AIM: To assay the level of ferulic acid in rabbit taking Siwu Decoction(Radix angelicae sinensis,Rhlzoma chuanxiong,Radix rehmanniae praeparata) and the influence result of the blood picture. METHODS: The level of ferulic acid in rabbit blood was measured with HPLC after medication and the blood examination was carried out. RESULTS: During the period of medication,the levels of red and white blood cells,hemoglobin,Fe,Zn and Cu increased,packed cell volume and whole blood low-shear viscosity decreased. CONCLUSION: Ferulic acid in Siwu Decoction might be one of effective constituents,promoting blood circulation and replenishing blood.

4.Activation of Wnt signaling pathway in the neuronal differentiation of bone marrow mesenchymal stem cells:a microarray analysis

BACKGROUND:It wil provide a new insight into the future application of bone marrow mesenchymal stem cels in the treatment of spinal cord injury and tissue engineering by studying the effect of activation of Wnt signaling pathway in the neuronal differentiation of bone marrow mesenchymal stem cels.
OBJECTIVE: To detect the expression of related genes by gene chip technology during the neuronal differentiation of bone marrow mesenchymal stem cels.
METHODS:Human bone marrow mesenchymal stem cels were isolated and purified, and passage 5 cels were obtained. GatewayTM technology was used to build lentiviral vectors that was used to transfect Wnt-1 into human bone marrow mesenchymal stem cels. Control, non-transduction and transduction groups were set in this study. Human bone marrow mesenchymal stem cels were then induced to differentiate into neurons. Cel morphology was observed under inverted phase contrast microscope. Gene chip was used to detect the regulation changes and the differential expression of related genes in the Wnt signaling pathway.
RESULTS AND CONCLUSION: Under the scanning electron microscope, the transfected cels were found to have the similar morphology of neuron-like cels. Analysis by the gene chip hybridization technique showed that 3 287 genes were up-regulated and 4 215 genes were down-regulated in the signal pathway. In the Wnt signaling pathway, genes related to the nervous system development and differentiation were up- or down-regulated. It is verified that the Wnt signal pathway is activated via Wnt-1 transduction, and the downstream genes appear to have genetic transcription so as to promote the neuronal differentiation of human bone marrow mesenchymal stem cels.
5.Operation of latero-anterior internal fixation by plate screw and vertebroplasty by calcium sulfate cement for treatment of thoracolumbar vertebral compression fractures with osteoporosis

[Objective]To evaluate the therapeutic effects of the operation of latero-anterior internal fixation by plate screw and vertebroplasty by calcium sulfate cement for treatment of thoracolumbar more vertebral compression fractures with osteoporosis.[Method]Twenty-two patients with osteoporosis and more vertebal compression fractures received latero-anterior internal fixation by plate screw and vertebroplasty by calcium sulfate cement.Functions of spinal cord nerve were damaged in different degrees.Preoperative auxiliary examination of X-ray,CT and MRI was used.The spinal stabilization,spinal cord function and the state of chronic lumbodynia were observed by postoperative photographs.[Result]All the patients were followed up,and the implants showed bone union.Sensation and motor function were improved in various degrees.The pain of chest and back or other complaints were obviously relieved.No recurrence occurred.Vertebral bodies by vertebroplasty had no leakage and the short-term effects were satisfactory.[Conclusion]The operation of latero-anterior internal fixation by plate screw combining vertebroplasty is profitable to simultaneously improve nerve compression of thoracolumbar kyphosis,spine instability,and other chronic pain caused by osteoporotic vertebral compression fractures.

8. The effects of radiotherapy and concurrent chemoradiotherapy on cell proliferation, apoptosis and cell cycle of cervical squamous carcinoma

Objective: To clarify the molecular mechanism underlying the response of locally advanced cervical squamous carcinoma to radiotherapy or concurrent chemoradiotherapy. Methods: Forty-nine patients were divided in to two groups. The 25 patients in radiotherapy (RT) group were given radiotherapy alone. The 24 patients in the concurrent chemoradiotherapy (CCRT) group were given 3 cycles of chemotherapy (DDP + 5-FU) in addition with RT. The specimens of the cervical tumors were obtained by biopsy before and during the treatment (10 Gy irradiation for RT group and 10 Gy irradiation plus one cycle of DDP + 5-FU chemotherapy for CCRT group). The cell cycle distribution was analyzed by flow cytometry (FCM). The apoptosis and PCNA expression in both groups were detected by TUNEL and immunohistochemical methods. Results: Apoptosis index (AI) and the positive rate of apoptosis were significantly increased after initiating RT and CCRT, respectively (P < 0.05, P < 0.001). There were also marked reduction of PCNA after 1 week of RT and CCRT (P = 0. 000 and P = 0.000). The CCRT significantly increased the apoptosis rate and decreased PCNA expression than RT (P = 0.03 and P = 0.005). Most cervical cancer cells were arrested in G2/M phase during RT treatment. However, CCRT induced S and G2/M phase arrest in a majority of cervical carcinoma cells. Conclusions: The additive or synergistic effects between chemotherapy and radiotherapy may be the mechanism for the effect of CCRT on locally advanced cervical squamous carcinoma CCRT induces apoptosis of tumor cells by inhibiting cell proliferation, inducing S and G2/M phase arrest, and synchronizing cell cycle.
9.Establishment of reference range for thromboelastography in pregnancy

Objective To establish a reference range for the normal value of thromboelastography (TEG) in pregnant females.Methods According to the results of pregnancy and physical examination,166 pregnant females and 64 healthy females without pregnancy were selected as the pregnant group and the non-pregnant control group,respectively.The TEG value and the traditional coagulation index were measured.The TEG parameters of the two groups were compared and analyzed,establishing a reference range for the parameters.We further analyzed the effect of full-term pregnancy on TEG results and the correlation between traditional coagulation index and TEG test results.Results The traditional coagulation index and TEG test results of the pregnant females andthe non-pregnant females were significantly different.According to the results,a new TEG reference range was established:R 3.9-7.5 min,K 1.0-2.4 min,α 57.6°-74.9°,MA 55.7-75.7 mm,LY30 0-0.56%,CI(-0.97)-3.6.Full-term pregnancy had no significant effect on TEG results.In addition to LY30,other parameters of TEG had some correlation with the traditional coagulation index.Conclusions The general TEG reference range does not apply to pregnant females and established TEG normal reference range for pregnant females can be applied for clinical use.
10.Synchronizing presentation of cyclins and merlin in single cell in vestibular Schwannoma tissues with confocal laser microscopy

Objective To explore the expression of merlin and cyclins and the nucleocytoplasmic shuttling of merlin in abiogenous vestibular schwannoma tissues. Methods Four paraffin imbedding vestibular Schwannoma samples were included in the study.The cyclins and merlin were analyzed by immunofluorescence at the same time,and the expression and location were synchronously observed with confocal laser microscopy(CLM) in a single schwannoma cell. Results Cyclins and merlin were perspicuously and synchronously presented in a single cell in vestibular schwannoma tissues.In early G1 phase of the tumor cell,merlin was mainly stained in the nucleus,followed by perinucleus and cytoplasm.In G1/S phase,merlin was concentrated in the perinucleus,less in the nucleus and cytoplasm.Merlin staining showed uniform distribution in the nucleus in G2/M phase,while was absent from the nucleus and concentrated in the perinucleus in late G2/M phase. Conclusion The expression of merlin and various kinds of cyclins can be synchronously presented in abiogenous vestibular schwannoma cells with immunofluorescence and CLM.Merlin is shuttling among the nucleus,cytoplasm and perinucleus along with the progression of cell cycle.
