3.Ultrasonic diagnosis of fetal cleft palate

Objective To explore the ultrasonic diagnosis of fetal cleft palate.Methods By applying coronal,sagittal and transverse planes to scan the fetal facial anatomy routinely, 2125 pregnant women with 20 odd weeks′ gestation were examined.Results Complete cleft palate was diagnosed in 4 cases by ultrasonography, all of which were accompanied with complete cleft lip and alveolar process.Incomplete cleft palate were missed in 2 cases.Conclusions Complete cleft palate is accompanied mostly with cleft alveolar process and complete cleft lip, and cleft palate can be found out through cleft alveolar process.To those fetuses without cleft lip or with incomplete cleft lip,their palate cannot be explored by ultrasound,and their cleft palate cannot be displayed.
4.Effects of Compound Konjac Glucomannan on Hyperlipidemia in Rats

Objective: The experiment was designed to study the effects of compound Konjac glucomannan on hyperlipidemia for the purpose of improving the therapeutic efficacy. Method: First, hyperlipidemia rat models were established. Then the effects of compound Konjac glucomannan and Konjac glucomannan on blood lipids levels and hepatic tissue morphous of rats were detected. Result; The compound Konjac glucomannan could decrease LDL-C and ApoB levels of serum TC significantly, while notably increase HDL-C and ApoA-1 levels. The result also shows that compound Konjac glucomannan could obviously improve the TG metabolism and decrease blood glucose concentration. The morphologic observation indicates that both Konjac glucomannan and flavonids could protect liver tissues of rats. Conclusion; Compound Konjac glucomannan could improve regulative ability of lipids metabolism and decrease serum lipids levels.
5.The relationship between sarcoplasmic reticulum Ca2+modulation proteins and postresuscitation myocardial dysfunction

Objective To investigate the relationship between sarcoplasmic reticulum Ca2+modulation proteins and postresuscitation myocardial dysfunction. Methods Thirty-eight SPF male Sprague-Dawley (SD) rats were randomly divided into control group(n=12)and cardiac arrest(CA)group(n=26). CA was induced by intravenous bolus of potassium chloride(40μg/g),and cardiopulmonary resuscitation(CPR)was conducted 8 minutes later. No CA was induced in control group except catheter placement for monitoring cardiopulmonary parameters after anesthesia. Invasive hemodynamic parameters were monitored for 1 hour after CPR. Echocardiogram was performed to evaluate cardiac function. Myocardial samples were harvested 5 minutes and 1 hour after restoration of spontaneous circulation (ROSC),and sarcoplasmic reticulum Ca2+ ATPase (SERCA2a),phosphorylated phospholamban (p-PLB) and rynodine receptor(RyR)were determined by Western Blot. Results ROSC rate of CA group was 92.3%(24/26),and mean recovery time was (68 ±39)seconds. Cardiac function was significantly impaired in CA group at 1 hour after resuscitation, and ejection fraction, fraction shortening (FS), the maximal rate of left ventricular pressure increase/decline (±dp/dt max)were significantly decreased compared with those in control group 〔ejection fraction:0.548±0.060 vs. 0.809±0.043,F=71.692,P=0.000;FS:(34.4±4.4)%vs. (46.0±3.5)%,F=55.443,P=0.000;+dp/dt max(mmHg/s):4 718±743 vs. 7 098±394,P<0.01;-dp/dt max(mmHg/s):-3 824±612 vs.-6 187±473,P<0.01〕. Compared with control group,the expression levels of p-PLB (gray value)was significantly decreased at 5 minutes and 60 minutes(5 minutes:0.64±0.15 vs. 1.29±0.13,P<0.01;60 minutes:0.95±0.08 vs. 1.30±0.09,P<0.05)after resuscitation in CA group,while the level of sarcoplasmic SERCA2a(gray value)and RyR (gray value)showed no significant differences(SERCA2a 5 minutes:1.01±0.18 vs. 1.24±0.07,60 minutes:1.03± 0.14 vs. 1.25 ±0.06;RyR 5 minutes:0.96 ±0.13 vs. 0.97 ±0.13,60 minutes:0.88 ±0.14 vs. 0.99 ±0.11,all P>0.05). Conclusions The impairment of the p-PLB is closely related to postresuscitation myocardial dysfunction.
6.Analysis of the Efficacy of Low-dose Progesterone Injection Combined with Baotai Wuyou Capsule in the Treatment of Threatened Abortion

