1.Apoptotic cells and related factors in ovariectomized rat osteoporosis model

Objective To investigate the status of apoptosis of osteoblast, osteoclast and the relative factors. Methods Apoptotic cells were examined using in situ end labeling technique. Expressions of bcl-2, Fas and transforming growth factor(TGF)β1 were observed with immunohistochemical method. Results Positive expression rate (40.5±5.2)% of apoptosis cells was significantly increased in the osteoblast, but the expression rate of apoptosis cells (8.4±1.2)% was significantly decreased in the osteoclast of the post-ovariectomy rats than in the sham-operated rats. The expression of Fas was decreased in osteoclast (20.0%) and increased in osteoblast (80.0%) of the post-ovariectomized rats. The expression of TGFβ1 was significantly decreased in the osteoclast (0) and the osteoblast (20.0%) of the post-ovariectomized rats. The above-mentioned targets in post-ovariectomized rats treated with nilestriol/levonorgestrel were close to those sham-operated rats (P>0.05). Conclusions Bone loss in the post-ovariectomized rats is attributable to the lower level of estrogen so that the apoptosis cells were decreased in osteoclast but were increased in osteoblast. As a result, the process of bone absorption prevails over that of bone formation. Expression of TGFβ1 depends on stimulation of estrogen. TGFβ1 may stimulate apoptosis of osteoclast and restrain apoptosis of osteoblast. Fas can induce the apoptosis of osteoclast.
2.Determination of copper,zinc and maganese in Ejiao by derivative flame atomic absorption spectrometry

OBJECTIVE:To determine the contents of Cu,Zn and Mn in Ejiao,a kind of Chinese traditional medicine.METHOD:The contents of Cu,Zn and Mn were determined by derivative flame atomic absorption spectrometry (DFAAS) after digested by HNO3-HClO4.RESULTS:The contents of Cu,Zn and Mn were 10.48,12.38 and 18.09μg.g-1 respectively.CONCLUSIONS:The DFAAS possesses higher sensitivities,lower detection limits and better precision.
3.Effect of salvianoli acid B and hyperoxic solution on experimental ischemia reperfusion injury of extremities

[Objective]To investigate the effect of salvianoli acid B and hyperoxic solution on experimental ischemia reperfusion injury of extremities.[Method]Twenty-four healthy New Zealand rabbits were divided into 4 groups randomly.Equivalent 0.9%Nacl(group A),hyperoxic solution(group B),salvianoli acid B(group C),salvianoli acid B and hyperoxic solution(group D) were injected through ear edge vein before ischemia.The femoral artery and vein were blocked and the model of ischemia reperfusion injury of extremities was set up.Before ischemia and after 4 hour's reperfusion,the blood samples were taken to measure the levels of blood malondialdehyde(MDA) and superoxide dismutase(SOD).The gastrocnemius muscles were observed by pathologic test.[Result]Serum enzymatic activity of MDA improved obviously.Salvianoli acid B and hyperoxic solution could prevent the improvement of it(P
4.Effect of Comprehensive Treatment on Common Peroneal Nerve Injury

Objective To study the effect of comprehensive treatment on common peroneal nerve injury. Methods 80 patients with common peroneal nerve injury were divided into treatment group (n=40) and control group (n=40). The treatment group accepted ultrashort wave, modulated medium frequency electrotherapy and electromyographic biofeedback therapy, while the control group accepted medication,acupuncture and excecise. All the patients were assessed with Fugl-Meyer assessment (FMA) in lower limbs before and after treatment.The effectiveness were recorded with MCRR. Results The scores of FMA of lower limb significantly improved after treatment and improved more in the treatment group than in the control group (P<0.05). The excellent and good rate was 80.00% in the treatment group, and 50.00% in the control group (P<0.01). Conclusion It is effective of comprehensive treatment on common peroneal nerve injury.
5.Effect of Electromyographic Biofeedback Therapy and Kinesitherapy on Radial Nerve Injury

Objective To observe the effect of electromyographic biofeedback therapy and kinesitherapy on patients with radial nerve injury.Methods 82 patients with radial nerve injury were divided into control group (n=41) and treatment group (n=41). The control group was treated with medcine and acupuncture, while the treatment group received electromyographic biofeedback therapy and kinesitherapy in addition. The radial nerve function was assessed and analyzed by electromyogram (EMG) and motor conduction velocity (MCV) before and after treatment. Results After 12 weeks of treatment, limb motor function, EMG and MCV were better in the treatment group than in the control group (P<0.05). Conclusion Electromyographic biofeedback therapy and kinesitherapy can effectively improve the limb motor function of patients with radial nerve injury
6.Preparation and pharmacokinetics of imperatorin sustained-release tablets

