3.Expression of aquaporin-1,3,8,9 mRNA in human amniotic membranes in polyhydranmios

Objective To determine the expression of aquaporin-1,3,8,9 mRNA (AQP-1,3,8, 9) in amniotic membranes in pregnant women with polyhydramnios. Methods Amniotic membranes were collected from women who presented with either polyhydramnios (n= 5)or normal amniotic fluid volume (control, n= 5) underwent elective cesarean sections at term. The AQP-1,3,8,9 mRNA expression were detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The expression of AQP-9 mRNA on fetal membranes were significantly higher in polyhydramnios groups (1. 1403±0. 0831) than that of control (0. 5903±0. 1909) (P = 0. 002), although the expression of AQP-1, 3 and 8 showed no significant difference between the two groups (P= 0. 972, 0. 242,0. 608, respectively). Conclusions AQP-9 may play an important role in maintaining the volume and the balance of different components of amniotic fluid in polyhydramnios cases.
4.The application of Herbert screw in the treatment of navicular bone fracture

Objective To investigate the treatment of Navicular bone fracture with Herbert screw.Methods Twenty-three patients with Navicular bone fracture were collected.They were all conducted with CT scan diagnosis and type,the small incision and targeting wire was used in operation.The Herbert screw was used to fix with pressure,plaster was also used to fix.After operation,the activity training was executed gradually.Results We conducted follow-up of 6-18 months in 23 patients,with an average of 12 months.All patients got bone-healing,the healing time ranges from 7 to 10 weeks,with an average of 9 weeks.There was one case of screw penetrating cortical bone,and one case of mild deformity healing,among the other 23 cases there were no complications including infection,nonunion,necrosis,osteoarthritis'etc.Conclusion The treatment of Navicular bone fracture with Herbert screw fixation can get firm fixation,function training and healing early,and it's worth of clinical promotion.
5.A study of the variability of the parameters of the pedicle screw path and its related factors

Objective To study the variability of the parameters of the pedicle screw path and influencing factors of clinical significance. Methods A Picker PQ6000 Spiral CT was used to scan 736 vertebrae from C 3 to L 5 of 118 men and 80 women aged between 17 and 58. The largest transverse diameter, screw path length and e angle (the angle between the axis of the pedicle and sagittal plane) of each pedicle were measured. The contour of the vertebrae and the relationship of the sagittal axis of the spinal process, vertebral body and human body were inspected. Any discrepancy of these axes was measured and recorded. The gender, age, height and weight were recorded at the same time. Results Measurement of the biggest transverse diameter, screw path length and e angle at each vertebral level showed a wide range of value and quantity. Although the screw path length and the transverse diameter of the pedicle showed significant difference between man and woman, further covariance analysis for between subject effects revealed that no inter gender difference existed after the effects of height and weight were removed. Both height and weight showed significant positive correlation with the screw path length and transverse diameter of the pedicle, but no correlation with the e angle. Observation on the deformity of the vertebrae showed that the sagittal axes of the vertebral body and human body was discrepant in 14 thoracic and lumbar vertebrae; the sagittal axes of the spinal process and vertebrae was discrepant in 11 vertebrae; the transverse diameter of the pedicle was smaller than 4 mm in 14 vertebrae below T 9; 4 vertebrae showed lateral convex pedicle. Conclusion The parameters of the pedicle screw path varied in different subjects and different vertebral level with a large variability. The weight and height are two important factors to influence the parameters. Deformity is another factor which may lead to mistake of placement of the pedicle screw. To improve the accuracy of the pedicle screw, individual identification of the entering point and orientation should be considered and in some circumstances even special screws, should be used.
6.THERAPEUTIC EFFECTS OF rhIL-11 AND rhG CSF ON MONKEYS IRRADIATED WITH 8.0 Gy ~(60) Co ?-RAY

To evaluate the effects of combined therapy of rhIL 11 and rhG CSF on monkeys irradiated with 8 0 Gy 60 Co ? ray. Animals were divided into control ( n =4) and rhIL 11+rhG CSF treatment group ( n =4). After irradiation the control was given no treatment, while the treatment group was given rhIL 11 50?g/(kg.d)+rhG CSF 10?g/(kg.d) and other symptomatic supportive treatment whenever needed. The results showed that all animals underwent nausea, diarrhea and fever. After irradiation, all blood cells in peripheral blood of the animals declined in quantity rapidly. Animals of the control died of hemorrhage and infection. Their mean surviving time after ? ray exposure was 18 2 days. Aninals of the treatment group all survived on the 45th day after exposure. Their peripheral blood cell counts recovered to near baseline level. Histopathological observation revealed that bone marrow cells of treated animals proliferated actively. The results suggested that a combination of rhIL 11 and rhG CSF treatment was significantly effective in extremely severe hematopoietic acute radiation sickness.
7.THE EFFECTS OF RECOMBINANT HUMAN STEM CELL FACTOR ON MOBILIZATION OF PERIPHERAL BLOOD STEM CELLS IN RHESUS MONKEYS

