Objective To investigate the expression of nuclear factor-κB in alveolar macrophages of paraquat-induced rats and the effect of diallyl sulfide on it.Methods Forty five male wistar rats were randomly divided into three groups,namely control group,model group,and DAS treatment group (n =15 in each).The model of paraquat poisoning was reproduced by single does of 70 mg/kg given by intra-gastric administration,while the equal volume of normal saline (NS) was given to the rats in control group instead.The dose of 100 mg/kg of DAS was given to rats by intra-peritoneal injection in DAS treatment group.The equal volume of NS was given to the rats by intra-peritoneal injection in model group and control group instead.The rats of model group and DSA treatment group were exposed to paraquat once a day for 14 days.Five rats in each group were sacrificed at 3,7,14 days.Alveolar maerophages were harvested by bronchalveolar lavage (BAL).The protein content of BAL fluid were examined.The exprossion of NF-κB was measured with immunocytochemistry technique.Results Alveolar macrophage cultures were carried out by using differential adherence of isolated and purified alveolar macrophages,and after 30 minutes culture,more adherent macrophages can be seen.Compared with model group,the protein content of BAL fluid at dfferent intervals in the control group were obviously lower,especially on the 3 rd day (261.6 ± 17.16) μg/mL vs.(673.4 ± 151.9) μg/mL;7 d (265.6 ± 18.37) μg/mL vs.(581.3 ± 134.58) μg/mL;14 d (253.8 ± 11.43) μg/mL vs.(589.07 ± 33.85) μg/mL,P ＜ 0.05.Comparisons of protein content in BLA fluid between PQ group and DAS treatment group were on the 3 rd day (673.4 ± 151.9) μg/mL vs.(342.9 ±39.03) μg/mL;on the 7 th day (581.3 ± 134.58) μg/mL vs.(383.7 ±7.37) μg/mL,P＜0.05;on the 14 th day (589.07±33.85) μg/mL vs.(282.9±15.59) μg/mL,P＜0.05.The immunocytochemistry analysis revealed minimal NF-κBp65 expression in the cell cytoplasm in the control group,while high NF-κBp65 expression was found in nuclear in the model group.Minimal NF-κBp65 expression was found in the cell cytoplasm in the DAS treatment group,and integral A value was significantly lower in the DAS treatment groups than that in the model group (P ＜ 0.05).Conclusions Treatment with an intra-peritoneal injection of DAS is capable of attenuating the extent of PQ-induced ALI in rats by lowering BLA fluid protein content,inhibiting the expression of NF-κB in alveolar maerophages.

Objective To investigate the relationship between the plasma inflammatory cytokines and carotid atherosclerosis in patients with ischemic cerebrovascular disease(ICVD).Methods The levels of plasma interleukin (IL)-6,matrix metalloproteinase (MMP)-8 and soluble CD40 ligand (sCD40L) in 64 patients with ICVD were detected by enzyme-linked immunosorbent assay (ELISA),the severity of bilateral carotid atherosclerosis was measured by colour ultrasound.The results were compared with those of non ICVD controls. The association of the levels of plasma IL-6,MMP-8,sCD40L and the severity of carotid atherosclerosis were analysized. OBJECTIVETo observe the expression of peripheral blood CD4+ T lymphocyte apoptosis gene in rheumatoid arthritis (RA) patients with cold dampness type (CDT), and to explore its correlation with clinical indicators of RA.

METHODSSixteen RA patients with CDT (as the RA group) and 16 healthy subjects (as the normal control group) were recruited. CD4 T cell apoptosis rate was detected in the RA group and the normal control group using FCM. mRNA expressions of fas, fasL, caspase-3, caspase-8, bcl-2, and bax were detected using RT-PCR. Correlations between the expression of apoptosis gene and clinical activity indicators of RA (ESR, CRP, RF, CCP, integrals for Chinese medial symptoms, morning stiffness time, joint tenderness number, joint swelling number, DAS28-3) were analyzed.

