3.Study on the Antigens of Tumor FBL-3:I.Prediction of FBL-3 Antigen and Verification of Antigenicity of Synthetic Peptides
Chinese Journal of Cancer Biotherapy 1996;0(04):-
T lymphocytes play a very important role in tumor immunity, which recognize tumor antigenic peptide presented by MHC molecules on the surface of tumor cells through T cell receptor ( TCR) . Cytotoxic T lymphocytes kill tumor cells after recognizing antigenic peptide in the cleft of MHC class I molecules. It is now generally accepted that peptides naturally presented by a given MHC class I molecule have a specific motif which is referred to as MHC class I allele-specific consensus motif and that synthetic peptide corresponding to the identified naturally presented antigens exhibit remarkable activity in inducing T-cell responses. FBL-3 is a transplantable Friend virus-induced leukemia of B6 (H-2b) origin, which can induce the CTL against FBL-3 by immunizing B6 mice with atenuated FBL-3.Seven peptides were predicted as candidates of FBL-3 antigenic peptides in the light of H-2Db specific peptide binding motif and gag gene sequence of Friend virus. The synthetic peptides, named gagl to gag7, were obtainted. The results showed that CTL specific to gag3 could be induced by in vivo immunizing B6 mice with gag3 in IFA, but a primary T cell response could not be induced by in vitro immuniztion with the peptides. gag3 and gag5 could be recognized by specific CTL to FBL-3,because the CTLs obviously killed target cells (EL-4) pulsed with gag3 or gag5. Immunization with gag3 and gag5 could not induce an immune response to protect the subsequent challenge of FBL-3 in B6 mice which could be induced by immunizing B6 mice with parental FBL-3 tumor cells. This indicates these 7 synthetic peptides may not be the same antigens of the FBL-3.
4.Study on the Antigens of Tumor FBL-3: I . Analysis of Peptide Antigens from Peptides Pool of Tumor Cells by Acid Elution
Chinese Journal of Cancer Biotherapy 1995;0(02):-
T lymphocytes, which play a very important role in tumor immunity, recognize the tumor antigenic peptide presented by MHC molecules on the surface of tumor cells. FBL-3 is a transplantable Friend virus-induced leukemia of B6 (H-2~(b) ) origin, which can induce the CTL against FBL-3 by immunizing B6 mice with attenuated FBL-3. The present paper was to elute immunogenic peptides ( CD8+ T cell epitopes) from class I complexes at the cell surface of viable FBL-3 cells by acid treatment. The acid treated cells remained viable but lost sensitivity to be lysed by FBL-3 specific cytotoxic T lymphocytes (CTLs). The sensitivity of the acid-treated cell was restored to the FBL-3 specific CTLs by supplement of the acid-eluted cell-free supematants. Paptides were subsequently fractionated by reverse-phase high performance liquid chromatography (RP-HPLC) . The fractionated peptides in HPLC fractions 5~6 and 23 ~ 60 (tubes) showed the capacity to sensitize RAM-S cells, which could be lysised by FBL-3 specific CTLs. The experiment indicates that this method may be useful in the definition of petide epitopes relevant to tumor cells.
5.One case of pediatric obstructive sleep apnea hypopnea syndrome with pulmonary hypertension.
Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2015;29(7):664-665
The clinical manifestation included snoring and mouth breathing for 2 years, repeated coughing and shortness of breath in action for more than 1 year. Physical examination of oral cavity showed tonsils were in grade III. The endoscopy showed 2/3 of postnaris were blocked by the adenoids. The preoperative ultrasonic cardiogram revealed the right atrial and right ventricular dilatation, pulmonary artery widened. The preoperative polysomnography (PSG) showed apnea-hypopnea index (AHI) was 28.5 events an hour, and the lowest oxygen saturation (LSaO2) was 39%. The patient was diagnosed as severe obstructive sleep apnea hypopnea syndrome with pulmonary hypertension. The postoperative PSG showed the AHI was 11.7 events an hour, and the LSaO2 was 86%. The ultrasonic cardiogram at 5 months after surgery didn't show any abnormalities.

6.Research progress on induction of bone marrow mesenchymal stem cells into muscle cells
International Journal of Biomedical Engineering 2006;0(01):-
Mesenchymal stem cells belong to multipotential stem cells, which is easy to isolate and culture. Many researchers have been exploring various ways to induce bone marrow mesenchymal stem cells into muscle cells. MSCs were induced into muscle cells by using biochemical and biomechanics approaches. These muscle cells could be used for such clinical applications, as treatment for ischemic cardiomyopathy and post-traumatic repair for muscular tissue. In this article we reviewed the research progress on induction of bone marrow mesenchymal stem cells into muscle cells and isolation, purification, and identification of MSC.

