1.The influence of Sodium Butyrate on hybridoma proliferation and antibody production

Using different concentration of sodium butyrate in RPMI 1640-20 or serum-free medium to culture hybridoma cells which secreted monoclonal antibodies (McAbs) against tissue-type plasminogen activator (t-PA). The results indicate that 0.25mmol/L-0. 5mmol/L of sodium butyrate could incite hybridoma cells to produce McAbs and had no inhibition on hybridoma cell growth. The maximum titer of McAb was 1:2500, 2-4 fold more than control. Sodium butyrate may be as an additive to enhance the ability of hybridoma secreting McAbs.
2.Nomograms prediction model for bladder cancer

There is remarkable heterogeneity in the history of the bladder cancer,which makes it essential to evaluate individualized prognosis risk of each patient.The Nomograms model is more convenient and accurate than other prognostic models,can provide significantly accurate individualized risk estimations that facilitate management decisions.The Nomograms model has been regarded as the most accurate and discriminating prognosis model to the patient of bladder cancer,and its development has a bright future.While the Nomograms Model has several weakness itself,which would be remedied through more researches and new predictors.Before Nomograms Model can be accepted in practice,we really need better evidence that they improve patient care and outcomes.
3.A novel biomimetic nanofibrous heparin/gelatin scaffold with sustained-release recombinant human bone morphogenetic protein-2:preparation and characterization

BACKGROUND:Bionic structure is one of the most important goals of scaffold design in tissue engineering. However, the majority of the bionic scaffolds do not have the capacity of sustained release of growth factors.
OBJECTIVE:To fabricate a novel biomimetic nanofibrous heparin/gelatin scaffold and investigate the characterization of the scaffolds including the microstructure, the mechanical strength andin vitro recombinant human bone morphogenetic protein-2 (rhBMP-2) release.
METHODS:Heparin-conjugated gelatin was prepared by chemical crosslinking and mixed with gelatin at different ratios. Nanofibrous composite scaffolds with different ratios of heparin-conjugated gelatin (0, 20% and 40%) were fabricated by using a thermaly induced phase separation combined with particle leaching technique. The amount of conjugated heparin in the scaffolds, scaffold microstructures and mechanical strength were detected and observed. The release kinetics of rhBMP-2 from the scaffolds with different concentrations of heparin conjugated gelatin was detected using ELISA method.
RESULTS AND CONCLUSION:The composite scaffolds possessed abundant porosities, good mechanical properties, and biomimetic nano-structures. The macroporous size was 250-420 μm. When the ratio of heparin-conjugated gelatin was 0, 20% and 40%, the content of heparin in the scaffolds was 0, (12.82±2.49) and (25.08±3.69) mg/g, respectively, with significant differences; the elastic modulus of the composite scaffolds was (299±38), (286±40) and (236±33) kPa, respectively, with insignificant differences (P > 0.05). With the increase in the content of heparin-conjugated gelatin, the rhBMP-2 adsorption capacity of the nanofibrous heparin/gelatin scaffold is improved, thereby significantly reducing the burst release of rhBMP-2 and prolonging the releasing time.
4.The Diagnostic Value of Multiplanar Reconstruction(MPR) of Spiral CT inDegenerative Disorders of Lumbar Spine

Objective To explore the diagnostic value of multiplanar reconstruction(MPR)of spiral CT in the disorders of lumbar intervertebral discs.Methods Fourty-one cases with the disorders of lumbar intervertebral discs were examined with conventional CT scans and MPR of spiral CT.CT findings of intervertebral discs at L 3/4 ,L 4/5 and L 5/S 1 were compared and observed.Results The conventional scans showed bulging and /or herniation of intervertebral discs in 35 cases,and normal in 6 cases which compression of nerveroots by intervertebral discs or osseous narrow of intervertebral discs were showed on MPR.The MPR showed compression of nerve roots,compression of dural sac or osseous narrow of intervertebral foramens in 36 cases.Conclusion MPR possesses important value in diagnosing the disorders of lumbar intervertebral discs,showing the causes of lumbar vertebral diseases and distinguishing the various diseases of lumbar vertebrae.
5.Effects of xeroderma pigmentosum group D and p53 on replication of HBV

AIM:To investigate the effects of xeroderma pigmentosum D ( XPD) and p53 on the replication of hepatitis B virus ( HBV) .METHODS:Recombinant plasmid pEGFP-N2/XPD and vacant vector plasmid pEGFP-N2 were transfected into HepG2.2.15 cells by liposome.On the next day, these cells were incubated with pifithrin-α, a p53 inhibi-tor, at a concentration of 20 μmol/L for 24 h.The cells were divided into 5 groups: blank control group, pEGFP-N2 group, pEGFP-N2/XPD group, pEGFP-N2/XPD+pifithrin-αgroup and pifithrin-αgroup.The mRNA expression of XPD, hepatitis B surface antigen ( HBsAg) , hepatitis B e antigen ( HBeAg) and hepatitis B virus X protein ( HBx) was detected by RT-PCR.The content of HBsAg and HBeAg in the supernatants of culture medium was measured by ELISA.The con-tent of HBV-DNA in the supernatants of culture medium was examined by fluorescence quantitative PCR.Using the method of bDNA, the content of HBV-DNA in the core particles was assessed.RESULTS:The expression of XPD mRNA was ele-vated by the transfection of recombinant plasmid pEGFP-N2/XPD.The increase in XPD expression significantly down-regu-lated the mRNA expression of HBsAg, HBeAg and HBx.The content of HBsAg and HBeAg in the supernatants of culture medium was significantly decreased by the increase in XPD expression.The results of fluorescence quantitative PCR showed that the content of HBV-DNA in the supernatants of culture medium was significantly down-regulated by the increase in XPD expression.bDNA results showed that the content of HBV-DNA in the core particles was significantly decreased by the increase in XPD expression.Pifithrin-αabolished the above-mentioned effects of XPD (all P<0.01).CONCLUSION:XPD inhibits the replication of HBV through p53 pathway.Therefore, XPD and p53 may be the targets for antiviral therapy of hepatitis B.
6.Application of low damage endotracheal intubation in patients undergoing extra long-term endotracheal intubation:a case report