RESULTSThe apoptosis rate of CD4+ T was significantly lower in the RA group than in the control group [(2. 6 +/- 0.9) % vs. (7.7 +/- 1.3) %, P < 0.01]. mRNA expression levels of fas, fasL, caspase-8, caspase-3, and bax mRNA of CD4+ T significantly decreased, but bcl-2 mRNA expression increased in the RA group (P < 0.01). The apoptosis rate of CD4+ T was negatively correlated with ESR (P < 0.05). The mRNA expression of caspase-8 was negatively correlated with joint swelling number (P < 0.05). The mRNA expression of bcl-2 was negatively correlated with integrals for Chinese medial-symptoms and joint function classification (P < 0.01, P < 0.05).

CONCLUSIONApoptosis obstacle exists in peripheral blood CD4 +T lymphocyte of RA patients, and is closely related to disease activity.

Objective To investigate whether 5, 7-dihydrox-8-nitrochrysin (NOC) induces apoptosis in U937 monocytic leukae-mia cells is involved in the regulation of the activity of PKD 2.Methods U937 cell line cells were cultured in vitro .Apoptosis rate was analyzed by flow cytometry (FCM) using propidium iodide (PI) staining.DNA ladder bands were observed by DNA agarose gel electro-phoresis.The phosphorylated protein expression of PKD 2 was analyzed using Western blot .Results NOC (2.5, 5.0, and 10.0μmol/L) increased apoptosis rate in U937 cells in a concentration-dependent manner ( P <0.05).After treatment with NOC (5.0 and 10.0μmol/L) for 24 h, U937 cells presented typical DNA ladder bands .At the same time, not only did NOC effectively down-regulate the ex-pression of PDK2 phosphorylated protein , but also increased apoptosis rate in U 937 cells in the presence of G?6976, a specific inhibitor of PKD2.Conclusions The effect that NOC induces apoptosis in U 937 cells is related to the inhibition of the activity of PKD 2.

Objective To study the inlfuence factors on detection of activity of four coagulation factors in prothrombin complex concentrates (PCC) by several factors. Methods Using Pharmacopoeia of the People’s Republic of China (2010) as reference, the activity of four coagulation factors in PCC were investigated by choosing different pre-treatments, different diluents, different salt concentration, different standard human plasma and different company reagents. Results The activity of FII, FVII, FX were decreased and FIX was increased in the condition of adding protamine sulfate, and there were no differences of four coagulation factors whether warm bath in 37 for 15 min or not. However, the differences of four coagulation factors were significant by using deficient plasma, saline, distilled water and commercial dilution buffer(P<0.05). The activity of coagulation factor II, X in 1 mol/L salt concentration of PCC were significantly lower than in 0.25 mol/L(P<0.05), while coagulation factor VII, IX were not. The activity of FII, FVII, FIX, and FX were different by using different standard human plasma to make standard curve. The activity of four coagulation factors existed significant difference(P=0.00) by using SIEMENS company reagents and domestic reagents. Conclusion Choosing different pre-treatments, different dilution buffers, salt concentration, standard human plasma and commercial kits will inlfuence the detection result of coagulation factors.

