3.Preparation and pharmacokinetics of imperatorin sustained-release tablets

Objective To prepare imperatorin sustained-release tablets and study their pharmacokinetics and bioavailability in rabbits.Methods Hydroxy-propyl methyl cellulose(HPMC) and carbomer were used to constitute the binary polymer matrix.Orthogonal design was used to optimize the formulation of imperatorin sustained-release tablets.The preliminary pharmacokinetic study in the rabbits was carried out by single-dose crossover design using imperatorin plain tablets as control.An appropriate GC-MS method was developed for measuring the concentration of imperatorin in the blood.Results Optimized formulation contained 1∶20 HPMC K100M and 2∶25 carbomer.Single dose test with 300mg showed that the relative bioavailability of imperatorin sustained-release tablets was(115.37?45.46)%;AUC of the studied tablets and control tablets was(391.08?47.22) and(368.25?136.15)?g/(h?L),tmax(2.26?0.25) and(0.33?0.05)h,Cmax(59.66?6.28) and(295.91?127.00)?g/L,T1/2ke(2.27?0.09) and(0.60?0.10)h,respectively.Conclusion The release behavior of the imperatorin sustained-release tablets prepared with binary polymer system shows evident sustained-release characteristics.
4.Treatment of large persistent primitive trigeminal artery aneurysm with blood flow diverter: a case report and literature review

Objective:To report a case of large persistent primitive trigeminal artery aneurysm treated with dense reticular blood flow guide device (flow diverter, FD).Methods:The imaging and clinical data before and after treatment with FD of a 54 years old female patient with large persistent primitive trigeminal artery aneurysm with abducent nerve and oculomotor nerve insufficiency in the Department of Neurosurgery, the First Affiliated Hospital of Jinan University in December 2018 were analyzed retrospectively, and the related literature was reviewed.Results:Digital subtraction angiography (DSA) showed that the left primitive trigeminal artery was not closed, and the internal carotid artery communicated with the basilar artery through the original trigeminal artery. The aneurysm was located at the beginning of the original trigeminal artery and the size was about 18.41 mm×12.8 mm. The contrast agent in the aneurysm was significantly retained after the cavernous sinus segment of the internal carotid artery was implanted with FD. Eight months after operation, the left blepharoptosis disappeared, strabismus disappeared and eyeball movement was normal. DSA showed that there was no obvious development of aneurysm in the original trigeminal artery.Conclusion:For mature persistent primitive trigeminal artery related large or giant aneurysm, FD may be a better means of treatment.
5.Mechanics Analysis on Novel Micro-Movement Elastic Implant of Distal Tibiofibular Syndesmosis in Dorsiflexion Position

Objective To study mechanical properties of the novel micro-movement elastic implant of distal tibiofibular syndesmosis in dorsiflexion position.Methods A combination of simulation and experiment was used. The normal ankle dorsiflexion model, the bone nail repaired model and the micro-movement elastic implant repaired model were established by using Mimics and CT data. Besides, ANSYS Workbench was used for finite element analysis. Mechanical experiments on lower limb specimens were conducted on Instron E10000 mechanical test instrument, and five sets of experimental data were measured and analyzed.Results Mechanical properties of the micro-movement elastic implant repaired model were closer to those of the normal ankle dorsiflexion model, but stress of the micro-movement elastic implant repaired mode was greater than that of the bone nail repaired model. The results of two-sample heteroscedasticity t test indicated that there was no significant difference in resistance torque between the micro-movement elastic implant repaired model and the normal ankle dorsiflexion model.Conclusions For dorsiflexion position, repairing mechanical properties of the novel micro-movement elastic implant are much better than those of the bone nail, and there is still room for optimizing the micro-movement elastic implant.
