1.Cardiac Protection during Open Heart Surgery: A View from Coronary Endothelial Function

Cardioplegic (and organ preservation) solutions were initially designed to protect the myocardium (cardiac myocytes) during cardiac surgery (and heart transplantation). Due to the differences between the cardiac myocytes and vascular (endothelial and smooth muscle) cells in structure and function, the solutions may have adverse effect on coronary vascular cells. However, such effect is often complicated by many other factors such as ischemiareperfusion injury, temperature, and perfusion pressure or duration. In evaluation of the effect of a solution on the coronary endothelial function, a number of points should be taken into consideration. First, the overall effect on endothelium should be identified. Second, the effect of the solution on the individual endothelium-derived relaxing factors (nitric oxide, prostacylin, and endothelium-derived hyperpolarizing factor) must be distinguished. Third, the effect of each major component of the solution should be investigated. Fourth, the effect of a variety of new additives in the solution may be studied. In the last decades, we have focused our research on the endothelial function during open heart surgery and have for the first time found that high potassium concentration impairs the EDHF-mediated function. This review attempts to discuss the above issues based on available literature in order to provide information for further development of cardioplegic or organ preservation solutions.
3.Effects of Gynostemma Pentaphyllum on serum markers of liver fibrosis in cholesterol-induced non-alcoholic fatty liver disease in rabbits

Objective To investigate the effects of Gynostemma Pentaphyllum on serum markers of liver fibrosis in cholesterol-induced non-alcoholic fatty liver disease (NAFLD) in rabbits. Methods Forty adult male white rabbits were randomly divided into a normal control group, a model group, a treatment group and a simvastatin group, with 10 rabbits in each group. NAFLD was induced with a high-cholesterol diet. After modeling, the rabbits in the treatment group were intragastrically administrated with Gynostemma Pentaphyllum 5 mg/(kg?d), the simvastatin group with simvastatin 5 mg/(kg?d), and the model and normal control groups with the equal volume of distilled water for 9 weeks. The serum levels of total cholesterol (TC) and triglyceride (TG), the serum inflammatory maker C-reactive protein (CRP), the serum markers of liver fibrosis such as hyaluronic acid and laminin (LN), and the TG level in the liver tissue were detected. Results Compared with the normal control group, the serum levels of TC (60.50 ± 9.77 mg/L vs.1.30 ± 0.44 mg/L), TG (1.72 ± 0.61 mmol/L vs. 0.85 ± 0.39 mmol/L), CRP (256.79 ± 30.78 mg/L vs. 8.71 ± 1.41 mg/L), HA (798.8 ± 69.4 ng/ml vs. 121.2 ± 6.8 ng/ml),LN (964.8 ± 62.8 ng/ml vs. 142.4 ± 12.2 ng/ml) in the model group were increased significantly (all P<0.01). Compared with the model group, the serum levels of TC (36.44 ± 6.57 mmol/L vs. 60.50 ± 9.77 mmol/L), TG (1.31 ± 0.39 mmol/L vs. 1.72 ± 0.61 mmol/L), CRP (68.77 ± 10.78 mg/L vs. 256.79 ± 30.78 mg/L), HA (420.8 ± 60.2 ng/ml vs. 798.8 ± 69.4 ng/ml), LN (449.8 ± 56.6 ng/ml vs. 964.8 ± 62.8 ng/ml) and the TG level in the liver tissue (0.52 ± 0.10 mmol/L vs. 0.77 ± 0.08 mmol/L) in the treatment group were decreased significantly (all P<0.01). The TG level in the liver tissue in the treatment group was significantly lower than that in the simvastatin group (0.52 ± 0.10 mmol/L vs. 0.59 ± 0.09 mmol/L;P<0.05). There were no significant difference in the serum levels of TC, TG,CRP,HA and LN between the treatment group and the simvastatin group (all P>0.05). Conclusions Gynostemma Pentaphyllum may regulate lipid metabolism, alleviate inflammation, and decrease serum markers of liver fibrosis, and might protect against liver fibrosis in rabbits with NAFLD.
4.CT findings in adrenal ganglioneuroma

Objective To find the features and diagnostic values of CT for adrenal ganglioneuroma.Methods CT examinations were performed in 16 cases(7 with plain scan and 9 with both plain and enhanced scan).All cases were confirmed by pathological evidence.Results 2 cases showed solid -cystic mass,14 were solid mass,and 5 accompanied with calcification.The average CT value was from 20 to 40 HU.In 9 cases with enhanced scan,5 slightly enhanced on delayed phase and no enhancement was seen in the others.Conclusion Adrenal ganglioneuroma showed some characteristic CT appearance and CT scanning plays an important role in diagnosis of adrenal ganglioneuroma.
5.A novel lupane triterpene from seeds of Ziziphus jujuba var.spinosa

