Objective To implore the characteristics of epithelial-mesenchymal transition(EMT) in human prostate cancer stem progenitor (S/P) cells isolated from LNCaP cell lines. Methods The S/P cells were obtained through florescence-activated cell sorting (FACS).Western blot and immunofluorescence assay were used to detect the S/P cells' EMT markers expression, such as E Cadherin,N Cadherin,Vimentin and Snail.Soft agar assay was used to detect the tumorigenesis ability of S/P cells.Cell migration assay was used to detect the migration ability of S/P cells. Results Compared with non S/P cells, the expressions of EMT markers, such as N Cadherin,Vimentin and Snail, were increased in S/P cells, while the expressions of epithelial marker and E Cadherin were decreased in S/P cells. After cultured for three weeks,S/P cells and non S/P cells both clonally grew. The colony numbers were (18.34±1.21) and (82.27±7.54),respectively (t =8.617,P =0.001).After cultured for 48 hours, the migration cells number was (25.33±5.13) in non S/P cells and (74.33±7.64) in S/P cells (t =7.953,P =O.001).Conclusions Human prostate cancer S/P cells isolated from LNCaP cell line have some characteristics of EMT,such as stronger tumorigenesis and migration ability, which could promote tumor invasion and metastasis.

Objective To analysis residents'death causes in the Three Gorges Reservoir Area, and analysis the distribution of causes of death and age, gender characteristics, therefore provide basis for governments at all levels to develop disease prevention. Methods Data for classification of death cause was analyzed by Excel 2003,according to the ICD-10 classification, the years of po-tential life lost(YPLL)was calculated by SPSS1 7.0.Results Totally 62 702 death date of resident population in Three Gorges Reservoir Area from July,2003 to December,2013 were collected. The crude death rate for males and standardized mortality rate was significantly higher than that in female;Cause of death of the top five were:circulatory system disease, respiratory system dis-ease,malignant tumor, injury and poisoning and digestive system diseases;Chronic disease was the main cause of death which ac-counting for 87.76%;years of potential life lost rate of top five were:injury and poisoning, tumor, circulatory system disease,respir-atory system diseases and perinatal diseases. Conclusion The main cause of death in Three Gorges Reservoir area is a county in the circulatory system diseases, malignant tumor, respiratory system disease, and show a younger trend. Therefore, prevent premature death has become one of the focal points of disease prevention and control.

A quantitative analysis method of multi-components with a single marker (QAMS) for simultaneous determination of six marker compounds (one from phenolic acids and five from phthalides) in Chuanxiong Rhizoma was established by applying HPLC and using butylidenephthalide as the internal reference substance. And also the feasibility and accuracy of the established method for quality evaluation and application of Chuanxiong Rhizoma were investigated and validated. The analysis was performed with the mobile phase consisting of acetonitrile - 0.2% aqueous formic acid. The flow rate was 1.0 mL . min-1 and the column temperature was maintained at 30 °C. The detection wavelengths were set at 252 nm (for ferulic acid, Z-ligustilide, and butylidenephthalide) and 266 nm (for senkyunolide I, senkyunolide A, and coniferyl ferulate), separately, and 20 µL was injected for analysis with gradient elution. The results showed that there were no significant differences observed between the HPLC-QAMS method and the external standard method (RSD <5%). The relative correction factors were credible (RSD < 5%) in changed chromatographic conditions. The established HPLC-QAMS method can be accurately used for simultaneously evaluating and controlling the quality of Chuanxiong Rhizoma with multi-components.

The general system of clinical engineering professional certification in Germany was introduced from the aspects of history, certification authority, requirement of certification, skills and knowledge points, methods of implementation and so on. Through comparative study, some suggestions were proposed to promote clinical engineering staff technology level assessment and certification, which might provide some references for the further improvement of clinical engineering certification system in China.

10. Effects and mechanism research of total saponins of Panax japonicas improving lipid metabolism in HepG2 cells based on computer aided drug design
Chinese Traditional and Herbal Drugs 2018;49(8):1835-1840
Objective To research the effects of total saponins of Panax japonicas (TSPJ) improving lipid metabolism in HepG2 cells, and to predict and verify TPSJ possible targets based on computer aided drug design. Methods HepG2 cells fatty degeneration model was induced with palmitic-acid (PA). The HepG2 cells were divided into five groups: the control group, the model group, the high-dose group (50 mg/L), the middle-dose group (25 mg/L), and the low-dose group (12.5 mg/L). The cells of five groups were cultured continuously for 24 h. The intracellular lipid accumulation was qualitative and quantitative detected by Oil red and Nile red staining. The content of triglyceride (TG) was detected by detection kit. TPSJ possible targets were predicted by computer. The expressions of related proteins were detected by immunofluorescence. Results The lipid accumulation model of HepG2 cells was successfully established for 24 h with the 100 μmol/L concentration of PA. TSPJ can significantly improve the lipid accumulation (P < 0.01), and decrease the content of triglyceride (TG) of HepG2 cells. The possible target of TSPJ may be estrogen-related receptors based on computer aided drug design. Compared with the control group, the expression levels of estrogen receptor β (ERβ) protein in model group were decreased. Compared with the model group, the expression level of ERβ protein in high-, middle-, and low-dose group were upregulated. Conclusion TSPJ can significantly improve the lipid metabolism of HepG2 cells, and the target of TSPJ might be ERβ.