8.Dynamic changes of calcium ions in retinal ganglion cells exposed to hypoxia and their relationship with TNF-? and lipid peroxidation
Journal of Third Military Medical University 2003;0(22):-
Objective To investigate the changes of retinal ganglion cells (RGCs) exposed to hypoxia and the mechanism. Methods RGCs were isolated from the retina of neonatal Long Evans rats aged 1 day and cultured, then divided into normal control group and hypoxia group (cultured in incubator containing 1 O_2, 5 CO_2 and 94 N_2). At 1, 3, 12 and 24 h after incubation, the calcium ion level by laser scanning confocal fluorescence microimaging system, the activity of SOD and the content of MDA by biochemistry technology, TNF-? by ELISA were detected. Results No changes of calcium ion level in RGCs were observed in normal control group. The calcium ion level increased significantly in the hypoxia group (P
9.Clinical analysis of phacoemulsification on patients with cardiovascular disease
Journal of Third Military Medical University 1984;0(01):-
Objective To investigate the changes of systolic pressure, diastolic pressure, oculocardiac reflex(OCR) and vision of patients suffering from cardiovascular disease when they applied phacoemulsification. Methods Totally 168 cases(182 eyes) who had underwent phacoemulsification were analyzed in random selection.Of all the cases,97(106 eyes) were combined with cardiovascular disease, the others were without any general disease. The changes of blood pressure,OCR,ECG and vision were observed in two groups. Results Compared with preoperative period, the mean systolic blood pressure increased significantly during operation, but the differences of diastolic pressure,OCR,ECG and vision were not significant between the two groups. Conclusion The safety and reliability of the surgery depend on reasonable medication, careful surgical procedure and monitoring for the patients with cardiovascular disease in the peri-operation period.
10.Effect of different concentrations of fetal bovine serum on bone marrow mesenchymal stem cell purity and cycle
Acta Universitatis Medicinalis Anhui 2014;(6):742-747
Objective To compare the influence of three kinds of complete media with 0. 10 ,0. 15 ,0. 20 fetal bo-vine serum( FBS) on purity and cycle of rat bone marrow mesenchymal stem cells ( BMSCs) cultured in vitro and to seek suitable FBS concentration for the cultivation of the stem cells. Methods SD rats were executed by cervical dislocation method and used whole bone marrow adherence to isolate rat BMSCs. Experiment was divided into A, B,C,3 groups. Compared the expression of CD45, CD29, CD90, CD44 in the three groups in 2,3,4,5 passages ( P2 , P3 , P4 , P5 );the cells of P3 in group A were digested and cultured in three different concentration of com-plete culture media for four days, measuring cell cycle in 24,48,72,96 h by flow cytometry instrument. BMSCs of P3 were collected and inoculated to 6 pieces of 96-well plates, then vaccinated with complete media with 0. 10, 0. 15,0. 20 FBS in every plate, one culture plate was taken out for optical density(OD) meaturement every day with CCK-8. Results CD45 was negative, CD29, CD90, CD44 were positive. The difference of BMSCs surface markers cultured in the three kinds of complete media was bigger in the first two passages, but the difference was less in P3 , P4 , all could obtain pure BMSCs in P4 relatively;according to the results of the cell cycle at the same time, G0/G1 phase:with the increase of concentration of fetal bovine, G0/G1 phase reduced and had no diference in A, B, C groups;G2/M phase:there was difference between them after 24 h(P<0. 05,F=12. 412), but with the extension of time, the differences disappeared;S phase:there was no difference in the three groups;S+G2/M phase increased with the concentration of FBS. According to the result of cell vitality, the OD of ABC three groups increased in turn in 24 h, and there was difference(P<0. 05,F=5. 002), but with the extension of time, there was no obvious difference between them. Conclusion The three kinds of culture media in P4 can obtain pure BM-SCs, cell cycle and vitality show three complete media can promote the growth of BMSCs. There is no difference between them. Culture media with 0. 10 FBS can satisfy the isolation and amplification of BMSCs, in order to ob-tain pure BMSCs in the short term, using culture media with 0. 15 and 0. 10 FBS in the primary culture and subcul-ture respectively.
