2.Comparative study on the inhibitory effect of several antihistamines on the release of inflammatory mediators from lymphocytes
Chinese Journal of Dermatology 2008;41(12):803-805
Objective To observe the effect of several antihistamines, including epinastine,on the release of leukotriene B4(LTB4),interleukin 4(IL-4)and interleukin 5(IL-5)from spleen lymphocytes of allergic mice challenged by ovalbumin(OVA).Methods OVA-sensitized BALB/c mice were used as animal model of allergy.Spleen lymphocytes isolated from these mice were treated with epinastine(0.1,1.0,10 μmol/L),ebastine(10 μmol/L),cetirizine(10 μmol/L),and dexamethasone(1.0 μmol/L),respectively,followed by incubation with OVA.Those cells without pretreatment with antihistamines served as the control. One hour after the challenge,enzyme-linked immunosorbent assay(ELISA)was applied to measure the level of LTB4,IL-4 and IL-5 in the cell culture supernates. Results Following the incubation with OVA,the levels of LTB4,IL-4 and IL-5 increased significantly in culture supernates of control spleen lymphocytes.Epinastine significantly inhibited the release of LTB4,IL-4 and IL-5 in a dose-dependent manner. At the concentration of 10 μmol/L,the inhibitory effect of epinastine was more potent than that of ebastine and cetirizine for IL-5 and LTB4 release(all P<0.05):for IL-4,the efrect of epinastine was similar to that of ebastine (P>0.05)but more potent than that of cetirizine(P<0.05).Conclusion Epinastine has a potent anti-inflammatory effect.

Chinese Pharmacological Bulletin 2003;0(10):-
Flavin-containing monooxygenase 3 (FMO3) is an important hepatic microsomal enzyme that oxidizes a number of drugs, xenobiotics and other chemicals. Many variants in the gene encoding FMO3 have been identified, some of which result in altered enzymatic activity, consenquently, altered substrate metabolism.Studies also implicate individual and ethnic differences in FMO3.Thus,it is anticipated that knowledge regarding functionally-relevant FMO3 genetic variability will become increasingly important for drug development and patient therapeutic choices.
8.The stable Expression of VEGF_(165) in Mammalian Cells
Journal of Chinese Physician 2001;0(07):-
Objective To investigate the transfection and expression of pcDNA3 1-VEGF 165 plasmid in mammalian cells. Methods pcDNA3 1-VEGF 165 plasmid was transfected into CHO cells by electroporation and the positive clones were screened by G418. The expression of VEGF 165 gene in the transfected CHO cells were detected by RT-PCR, ELISA and western blotting analysis. The biologic activities of VEGF 165 secreted by the transfected CHO cells were measured by Mile's assay and MTT method. Results G418-resistant CHO cell clones were obtained. RT-PCR, ELISA and western blotting analysis showed that there were the transcription and translation of VEGF 165 gene in anti-G418 CHO cells. The mile's assay showed that the expressed VEGF 165 could increase the vessel permeability, and MTT assay proved that it could promote the proliferation of endothelial cells. Conclusion pcDNA3 1-VEGF 165 plasmid could express VEGF 165 with biological activities in mammalian cells.

Chinese Journal of Endemiology 2016;35(7):517-519
Objective To understand the city residents' iodine nutrition level after implementation of a new iodine concentration standard. Methods Using random sampling, four counties were selected to monitor iodine nutrition level;in every county 50 children aged 8-l0 and 20 pregnant women were selected as monitoring subjects;B ultrasound scan was used to detect goiter and children urinary iodine and salt iodine levels were tested;urinary iodine concentration of pregnant women and nursing mothers was determined. Results Thyroid of 200 children aged 8-10 years were detected, the enlargement rate of thyroid gland was 1% (2/200);200 salt samples were tested, qualified iodized salt consumption rate was 96% (192/200);200 copies of children urine samples were tested, the median of urinary iodine was 229.3 μg/L;80 copies urine samples of pregnant women were tested, urinary iodine median was 183.7 μg/L.Conclusions After implementation of a new standard of salt iodine content, the iodine nutrition level of children in the four counties (districts) of Zunyi is still high, but the level of iodine nutrition of pregnant women appears imbalanced. We suggest to supply different concentrations of iodized salt for different groups of people,to strengthen the monitoring, and to prevent iodine deficiency disorders.

OBJECTIVE To investigate the effect of pranlukast(Pran) on learning and memory impairment and neuroinflammatory and apoptotic response in streptozocin(STZ)-induced type 1 diabetic mice. METHODS Male ICR mice were injected through the tail vein with STZ(150 mg·kg-1)to induce the type 1 diabetes model. Diabetic mice were administered orally with Pran. After 4 consecutive weeks of administration, the escape latency in hidden platform trials, number of platform crossings and time spent in the target quadrant of mice were assessed by the Morris water maze(MWM)test. Western blot was used to detect the proteins of cysteinyl-leukotrienes receptor-1(CysLT1R)and pro-inflammatory factors, nuclear factor-κB p65 subunit(NF-κB p65),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and cleaved caspase 3,Bax and Bcl-2 in the hippocampus and prefrontal cortex of diabetic mice. We also determined fasting blood glucose, serum insulin and lipids such as triglyceride, total cholesterol, high density lipoprotein cholesterol, and low density lipoprotein cholesterol. RESULTS The data of the MWM test showed that untreated diabetic mice displayed a higher escape latency in hidden platform trials(P<0.05),and a smaller number of platform crossings(P<0.05)as well as shorter per?centage of time spent in the target quadrant(P<0.05). The data of Western blotting showed that treat?ment with Pran 0.6 and 1.2 mg·kg-1 significantly reduced the levels of CysLT1R,nuclear NF-κB p65, IL-1βand TNF-α,cleaved caspase 3,and the ratio of Bax and Bcl-2 in the hippocampus and prefrontal cortex of diabetic mice(P<0.05). However,Pran did not improve the fasting blood glucose, serum insulin or lipid metabolism disorder in diabetic mice. CONCLUSION Pran improves memory impairment and nerve injury in STZ-induced type 1 diabetic mice.