Objective To approach the effect of low damage endotracheal intubation on reducing the occurrence of cuff-related intubation complication and prolonging the intubation time. Methods On January 7th, 2015, 1 patient with respiratory failure after subarachnoid hemorrhage were admitted to Huludao Central Hospital. Immediate endotracheal intubation and ventilator assisted ventilation were performed. When the trachea was difficult to be removed in a short time, and tracheotomy was refused, a low damage endotracheal intubation was used for a long term. On the basis of the original high volume low pressure cuff, this tube was designed for inner cuff, the hole was allowed in the inner cuff to connect with the tube. During the period of ventilation, the cuff pressure changed with airway pressure automatically, therefore it would reduce the compression injury of tracheal mucosa, improve the tolerance of the patients, and prolong the intubation time. Results The patient was removed from the tube on October 12th, 2015 with an intubation day of 279, the intubation-related complications and severe aspiration pneumonia had not been observed during the application of low damage endotracheal intubation. Conclusions Design of this intubation ensured the cuff pressure changes with airway pressure, therefore, it could effectively avoid the cuff pressure become too high, and reduce the occurrence of intubation-related complication. This low damage endotracheal had an evident superiority in the aspects of cuff management. It has a better practical significance, especially for patients with long intubation time.
7.The effect of HGF-mediated Skp2 down-regulation on suppression of proliferation of human hepatocellular carcinoma cells

Objective To investigate the effect of hepatocyte growth factor(HGF)on the proliferation of human hepatocellualr carcinoma cells,and the mechanism of HGF-induced proliferation inhibition.Methods Human hepatocellualr carcinoma cell line HepG2 were treated with different concentrations of HGF for different time periods,and the proliferation of these cells was examined by colorimetric BrdU cell proliferation enzyme-linked immunosorbent assay.The expression of S-phase kinase associated protein 2(Skp2)was examined using Western blot and RT-PCR.Plasmids pcDNASkp2 was introduced into HepG2 cells,then the clones showing up-regulation of Skp2 were selected,and the effect of HGF on the proliferation in these clones was investigated.Results HGF inhibited the proliferation of hepatoma cells in a dose and time dependent manner.The expression of Skp2 was significantly suppressed by HGF.Furthermore,HGF did not suppress the proliferation of HepG2 cells transfected with Skp2.Conclusions This study suggests that HGF could inhibit HepG2 cell proliferation,and the down-regulation of Skp2 could be closely related to this suppressed proliferation.
8.Laparoscopic mesh repair of parastomal hernia: Experience of 7 cases

Objective To investigate the effectiveness of laparoscopic mesh repair of parastomal hernia. Methods Laparoscopic mesh repair was performed in 7 patients with parasromal hernia from September 2004 to December 2005 in this hospital.Results Laparoscopic mesh herniorrhaphy was successfully performed in 6 patients, while a conversion to open herniorrhaphy was required in 1 patient because of extensive intraperitoneal adhesion. The operating time was 45~180 mi (mean, 109 min), and the hernial diameter was 4~6 cm (mean, 5.6 cm). Transient abdominal distention happened in 2 patients. Five patients complained of a pain in operative area within 3 weeks. Seroma occurred in 4 patients and was cured by needle aspiration and pressure bandaging for 2~ 4 times. No hematoma or surgery-related infection was seen. The length of postoperative hospital stay was 3~8 d (mean, 5.1 d). Postoperative follow-up for 2~15 months (mean, 8.3 months) revealed no recurrence. Conclusions Laparoscopic mesh repair of parastomal hernia is safe, effective, and feasible.
9.Treatment for Complications of Laparoscopic Incisional Hernias Repair

Objective To explore the causes and treatments for complications of laparoscopic incisional hernia repair.Methods A total of 110 patients with incisional hernia(42 men and 68 women,aged 33 to 89 years with a mean of 65)were treated with laparoscopic mesh repair in our hospital from March 2004 to May 2007.The clinical records of the patients were analyzed retrospectively.Results The laparoscopic repair was completed in all of the patients expect in one,who was converted to open surgery because of intraperitoneal adhesion.After the operation,15(13.6%)patients developed abdominal distension,86(78.2%)had severe pain in the operative area(not less than 6 weeks in 2 cases),23(20.9%)had seroma,6(5.4%)had urinary retention,2(1.8%)had acute gastric dilation,2(1.8%)had intestinal fistula,2(1.8%)had respiratory dysfunction,and 2(1.8%)showed recurrence of the hernia.Conclusion Most of the complications of laparoscopic incisional hernia repair are caused by insufficient preoperative preparation,intraperitoneal adhesion,difficulty in exposure of the visual field,improper use of electrocoagulation,small-sized masses,or weak fixation.
