UCH-L1(ubiquitin carboxy-terminal hydrolases L1)is a member of the carboxyl-terminal ubiquitin hydrolase family,naturally,that there is hydrolase activity,however,and that there is ligase acitivity,which is different from the other members of UCHs.UCH-L1 hydrolase activity could keep the pool of free ubiquitin and compromise the ubiquitin-dependent degradation pathway,while UCH-L1 dimerizational-dependent,ubiquityl ligase activity could produce undegradable,K63-linked polyubiquitin chains that could inhibit proteasomal activity.Therefore,UCH-L1 is involved in the more diverse physiological activities,including neuron formation,gonadal development and fertilization.Mutation of UCH-L1 is linked to the neurodegenerative diseases,such as Parkinson's disease.Moreover,abnormal expression of UCH-L1 is response to carcinogenesis in many tissue such as thyroid lung.

Objective: To explore the effects of different positions, epoch lengths and cut points of accelerometers on the measurement of sedentary behaviour (SB), light physical activity (LPA) and moderate vigorous physical activity (MVPA) in school aged children, so as to provide a reference for optimizing the criteria of accelerometers to measure SB and physical activity in school aged children. Methods: From May 2021 to September 2022, 110 school aged children aged 8-12 years old were convenient selected from a primary school in Hangzhou, and the accelerometers were worn on the waist, back, thigh, and wrist during the school time, and the collected data were transformed into six datasets with different epoch lengths (1, 5, 10, 15, 30, and 60 s) and compared using 4 common cut points developed by Puyau, Freedson, Evenson, and Pulsford (a total of 96 combinations) for classifying the percentages of time spent in SB, LPA, and MVPA. The effects of different positions, epoch lengths and cut points and their interactions on the assessment results were analyzed using a three way repeated measures ANOVA, and the variability of multiple comparisons between groups was analyzed using a Bonferroni post hoc test. Results: Except for the interaction of cut point × position × epoch on the percentage of time spent in SB and the main effect of epoch on he percentage of time spent in MVPA were not statistically significant ( F =0.66，1.18， P >0.05), the remaining main effects and interactions of the three factors on the assessment results were all statistically significant ( F =6.28-11 662.28， P <0.01). The differences between the results of different positions were statistically significant ( F =90.98-308.79, P <0.01). The percentages of time spent in SB and MVPA decreased with the increase of epoch lengths, while the percentage of time spent in LPA increased with the increase of epoch lengths ( F =16.54- 676.35 , P <0.01). The difference between the results using different cut points measurements was statistically significant ( F = 98.14- 976.40, P <0.01). Conclusions Accelerometer methodological factors including positions, epoch lengths and cut points will affect the estimates of SB and physical activity in school aged children. Therefore, it is recommended that future studies need to optimize the applicability of accelerometer analysis methods in school aged children, and the comparisons of results between studies need to be treated with caution.

The effects of ghrelin on the proliferation and differentiation of 3T3-L1 preadipocytes and the possible mechanisms were investigated in this study.3T3-L1 preadipocytes were cultured in vitro and treated with different concentrations of ghrelin.Proliferation of 3T3-L1 preadipocytes was evaluated by MTT method and mRNA levels of c-myc and thymidine kinase were detected by RT-PCR.Morphological changes of 3T3-L1 preadipocytes were observed and cell differentiation was measured by oil red O staining.The mRNA levels of peroxisome proliferator-activated receptor γ (PPARγ) and CAAT/enhancer binding protein (C/EBPα) in the cells at different differentiation stages were detected by RT-PCR.The results showed that ghrelin at concentrations of 10-7 to 10-15 mol/L could significantly promote preadipocyte proliferation (P<0.05),with the most pronounced effect observed at 1011mol/L (P<0.01).Treatment of 3T3-L1 preadipocytes with ghrelin significantly in-creased the mRNA levels of c-myc and thymidine kinase (P<0.01).Morphological findings demonstrated that the great amount of lipid droplets appeared in the 3T3-L1 preadipocytes treated with ghrelin.Ghrelin could morphologically induce the differentiation of 3T3-L1 preadipocytes into mature adipocytes.Ghrelin significantly increased the mRNA levels of PPART and C/EBPα during the differentiation,when compared with control group (P<0.05).The mRNA levels of PPARγ and C/EBPα were obviously up-regulated with the differentiation of preadipocytes after the treatment of ghrelin.There were significant difference in the mRNA levels of PPARγ and C/EBPα on day 2 and day 8 of the differentiation of 3T3-L1 preadipocytes (P<0.01).In conclusion,ghrelin could promote the proliferation and differentiation of 3T3-L1 preadipocytes by increasing the mRNA levels of PPARγ and C/EBPα and therefore enhance the sensitivity of adipocytes against insulin.

Two-dimensional and real-time three dimensional echocardiography (RT3DE) was performed in 26 patients with atrial septal defect, and the echocardiographic data were compared with the surgical findings. Significant correlation was found between defect diameter by RT3DE and that measured during surgery (r=0.77, P<0.001). The defect area changed significantly during cardiac cycle. Percentage change in defect size during cardiac cycle ranged from 6%-70%. Our study showed that the size and morphology of atrial septal defect obtained with RT3DE correlate well with surgical findings. Therefore, RT3DE is a feasible and accurate non-invasive imaging tool for assessment of atrial septal size and dynamic changes.

Folliculogenesis is essential for production of female gametes in vertebrates. However, the molecular mechanisms underlying follicle development, particularly apoptosis regulation in ovary, remain elusive. Here, we generated sox3 knockout zebrafish lines using CRISPR/Cas9. sox3 knockout led to follicle development retardation and a reduced fecundity in females. Comparative analysis of transcriptome between sox3 and wild-type ovaries revealed that Sox3 was involved in pathways of ovarian steroidogenesis and apoptosis. Knockout of sox3 promoted follicle apoptosis and obvious apoptosis signals were detected in somatic cells of stages III and IV follicles of sox3 ovaries. Moreover, Sox3 can bind to and activate the promoter of cyp19a1a. Up-regulation of Cyp19a1a expression promoted 17β-estradiol synthesis, which inhibited apoptosis in follicle development. Thus, Sox3 functions as a regulator of Cyp19a1a expression, via 17β-E2 linking apoptosis suppression, which is implicated in improving female fecundity.