OBJECTIVE:To observe the efficacy and safety of low-dose Progesterone injection combined with Baotai wuyou capsule in the treatment of threatened abortion. METHODS:107 patients with threatened abortion were retrospectively analyzed and divided into observation group(54 cases)and control group(53 cases)by different medication. All patients received bed rest orally taking folic acid and other measures. Meanwhile,control group was given Progesterone injection 20-40 mg,qd,intramuscu-larly injected. Observation group was given Progesterone injection 10-20 mg,qd,intramuscularly injected and given 4 Baotai wuy-ou capsule additionally,tid,taking with ginger decoction,no fish during the medication. The courses of 2 groups were 14 d. Clini-cal efficacy,hemostatic time,remission time of abdominal pain and backache in 2 groups were observed,and the incidence of ad-verse reactions during the treatment was recorded. RESULTS:The total effective rate in observation group was significantly higher than control group,hemostatic time,remission time of abdominal pain and backache were significantly shorter than control group, the incidence of adverse reactions was significantly lower than control group,with statistical significances (P<0.05). CONCLU-SIONS:Low-dose Progesterone injection combined with Baotai wuyou capsule shows good efficacy in the treatment of threatened abortion,which can shorten patients'hemostatic time,remission time of abdominal pain and backache,and reduce the incidence of adverse reactions,with good safety.
7.Heterologous Expression of Nanobodies: a Recent Progress

Heavy chain antibodies (HcAb) without light chains are naturally produced by camelids.The single domain antigen-binding fragment of HcAb is referred to as VHH or nanobody (Nb),which is the smallest antigen-binding entity at present.Several characteristics such as low molecular weight,high stability,and low immunogenicity make the use of nanobodies superior to the conventional antibodies.Currently,nanobodies are highly valuable antibodies for various applications,including fundamental research,diagnostics,and therapeutics.A variety of nanobodies are investigated in clinical researches.The recent progresses of heterologous expression of nanobodies in seven expression systems,including Gram-negative and positive bacteria,yeasts,filamentous fungi,insect cells,mammalian cells and plant cells were focused on,and the expression systems,hosts,characteristics of vectors,construction of vectors and yield of nanobodies were summarized.The strategies to increase the yield of nanobodies are discussed from molecular level,expression level and rational design.
8.Expression of CD35 on erythrocyte membrane and changes of serum inflammatory cytokines in children with recurrent respiratory tract infection before and after zinc treatment

Objective To investigate the expression of CD35 on erythrocyte membrane and the changes of serum inflammatory cytokines in children with recurrent respiratory tract infection (RRTI) and their relation to zinc therapy. Methods One hundred and sixteen RRTI children including 82 cases of upper respiratory tract infection and 34 cases of lower respiratory tract infection were enrolled in the study;40 children with acute respiratory infection and 50 healthy children were randomly selected as the controls. The expression of CD35 on erythrocyte membrane, positive rate of circulating immune complex (CIC), IL-6, IL-8 and TNF-α were detected. Sixty-eight RRTI children with hypozincemia were randomly divided into zinc treatment group ( n = 38) and control group ( n = 30). The above parameters were detected at the end of the treatment and 12 weeks after the treatment. Results The expression of CD35 on erythrocyte membrane was significantly lower in RRTI children ( upper respiratory group and lower respiratory group) than that in healthy controls ( t=6.17 and 6.46, P <0.01 ). CIC-pesitive rate and the contents of IL-6, IL-8 and TNF-α were increased in RRTI children, especially in those with lower respiratory tract infections. Compared with the children of acute respiratory infections, the expression of CD35on erythrocyte memhrane was much lower in RRTI children in the remission stage ( t = 20. 307, P < 0.01 ). The above parameters were improved in RRTI children who received zinc treatment. Conclusions Down-regulation of CD35, insufficient elimination of CIC, excessive production of serum IL-6, IL-8 and TNF-α were observed in RRTI children, which might be the immunopathologic mechanism of the repeated infection. These indexes can be improved after zinc treatment.
9.Angiotensin-Converting Enzyme Gene Polymorphism and Stroke

Angiotensin-converting enzyme (ACE) gene is one of major candidate genes with genetic susceptibility to stroke. However, its correlation with stroke remains controversial. The relationship between gene polymorphism and stroke also cannot be determined. A specific branch in the ACE phylogenic tree may replace the insertion/deletion polymorphic site as a risk marker of stroke for association study.
