1.A study of fomotidine preventing weight gain olanzapine-induced

Objeetive To study the therapeutic effect of the H2 antagonist fomotidine on preventing weight gain olanzapine-induced.Methods Forty first-episode CCMD-3 schizophrenia patients were randomly allocated to received either famotidine or placebo in addition to olanzapine for 8 weeks.Their body weights and heights were measured before treatment and 2 weeks,4 weeks,8 weeks after treatment to calculate the body mass index and the percentage of patients gained more than 7% of initial bpdy weight after 8 weeks.SAPS and SANS were selected to evaluate the therapeutic effect.Results While the weight and the body mass index of patients in two groups after 2 weeks,4 weeks,8 weeks post-treatment increased significantly compared with those of pre-treatment(P<0.05).The weight of patients in two groups gained 3.7kg at the edd of 8 weeks,and the BMI increased 2.5 or 2.6 kg/cm2,reapectively.However 8 weeks later,the difference between two groups wan not significant(P>0.05).The difference between two groups on the percentage of patients gained more than 7% of initial body weight at the end of 8 weeks Wan not significant(P>0.05).The sCOres of SAPS and SANS at the end of 8 weeks decreased significantly compared with those of the baseline(P<0.05),but the difference between two groups was not significant(P>0.05).Conclusion Allocated to the H2 antagonist fomotidine could not reduce weight gain olanzapine-induced.
2.Effect of atorvastatin on myocardial fibrosis induced by aldosterone

AIM: To investigate the effects and mechanisms of atorvastatin on myocardial fibrosis induced by aldosterone in SD rats. METHODS: Forty male uninephrectomized SD rats were limited to drink 1% NaCl water for 4 weeks and assigned to the follow groups: vehicle control rats (CON group);aldosterone treated rats (ALD group);spironolactone + aldosterone treated rats(SPI+ALD group);atorvastatin + aldosterone treated rats (ATO+ALD group). Blood pressure was measured by catheterization. Expression of platelet-derived growth factor (PDGF-A, PDGF-B), platelet-derived growth factor receptor (PDGFR-α, PDGFR-β) and ectodermal dysplasia-1 (ED-1) were analyzed by immunohistochemistry. Collagen volume fraction (CVF) and perivascular collagen area (PVCA) were analyzed by Sirius-Red staining. Myocardium osteopontin protein was detected by Western blotting analysis. RESULTS: Mean arterial blood pressure in ALD group, SPI+ALD group and ATO+ALD group was elevated, and significant difference was observed between the three groups and vehicle control group (P<0.01 or P<0.05). Myocardial fibrosis was observed in ALD group. Compared to other three groups, the index of CVF and PVCA was increased significantly (P<0.01 or P<0.05). No significant difference of the index of CVF and PVCA between ATO+ALD group and SPI+ALD group was observed (P>0.05). Compared to other groups, the levels of PDGF-A, PDGF-B, PDGFR-α, ED-1 and OPN in ALD group were significantly increased (P<0.01 or P<0.05). The levels of PDGF-A, PDGF-B, PDGFR-α and OPN were no significant difference between ATO+ALD group and SPI+ALD group (P<0.01 or P<0.05). However, the level of ED-1 in ATO+ALD group was significantly decreased compared to SPI+ALD group (P<0.05). No significant difference of PDGFR-β level among four groups was found (P>0.05). CONCLUSION: These results suggest that atorvastatin may attenuate myocardial fibrosis induced by aldosterone. The mechanisms concern with reduction of macrophage infiltration, expression of inflammatory cytokines OPN, partially inhibition of platelet-derived growth factor and its receptor expression.
3.Influence of diabetic self-management education on blood glucose control and inflammatory factor levels in inpatients with diabetes mellitus