Objective To study the chemical constituents from the seeds of Ziziphus jujuba var.spinosa,which is used as a sedative herbal medicine in China.Methods The constituents were separated and purified by silica gel column.Their structures were elucidated based on their physicochemical properties and spectral analysis.Results Six compounds were obtained from 95% EtOH extracts in the seeds of Z.jujuba var.spinosa.They were identified as 2?,3?-dihydroxy-lup-20(29)-en-28-oic acid methyl ester(Ⅰ),?-sitosterol(Ⅱ),betulin(Ⅲ),betulinic acid(Ⅳ),hexadexanoic acid 2,3-dihydroxypropyl ester(Ⅴ),and daucosterol(Ⅵ).Conclusion Compound Ⅰ is a novel lupane triterpene named as alphitolic acid methyl ester,compound Ⅴ is obtained from the seeds of Z.jujuba var.spinosa for the first time.
6.Construction of eukaryotic expression vector of pre-miR-15a and its inhibitory effect on Raji cells proliferation

Objective To construct an eukaryotic expression vector of pre-miR-15a,and to investigate the inhibitory effect of pre-miR-15a to Raji cells. Methods The pGCSIL-GFP vector encoding pre-miR-15a nucleotides was transfected into the bacterial competent cells,and then confirmed by PCR and sequencing analysis. The identified vector was transfected into Raji cells with oligofectamine 2000. The cells were divided into 3 groups,blank,negative control and pre-miR-15a group. Semi-quantitative RT-PCR was used to detect the expression of Bcl-2 mRNA,and immunofluorescence indirect for Bcl-2 protein expression. The growth of Raji cells was assayed by trypan blue dye exclusion method. Results PCR and sequences analysis indicated that the recombinant clones was identical with target sequences. Many green fluorescent cells were observed under fluorescent microscopy. The levels of Bcl-2 mRNA at every group had no obviously difference. Bcl-2 protein expression was obviously decreased at pre-miR-15a group compared with the other groups. Trypan blue dye exclusion method showed the cell growth was inhibited at 48 h and 72 h post-transfection. Conclusion We successfully construct the eukaryotic expression vector of pre-miR-15a,and it can inhibit the growth of Raji cells.

9. Optimization of the prescription of Calcein-loaded novel temperature-sensitive liposomes

OBJECTIVE: To optimize the prescription of the novel temperature-sensitive liposomes(NTSL). METHODS: Calcein-loaded temperature-sensitive liposomes with different phospholipid composition (DPPC-MSPC-DSPE-PEG200=86:10:4 and N (20% HSPC + 80% DPPC) -MSPC-DSPE-PEG200=92:4:4) were prepared by the thin film -hydration method. Based on the fluorescence self-quenching off characteristics of calcein, the release behavior of the above liposomes was compared when exposed to 37°C (normal temperature) as well as their respective phase transition temperature (Tm). RESULTS: Compared with the traditional temperature-sensitive liposomes (TTSL), the NTSL showed more favorable temperature-sensitive release performance and more stable encapsulation ability to calcein, even with the amount of lysophospholipids was reduced by 60 percent. CONCLUSION: The application of NTSL helps to enhance the effect of temperature-sensitive release.
10.Observation on the effect of psychological intervention of iron combined with erythropoietin in the treatment of anemia in hemodialysis patients

Objective And effect of psychological intervention observation and analysis of iron combined with erythropoietin in the treatment of anemia in hemodialysis patients and the effect of. Methods 110 cases of Shaoxing Hospital of Traditional Chinese Medicine in anemic hemodialysis patients with January 2015 to February 2016 underwent diagnosis and treatment as the research object of the study, all the patients were randomly divided into 2 groups, respectively for the test group and control group, 2 groups of 55 cases. The control group was treated with erythropoietin receptor activator and psychological intervention treatment, experimental group was treated with ferrous succinate combined with erythropoietin receptor activator for treatment, but also psychological nursing intervention, observation and comparison of the symptoms of anemia, 2 groups of patients after treatment of adverse reaction etc. Results The experimental group patients with red blood cells, serum ferritin, hemoglobin and hematocrit were significantly higher than control group, the difference was statistically significant (P<0.05); side effects in test group than in the control group, the difference was statistically significant (P<0.05). Conclusion Iron combined with erythropoietin treatment in anemic hemodialysis was more effective at the same time, then the implementation of the corresponding psychological intervention, can reduce the adverse reactions in patients with symptoms, improve the safety of the treatment, and has high applicationvalue.
