Objective In order to verify the validation of the industry standard WS/T 405‐2012 about adult blood cell analysis reference range ,and ensuring their clinical application .Methods According to Clinical and Laboratory Standards Institute(CLSI) C28‐A3 recommendation method ,40 health reference individuals were enrolled in the study whose blood samples were collected ,de‐tected and analyzed .Results The blood cell analysis indicators involved in the validation included WBC ,RBC ,hemoglobin(HGB) , hematocrit(HCT) ,mean corpuscular volume(MCV) ,mean corpuscular hemoglobin(MCH) ,mean corpuscular hemoglobin concen‐tration(MCHC) ,platelet(PLT ) ,etc .The probability of all results of selected 40 reference individuals beyond the WS/T 405‐2012 blood cell analysis reference interval were no more than 10% (value 90% or higher) .Conclusion The WS/T 405‐2012 blood cell a‐nalysis reference range is suitable for the laboratory .A perfective verification system should be established to ensure its application .
9.Clinical efficacy of liraglutide combined with metformin in treatment of type 2 diabetes mellitus

One hundred and twenty four patients with type 2 diabetes mellitus (T2DM) admitted to the Department of Endocrinology of Chuzhou First People's Hospital from April to October 2019 whose blood glucose levels were not controlled after treatment with metformin (2.0 g/d) for more than 3 months were enrolled in the study. The patients were randomly divided into oral drug group and liraglutide group with 62 cases in each group. On the basis of metformin, the oral drug group was treated with acarbose (300 mg/d); the liraglutide group was treated with liraglutide (1.2 mg/d). The course of treatment was 3 months. The body weight, fasting blood glucose (FPG), 2-h postprandial blood glucose (2 hPG), glycosylated hemoglobin (HbA1c), cholesterol (TC) and triglyceride (TG) levels were compared between the two groups before and after treatment, and the gastrointestinal symptoms and the incidence of hypoglycemia were analyzed. After 3-month treatment the body weight, FPG, 2 hPG, HbA1c of the two groups were lower than the baseline levels ( P<0.05). Compared with the oral group, the body weight, FPG, 2 hPG, HbA1c and TG in liraglutide group were more markedly decreased ( P<0.05). The gastrointestinal reaction of liraglutide group was significantly higher than that of the oral drug group, and there was no hypoglycemia in both groups. The data indicate that liraglutide combined with metformin can significantly improve the blood glucose, blood lipid and body weight of T2DM patients and no hypoglycemia occurs during the treatment.
10.Application of SF-36 scale on pulmonary tuberculosis patients in Yunnan province of China and southern Thailand.

OBJECTIVETo explore the effect of SF-36 scale being applied in different countries under different culture and to describe the quality of life of pulmonary tuberculosis patients in China and Thailand.
METHODSSF-36 scale was applied to pulmonary tuberculosis patients in both countries using face to face interview.
RESULTSMany coefficients among domains were greater than 0.5 when quality of life of tuberculosis patient in both countries was measured. Cronbach's coefficient of all domains were greater than 0.7 for tuberculosis patients in China while cronbach's Coefficient of most domains were equal or greater than 0.7 for tuberculosis patients in Thailand except for vitality and social domains. The score of social domain for patients in Thailand was greater than that of China.
CONCLUSIONStructure validity was not good for tuberculosis patients in both countries since there were some items overlapped in different domains. However, the reliability was good for measuring quality of life of tuberculosis patients both in China and in Thailand.