Objective:To explore therapeutic effect of Tanshinone IIA‐sulfonic sodium on patients with coronary heart disease (CHD) and its influence on hemorheology ,cytokines and blood lipid levels .Methods :A total of 98 CHD patients treated in our hospital were selected .According to random number table ,they were randomly and e‐qually divided into routine treatment group and Tanshinone IIA group (received intravenous drip of Tanshinone IIA sulfonate sodium based on routine treatment group ) . Therapeutic effect , hemorheologic indexes , cytokines and blood lipid levels before and after treatment ,and adverse drug reaction were compared between two groups .Re‐sults:Compared with routine treatment group , there was significant rise in total effective rate (73.47% vs . 93.88% ) in tanshinone IIA group ,P=0.003. Compared with before treatment ,after treatment ,there were signif‐icant improvement in levels of blood lipid ,blood viscosity ,and cytokines in Tanshinone IIA group , P< 0.01 all . Compared with routine treatment group after treatment ,there was significant rise in HDL‐C level [ (1.32 ± 0.20) mmol/L vs .(1.74 ± 0.26) mmol/L] ,significant reductions in levels of LDL‐C [(3.27 ± 0.50) mmol/L vs .(2.03 ± 0.29) mmol/L] ,TG [ (2.69 ± 0.48) mmol/L vs .(1.43 ± 0.30) mmol/L] and TC [ (6.89 ± 1.02) mmol/L vs . (4.29 ± 0.71) mmol/L];significant reductions in plasma viscosity [ (2.59 ± 0.61) mPa s vs .(1.45 ± 0.32) mPa s] , whole blood viscosity [(4.63 ± 1.37) mPa s vs .(2.97 ± 0.85) mPa s] ,platelet adhesion rate [(36.73 ± 4.51)% vs . (30.74 ± 3.37)% ] ,levels of Fg [(4.12 ± 0.99) g/L vs .(3.04 ± 0.78) g/L] ,ICAM‐1 [(106.79 ± 30.15) pg/ml vs . (78.43 ± 14.52) pg/ml] ,CRP [(19.98 ± 3.89) mg/L vs .(10.82 ± 2.19) mg/L] and TNF‐α[(27.91 ± 4.78) pmol/L vs .(17.84 ± 3.56) pmol/L] in Tanshinone IIA group ,P<0.01 all .No severe adverse reaction was found in two groups .Conclusion:Tanshinone IIA sulfonate sodium possesses significant therapeutic effect on CHD patients .It can significantly improve patient′s hemorheology ,blood lipid and reduce cytokine levels ,and it′s safe and reliable .

OBJECTIVE:To investigate the improvement effects of andrographolide dripping pills combined with Danxi yup-ingfeng granules on immunologic function of children with allergic rhinitis(AR). METHODS:Two hundred and eight AR children were randomly divided into control group,observation groupⅠ,observation groupⅡ,observation groupⅢ,with 52 cases in each group. Control group was given Loratadine syrup 10 mL,qd;observation groupⅠwas given Danxi yupingfeng granules 15 g,bid+Andrographolide dripping pills 0.6 g,tid;observation groupⅡwas given constant dose of Andrographolide dripping pills;observa-tion groupⅢwas given constant dose of Danxi yupingfeng granules. Four groups were treated for 2 weeks. 2 weeks later,they were given Loratadine syrup orally if the symptom appeared again. The levels of IgE and Th1/Th2 were compared among 4 groups be-fore treatment,1,2 weeks after treatment,3 months after the end of treatment. The children receiving Loratadine syrup orally due to the recurrence of AR 3 months after the end of treatment were observed. RESULTS:Before treatment,there was no statistical significance in serum levels of IgE and Th1/Th2 among groups(P>0.05). One week after treatment,serum levels of IgE and Th1/Th2 were improved significantly,with statistical significance (P<0.05);but there was no statistical significance among 4 groups (P>0.05). 2 weeks after treatment,serum levels of IgE and Th1/Th2 were improved significantly,and the observation groups were significantly better than the control group,with statistical significance(P<0.05). Three months afterthe end of treatment,serum lev-els of IgE and Th1/Th2 in observation group were improved significantly and better than control group,with statistical significance (P<0.05);there was no statistical significance in control group,compared to before treatment(P>0.05). The recurrence rate of ob-servation groupⅠwas significantly lower than that of observation groupⅡ,observation group Ⅲ and control group,with statistical significance(P<0.05). No obvious ADR was found in 4 groups. CONCLUSIONS:Andrographolidume dripping pills combined with Danxi yupingfeng granules can effectively improve immunologic function of AR children and reduce recurrence rate with good safety.