Objective:To explore the influence of diabetic self-management education on blood glucose control and inflammatory factor levels in inpatients with diabetes mellitus (DM).Methods:A total of 168 DM patients,who hospitalized in department of endocrinology in 163rd hospital of Chinese PLA from Jun 2012 to Jun 2013,were ran-domly divided into self-management education group (self-management group,n=88)and routine health education group (routine management group,n = 80).Self-management group received self-management education course namely AADE7TM ,provided by full-time DM health education staff,including healthy eating,being active,monito-ring,taking medication,problem solving,healthy coping and reducing risks,while routine management group re-ceived routine DM health education.Blood glucose control,changes of levels of myeloperoxidase (MPO),tumor necrosis factor-α(TNF-α)and intercellular adhesion molecule-1 (ICAM-1)were examined before and after interven-tion.Results:Compared with before intervention,there were significant reductions in levels of fasting blood glucose (FBG),postprandial blood glucose (PBG)and glycosylated hemoglobin (HbA1c)in both groups after interventions (P <0.05 all).Compared with routine management group,there were significant reductions in levels of FBG [(7.1 ±1.3)mmol/L vs.(6.0±1.1)mmol/L],PBG [(9.5±1.6)mmol/L vs.(8.8±1.4)mmol/L]and HbA1c [(7.9 ±1.5)% vs.(7.2±1.1)%]in self-management group,P <0.05 all.Before discharge,there were significant de-crease in inflammatory factor levels in both groups compared with before intervention (P < 0.05 all).Compared with routine management group after intervention,there were significant decrease in inflammatory factor levels [TNF-α:(201.0±57.3)pg/ml vs.(100.7±49.2)pg/ml,ICAM-1:(25.2±3.6)ng/ml vs.(18.2±2.9)ng/ml] in self-management group,P < 0.05 both.Conclusion:Self-management education provided by full-time diabetic health education staff is superior to routine diabetic health education method in controlling blood glucose and decrea-sing inflammatory factor levels,indicating it can improveβcell function and relieve insulin resistance.
4.Design and implementation of academic impact power information platform for secondary disciplines of clinical medicine

Objective To design and implement the academic impact power information platform for the secondary disciplines of clinical medicine in order to keep abreast of their advantages and disadvantages. Methods An index system for the assessment of secondary disciplines of clinical medicine was constructed with the bibliometric methods according to the factual data. Results Its functions of query analysis, batch input, and editing of the index data of different disciplines were achieved in higher education institutions using the ASP. NET technology based on the data processing, indexing and statistical analysis. Conclusion The platform can provide the data support for the develop-ment and scientific research management of secondary disciplines, and thus playing an important role in promoting the innovative research and management of clinical medicine.
5.Prevention of the adverse reaction application of bevacizumab in combination with chemotherapy in postoperative patients with gastric cancer

Objective To investigate the adverse reaction of gastric cancer patients after inquiry application of bevacizumab combined with chemotherapy,and put forward some corresponding preventive measures.Methods Sixty-five cases patients with gastric cancer who were treated with advanced gastric cancer radical operation application of bevacizumab in combination with chemotherapy in the Central Hospital of Jiangmen from January 2012 to January 2014,including 41 male,24 female,with the average age of 48.7 years old.The adverse reactions were observed,and give the corresponding prevention and control measures on the basis of the adverse reaction conditions,the experience in the prevention and treatment were summarized according to the therapeutic effect with clinical and related literature.Results The adverse reactions caused by bevacizumab combined with chemotherapy in patients with gastric cancer including proteinuria (a total of 24 cases(37%),including 18 cases(27.8%) of (+),6 cases(9.2%) of (+ +)),gastrointestinal reaction(a total of 10 cases (15.4%),including 8 cases (12.3 %) of grade Ⅰ,2 cases (3.1%) of grade Ⅱ),thrombosis (a total of 20 cases(30.7%),including 13 cases of grade Ⅰ (20%),7 cases(10.7%) of grade Ⅱ) and hypertension(a total of 11 cases(16.9%),including 9 cases(13.8%) of grade Ⅰ,2 cases(3.1%) of grade Ⅱ).Bevacizumab combined with chemotherapy in the treatment of gastric cancer patients with partial remission in 41 cases (63.1%),10 cases (15.4%) of stable disease progression,14 cases (21.5%) of disease progression,the complete remission rate was 63.1%.For adverse reactions of proteinuria,patients with asymptomatic proteinuria,if patients with urinary protein detection results for (+),without special treatment,when patients with urinary protein detection for (+ +),given the observation,if patients with urinary protein was more than or equal to 2 g/24 h,paused bevacizumab until proteinuria<2 g.When the patients with digestive system damage,such as ALT and AST elevation,given the treatment of liver until the two hepatic enzymes returned to normal;patients with gastrointestinal reactions were nausea and vomiting,asked patient to light diet and regular esophagoscopy,if the patients appeared visceral fistula immediately.stopped bevacizumab.For patients with thrombosis,should be closely monitored in patients with signs,once found regardless of any level of embolism events,all immediately stopped the chemotherapy drugs.For the main adverse reactions of hypertension,given patients with proper medication explain before the treatment to reduce the fear,tension,and avoiding mood disorders which lead to an increase in blood pressure,blood pressure should be regularly detected in patients who use drugs,if patients occurred a sharp rise in blood pressure during the treatment,immediately stopped bevacizumab and given antihypertensive treatment.Conclusion Clinicians should take the possible corresponding prediction in patients underwent application of bevacizumab in combination with chemotherapy on the possible different adverse reactions according to the actual situation of patients,and take corresponding preventive measures,can to some extent avoid serious adverse reaction occurred,and improve the quality of life of the patients.
6.Application of Interface Adsorption Theory for Microencapsulation Technology of Borneol