To evaluate the effects of recombinant human stem cell factor on mobilization of peripheral blood stem cells, 30 monkeys were divided into control, rhG CSF 10?g?kg -1 ?d -1 (?KD), rhSCF 50?KD, rhG CSF 10?KD+rhSCF 20?KD, rhG CSF 10?KD+rhSCF 50?KD and rhG CSF 10?KD+rhSCF 125?KD groups. The monkeys were given subcutaneous injections of rhSCF for 8 days and/or rhG CSF for 5 days. The highest value of leukocyte in rhG CSF and rhSCF groups was 206% and 200% respectively, and that of rhG CSF+rhSCF groups was 280% to 310%. CFU GM of peripheral blood was up to 1 1, 1 3 and 5 8~7 9 times respectively after the mobilization. CD34 positive cell number of joint mobilization group was 20, 24 and 35 times respectively of that before mobilization. rhSCF can mobilize the effect of peripheral blood stem cells, and the effect of joint rhSCF+rhG CSF is better.
8.Effect of hematopoietic factors mobilized peripheral blood stem cells on rhesus monkeys after irradiation of 7.0Gy ? ray

Objective To observe the curative effect of rhG-CSF or rhG-CSF+rhSCF mobilized peripheral blood stem cells (PBSC) on rhesus monkeys after irradiation of 7.0Gy ? ray.MethodsFifteen healthy adult rhesus monkeys were divided into placebo control, rhG-CSF, and rhG-CSF+rhSCF mobilized groups. rhSCF 20?g?kg -1 ?d -1 (?KD) was administered for 8 consecutive days. Then from the 4th day to the 8th day, animals of both rhG-CSF and rhG-CSF+rhSCF mobilized groups were also given rhG-CSF 10?KD. The placebo control animals were given 0.9% sodium chloride of the same volume per kilogram. On day 0 of the irradiation, PBSCs were collected from all subjects. 3~4 hrs after irradiation, all animals received autologous PBSC infusion. ResultsFrom day 21 to day 29 after TBI, rhG-CSF+rhSCF mobilized group showed higher WBC counts compared to placebo control and rhG-CSF mobilized group. Platelet counts of both mobilized groups recovered more quickly than those of placebo control. Bone marrow nucleated cells culture demonstrated that rhG-CSF and rhG-CSF+rhSCF mobilized PBSC had stimulated bone marrow nucleated cells to form more CFUs. Histopathological evaluation showed the number of hematopoietic cells in the bone marrow of rhG-CSF mobilized group was greater than those in placebo control but less than those in rhG-CSF+rhSCF mobilized group. ConclusionThe infusion of rhG-CSF or rhG-CSF+rhSCF mobilized PBSC can improve the hematopoietic recovery in rhesus monkeys after 7.0Gy ? ray irradiation. The combined use of two mobilizing factors gives better result than using single factor.
9.Determination of Alum Residual Amount in Rhizoma Pinelliae Preparata

Objective To establish a method for the determination of alum residual amount in Rhizoma Pinelliae Preparata from different companies,and to provide a scientific evidence for quality evaluation of Rhizoma Pinelliae Preparata.Methods Na2EDTA titration method was applied for the quantitative analysis.Results There were obvious differences in residual quantity of alum in Rhizoma Pinelliae Preparata from different companies.Conclusion The method is simple,reproducible and accurate,it can be used for the determination of alum residual amount in Rhizoma Pinelliae Preparata.
10.A Retrospective Analysis of Human Parvovirus B19 Infection in Childhood Immune Thrombocytopenia

Objective To confirm the role of human parvovirus B 19 ( B19) infection in childhood immune thrombocytopenia ( ITP) patients.Methods A total of 416 cases of newly diagnosed childhood ITP patients from Jan .2011 to Dec.2013 had been summarized to be cases group , Then a total of 130 childhood patients with common respiratory tract infection were selected randomly as the control group . All patients had been divided in grouped by age as <1 years group(n=187), 1-3 years group(n=127), 3-7 years group(n=71), and 7-14 years group(n=31).The incidence rate of B19 infection in all age groups, the prognosis conditions in B19 infection positive and negative groups after the same therapy protocol were obserred .Results The B19 infection rate had no statistical significance in re-search groups and control groups .And no significant difference in the same age groups compared with control groups (all P>0.05) was found.All the ITP patients had not been given anti -B19 treatment.The PLT remission rate,respectively, after treated with the same pro-tocol including glucocorticoid and/or immunoglobulin had a declining trend as the ages increasing .The B19 infection groups of all ages al-so had no significant difference PLT remission rate had been confirmed in non -B19 infection patients in each age groups (all P>0.05). Conclusions B19 infection may not be a major cause in childhood newly ITP , and treatment protocol with no anti -B19 treatment had no influence on the clinical curative efficacy .