Objective To prepare imperatorin sustained-release tablets and study their pharmacokinetics and bioavailability in rabbits.Methods Hydroxy-propyl methyl cellulose(HPMC) and carbomer were used to constitute the binary polymer matrix.Orthogonal design was used to optimize the formulation of imperatorin sustained-release tablets.The preliminary pharmacokinetic study in the rabbits was carried out by single-dose crossover design using imperatorin plain tablets as control.An appropriate GC-MS method was developed for measuring the concentration of imperatorin in the blood.Results Optimized formulation contained 1∶20 HPMC K100M and 2∶25 carbomer.Single dose test with 300mg showed that the relative bioavailability of imperatorin sustained-release tablets was(115.37?45.46)%;AUC of the studied tablets and control tablets was(391.08?47.22) and(368.25?136.15)?g/(h?L),tmax(2.26?0.25) and(0.33?0.05)h,Cmax(59.66?6.28) and(295.91?127.00)?g/L,T1/2ke(2.27?0.09) and(0.60?0.10)h,respectively.Conclusion The release behavior of the imperatorin sustained-release tablets prepared with binary polymer system shows evident sustained-release characteristics.

8.Treatment of post interventional syndromes in adenomyosis with Huayuxiaojie decoction

Objective To observe the clinical effect of treating adenomyosis uterine artery embolization (UAE)after interventional therapy for adenomyosis with Huayuxiaojie decoction. Methods A total of 40 patients with adenomyosis were randomly recruited into a control group and a treatment group. 20 patients in the control group were not given any medical treatment after interventional operation; while 20 patients in the treatment group were given the decoction of Huayuxiaojie after intervention. Changes of menstrual blood volume, menalgia degree, CA125 and uterine volume of all patients were investigated after the treatment. Results Patients in the treatment group showed significantly better results than the control group in the improvement of menstrual blood volume, dysmenorrhea, CA 125, and uterine volume (P<0.05) .Conclusion Huayuxiaojie decoction was effective in relieving menalgia, adjusting menorrhagia, soothing bellyache,reducing fever of syndromes after interventional operation of adenomyosis.

OBJECTIVE To investigate the transgenerational effects of a hypothala mic-pituitary-ad-renal (HPA)axis -associated neuroendocrine metabolic progra mming alteration fro m F1 to adult second generation (F2)with prenatal ethanol ingestion.METHODS Pregnant Wistar rats were ad ministered with ethanol (4 mg·kg -1·d -1 )fro m gestational day 1 1 until delivery.F1 rats were fed a high-fat diet fro m postnatal week 4 (PW4)and were cross-mated in PW 16 -20.F2 rats were fed a standard diet fro m PW4 and rectal te mperature was measured in PW20,oral glucose tolerance test (OGTT)was con-ducted in PW21 ,blood sa mples and hypothala mus were collected in PW24 to investigate seru m lipids and HPA axis activity.RESULTS Co mparing to the F2 control group ,rectal te mperature in F2 ethanol group were higher (P<0.01 ),sugar tolerance in F2 male group was i mpaired (P<0.05),seru m corti-costerone and hypothala mus arginine vasopressin (AVP) mRNA were increased (P <0.05);seru m insulin were decreased (P<0.05)and male rats showed i mpaired glucose tolerance (P<0.05);seru m high-density lipoproteincholesterol (HDL-C)decrease (P <0.05)and total cholesterol (TCH)/HDL-C and low density lipoproteincholesterol (LDL-C)/HDL-C ratios were markedly increased (P <0.05,P <0.01 ).CONCLUSION Prenatal ethanol exposure induced metabolic syndro me has transgenerational effects,which may originate fro m the intrauterine progra mming of altered HPA axis-associated neuroen-docrine metabolis m.

Objective To explore the expressions and significances of p 53、bcl-2、Ki-67 and apopto-sis in keratoacanthoma(KA)and well differentiated squamous cell carcinoma (WDSCC).Methods The expres-sions of p53、bcl-2、Ki-67 were detected by immunohistochemical technique in 44 cases of KA and 20 cases of WDSCC.Apoptosis was identified by using terminal deoxynucleotidyl transfers ( TdT) -mediated dUTP -biotin nick and labelling(TUNEL)method.Results The positive expressive rates of p53 in proliferative、mature and re-gressive of KA were 22.23%、26.18% and 6.52%, which was lower than the positive rate of WDSCC (41.82%).Significant differences were found between every period of KA with WDSCC in p 53 expression;The intensity of expression and pattern were similar in KA of Ki -67 and p53.There were a positive correlation be-tween p53 and Ki-67 expression rate(r=0.986,P<0.001).The bcl-2 staining showed weak expression in 1 case in WDSCC and 2 cases in KA ,but only a few positive tumor cells was limited to basal cell layer in KA .The average apoptosis rate in KA was 21.72%,which was apparently higher than in WDSCC (9.925%).There were a negative correlation between the apoptosis rate and proliferation rate of Ki -67(r=-0.824,P<0.001).Con-clusion The proliferation and apoptosis in KA can coexist .However ,the apoptosis will occupy a dominance posi-tion in regressive phase ,which results in the regression of tumor .The expressions of p53、Ki-67 and apoptosis would be a certain significance in differentiated KA and WDSCC .