In this paper, the theory of electrical character and adsorption of solidliquid interface was applied to study the preparation process for borneol-mi-croencapsule with co-coacervation using gelatin and arabic gum as materials. The optimum process conditions for preparing microencapsule was gained. This study method provided a reliable basis for raising the inclusion rate, decreasing the capsule materials in quantity and drawing up the process conditions.
8.Construction and expression of recombinant adenovirus carrying human endostatin gene

AIM:So far,endostatin is found playing the strongest effect to inhibit endothelial cells.Because endostatin protein is instable,it is difficult to prepare and apply.Gene therapy is used to assist endostatin to inhibit angiogenesis.This study constructed recombinant adenovirus of human endostatin gene using AdEasy-1 system to investigate the feasibility of expressing endostatin protein in ECV304 cells(human umbilical vein endothelial cells).METHODS:The experiment was performed at College of Life Science,Sun Yat-set University from March to October 2006.Endostatin was amplified from Pshuttle-Endostatin plasmid with PCR and subcloned into the pAdTrack-CMV shuttle vector after sequencing and identification.The resultant plasmid was co-transduced into E.coli BJ 5183 cells with pAdEasy-1 plasmid for homologous recombination.The recombinant plasmid was detected by restriction analysis and DNA sequence analysis.The linearized recombinant plasmid was transfected into AAV 293 cells with LipofectAMINE2000.The recombinant adenovirus was detected by GFP,PCR and restriction analysis.The virus was used to infect ECV304,and the expression of endostatin in vitro was detected by the techniques of RT-PCR and Western blot.RESULTS:The positive clones of the recombinants were constructed and confirmed by restriction endonuclease analysis and DNA sequence analysis.AAV293 cells were transfected.The titer of viral liquid was 2.06?1010 pfu/mL.The transcription of endostatin mRNA in ECV304 cells was checked by RT-PCR and endostatin protein could be detected in the culture by Western blot analysis.CONCLUSION:The recombinant adenoviral vector pAd-Endo carrying human endostatin is successfully constructed,and effective expression is found in ECV304 cells in vitro.
9.Progression on the medical nutrition therapy of type 2 diabetes mellitus

Medical nutrition therapy (MNT)is the basic and long-term treatment of diabetes patients. At present,the dietary structure of type 2 diabetes mellitus(T2DM)patients in China has yet to be further improved. This article focused on the effect,dietary structure,food selection and guarantee of compliance of MNT in T2DM.We recommended that the dietary pattern should be designed based on the traditional dietary habits around the country,combined with people living and working reality. The rational utilization of network platform can improve the efficiency and compliance of nutrition education.
10.Gene expression differences between murine intestine intraepithelial lymphocytes and splenic T lymphocytes

Objective: To compare the differences in gene expression between murine intestine intraepithelial lymphocytes (iIELs) and splenic T lymphocytes, establishing the cDNA subtractive library of iIELs, analyzing the iIELs special genes. Methods: The iIELs and splenic T lymphocytes were isolated by density gradient in Percoll and nylon column respectively. cDNA subtractive library of iIELs was established by improved subtractive hybridization. The cDNA was ligated with a different adaptor. After 2 rounds of hybridization and suppression PCR amplification, the PCR products were directly inserted into the plasmid vectors. The ESTs special for iIELs were screened by reverse Northern blot. Results: Fifty ESTs with obvious difference in the cDNA subtractive library of iIELs were screened. These ESTs were squenced and submitted to Genebank. The ESTs were analyzed through BLAST, and 14 genes special for iIELs were obtained. These genes were associated with the pattern recognition of pathogen, cytokine signal transduction and proteinase inhibitors. Conclusion: iIELs are important components of intestine natural immunity, it can recognize the conservative antigens of bacteria and can be affected by cytokines and proteinase inhibitors, leading to unique cellular behavior--more resting and less activation.