OBJECTIVE To investigate the expression and roles of IL-9 and its receptor (IL-9R) mRNA in the nasal mucosa of ovalbumin-induced allergic rhinitis (AR) mice. METHODS Sixteen Balb/c mice were selected and randomly divided into two groups with 8 mice in each group. Mice were used for establishing the animal model of AR with ovalbumin sensitization as AR group, at the same time, the physiological saline as the control group. In each group, 4 mice were randomly taken from each group, and the pathological examination showed that the model was successful. The nasal mucosa was taken from the remaining 4 mice in each group and then to detect IL-9 and IL-9R mRNA in nasal mucosa of the two groups by using real-time quantitative PCR. RESULTS The expression of IL-9 and IL-9R mRNA could be detected in both the control and AR groups. The expression levels of IL-9 and IL-9R mRNA in the nasal mucosa of the AR group were both higher than that in the control group (P<0.05). The expression level of IL-9 mRNA was positively correlated with the expression of IL-9R mRNA in the nasal mucosa of mice (r =0.857, P<0.05). CONCLUSION IL-9 and its receptor IL-9R are involved in the development of AR, and play an important role in the development of allergic rhinitis. It provides a new perspective for the further understanding of the pathogenesis of AR and provides a new clue for the treatment of allergic rhinitis.

Objective To explore the expressions of adiponectin and adiponectin receptor 2 (AdipoR2) in the liver of patients with hepatitis B virus (HBV)-related liver failure and their clinical implications in the pathogenesis of liver failure.Methods The healthy controls (HC),patients with chronic hepatitis B (CHB) and patients with HBV-related liver failure (HBV-LF) were enrolled in the study.Each group had 20 participants.The liver tissues from 11 CHB patients who were diagnosed by liver biopsy,6 patients with HBV-LF and 6 liver donors during liver transplantation were collected.Histological specimens were observed by hematoxylin-eosin staining and Masson trichrome staining under microscope.The mRNA and protein expressions of adiponectin and AdipoR2 in the liver were measured by semi-quantitative reverse transcription and polymerase chain reaction (SqRT-PCR) and immunohistochemical staining,respectively.The level of serum adiponectin was detected by enzymelinked immunsorbent assay.Serum biochemical parameters and HBV DNA levels were also measured.The pairwise comparison between groups was done by Student-Newman-Keuls and mann whitney U test.The relationship between two variables was analyzed using Spearman correlation.Results The level of serum adiponectin in HBV-LF group [(0.86 ± 0.15) ng/mL] was higher than those in HC [(0.59±0.15) ng/mL] and CHB groups [(0.62±0.13) ng/mL,Z=3.963,Z=3.774,both P＜0.01)],but showing no difference between CHB and HC groups (P＞0.05).The expressions of adiponectin and AdipoR2 mRNA in the liver were significantly higher in HBV-LF group (0.251 ±0.028 and 0.223 ± 0.021,respectively) than those in HC (0.091 ± 0.018 and 0.072 ± 0.020,respectively) and CHB (0.121±0.019 and 0.097±0.017,respectively) groups (q=18.428,17.069,19.807,18.673,respectively; all P＜0.01).Also,the expressions of adiponectin and AdipoR2 mRNA in CHB group were significantly higher than those in HC group (q=3.895,3.860,both P＜0.05).The expressions of adiponectin and AdipoR2 proteins in the liver were significantly higher in HBV-LF group (8.482±0.772 and 7.654±0.272,respectively) than those in HC (4.045± 0.815 and 2.804±0.623,respectively) and CHB (5.545±0.613 and 4.775±0.458,respectively) groups (q=15.327,11.542; Z=2.802,3.266; respectively; all P＜0.01).Also,the expressions of adiponectin,AdipoR2 proteins in CHB group were significantly higher than those in HC group (q=5.894,Z=3.266,both P＜0.01).In HBV-LF group,serum adiponectin levels as well as the expressions of adiponectin mRNA and protein in the liver were negatively correlated with serum albumin (r=-0.815,-0.886,-0.943; P＜0.01 or P＜0.05),but positively correlated with serum alanine aminotransferase (r=0.701,0.886,0.943; P＜0.01 or P＜0.05).The expression of AdipoR2 mRNA in the liver was negatively correlated with serum albumin (r=-0.943,P＜0.01),but positively correlated with serum aspartate aminotransferase (r=0.829,P＜0.05).Conclusions The expressions of adiponectin and AdipoR2 are up-regulated during HBV infection,especially in patients with HBV-LF,which might reflect the degree of necroinflammation in the liver.
