OBJECTIVE:To observe clinical efficacy and safety of Zishen jianpi huoxue decoction combined with Xiaoxintong capsule in the treatment of type 2 diabetes complicating with silent myocardial ischemia (SMI). METHODS:126 patients with type 2 diabetes complicating with SMI were selected and randomly divided into control group(62 case)and observation group(64 cases). Both group were given diet guidance and hypoglycemic drugs as sulfonylureas,biguanides. Control group was given Xiaox-intong capsule 10 mg,tid;observation group was additionally given Zishen jianpi huoxue decoction 400-500 ml,bid. Both groups received treatment for 7 d. Therapeutic efficacy of thoracic obstruction,improvement of coronary artery branch stenosis and times and duration of electrocardiogram ST segment low voltage outlet were observed in 2 groups. ADR was compared between 2 groups. RESULTS:Therapeutic efficacy of thoracic obstruction and improvement of coronary artery branch stenosis in observation group were significantly better than in control group,with statistical significance(P<0.05);the times and duration of electrocardiogram ST segment low voltage outlet in observation group were significantly lesser shorter than in control group,with statistical signifi-cance (P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:Zishen jianpi huoxue decoction complicating with Xiaoxintong capsule shows significantly therapeutic efficacy in the treatment of type 2 diabetes complicating with SMI with good safety.

Cytokine-induced killer cells (CIK) is the fourth largest cancer treatment after surgery,chemotherapy and radiotherapy,and it is the development direction of cancer treatment.It is a new type of immune cell,and it is named after natural killer cell samples T lymphocytes as it express CD3 and CD56.Currently,CIK treatment has a broader range of clinical applications,and it has achieved the better clinical efficacy in the blood system cancer and solid tumors,The CIK adoptive immunotherapy is considered to be a new hope for the anticancer treatment.

Objective To establish a method of obtaining the highly purified smooth muscle cells (SMCs) isolated from rabbit bladder and then to identify them after in vitro culture using methods of morphology and immunofluorescence. Methods Primary cell culture of SMCs was set up by enzymic method in total of 6 adult male New Zealand-White rabbits. The serosa and mucosa of bladder were carefully removed by micro-surgical instruments under the naked eyes. The morphology characteristics of cells and their pattern of cell growth were observed using inverted microscope. The cells were identified using methods of H-E staining, transmission electron microscope and immunofluorescence staining of α-actin. The curves of cell growth and proliferation were drawn after cell counting. Results The capability of adherence was observed in these isolated SMCs after in vitro culture for 24 hours, and passage was then conducted after culture for 7 days. The cells kept growing well and fast from the 1~(st) to 10~(th) generation. But starting from the 12~(th) generation, the deformed cells were found and then become those without typical morphological feature of SMCs at the 15~(th) generation, and finally died at the 20~(th) generation. The isolated cells after in vitro culture at 4~(th) passage were identified to be SMCs successfully using the methods of H-E staining, transmission electron microscope and immunofluorescence. Conclusions The highly purified bladder SMCs of rabbit were successfully obtained after isolated by enzymic method and in vitro culture within a short time. That would provide us the basis for future studies of pathophysiology and molecular biology of bladder SMCs.

Object To develop a new method for the determination of curcumol in essential oil from rhizoma Curcumae L.. Methods The contents of curcumol were determined by high performance capillary gas chromatography with sequential increase of temperature on a HEWLETT PACKARD 5890A gas chromatograph. Results The method can be used to determine curcumol with accuracy at a recovery of 101.4% and RSD of 0.40%. Conclusion The present study provided a satisfactory method for the determination of curcumol, and it was found that its contents in four different species (C. wenyujin, C. longa, C. aeruginose, and C. kwangsiensis) were markedly different.

BACKGROUND:Diabetic lower limb ischemia is prone to involve distal lower limb arteries, and a conventional treatment is often unable to obtain the ideal effect. OBJECTIVE:To investigate the effect and safety of umbilical cord blood stem cel transplantation in the treatment of diabetic lower limb ischemia. METHODS: A diabetic rat model of lower limb ischemia was established, and along the femoral artery, five points
were selected for injection of human umbilical cord mesenchymal stem cel suspension, 20 μL per point. At 1, 2, 4 weeks after transplantation, transcutaneous oxygen pressure, vascular density and vascular endothelial growth factor level in the ischemic region, and incidence of adverse reactions were recorded. RESULTS AND CONCLUSION:At 1, 2 and 4 weeks after transplantation, the transcutaneous oxygen pressure, vascular density and vascular endothelial growth factor level in the ischemic region were found increasing, which were significantly different from those before transplantation (P < 0.05). At different time after transplantation, al animals had no inflammatory reactions such as skin bleeding and dermatitis, and local red, sweling, hot, pain, and had no tumor-like growth in organs. These findings indicate that umbilical cord blood stem cel transplantation can safely and significantly improve symptoms of diabetic lower limb ischemia, which has certain application feasibility. Obej ctive To investigate the role of metastasis associated protein 1(MTA1)in estrogen reg-ulated expression of matrix metalloproteinase -9(MMP-9)and tissue inhibitor of metalprotease -1(TIMP-1) in estrogen receptor( ER ) positive breast cancer cells .Methods MTA1 knockdown cell model was generated based on MCF-7breast cancer cell line by transfected with MTA 1-shRNA.The mRNA and protein level of MMP-9 and TIMP-1 in wild type MCF-7(MCF-7WT)and MCF-7MTA1-shRNA before and after 17β-estradiol ( E2) treatment were examined by Real -time PCR and Western blot respectively .Results The MTA1-shRNA showed maximally 84.9%suppression of MTA1 expression in MCF-7,suggesting a satisfied MTA 1 knockdown cell model was established for subsequent experiments .After treated with E2 for 48 h,MCF-7WT showed an incre-ment of 46%(P<0.05)and 37%(P<0.05)of the mRNA and protein level of MMP -9 and a decrement of 32.3%( P<0.05)and 18.2%(P<0.05)of TIMP-1;MCF-7MTA1-shRNA showed a decrement of 32.3%(P<0.05)and 18.2%(P<0.05)of mRNA and protein expression of MMP -9 respectively but no significant differ-ence in TIMP-1 comparing with MCF-7WT before treated with Estradiol.After E2 treatment,MCF-7MTA1-shRNA didn′t show significant change of MMP -9 except decrements of 32.3%(P<0.05)and 18.2%(P<0.05)in the mRNA and protein levels of TIMP -1.Conclusion MTA1 may be involved in the pathway by which estrogen regulated the expression of MMP -9 but not TIMP-1 in ER positive breast cancer cells .

Objective To discuss the relationship between carotid atherosclerosis and the ischemic cerebrovascular disease(ICVD).Methods 1583 subjects with acute ICVD enrolled from June 2001 to December 2004 underwent carotid artery duplex ultrasonography.Logistic regression was performed to analyze the relationship between carotid atherosclerosis and ICVD.Results 86.5%(1369/1583)of all the patients had a varying degree of carotid atherosclerosis,in which the occurrence of the atherosclerotic plaques(1286/1583,81.2%)higher than that of stenosis of the extracranial carotid(214/1583,13.5%).There was a significant difference in the incidence of carotid atherosclerosis between patients from the TIA group(198/317,62.5%)and from the cerebral infarction group(1087/1266,85.9%).Carotid atherosclerotic plaques were most commonly seen in the bifurcations of the common carotid artery(665/1286,51.7%).The severity of stenosis of the extracranial carotid and the formation of the atherosclerotic plaques were significantly correlated to the risk factors of cerebrovascular disease.Conclusions Atherosclerotic plaques are dominant characteristics of carotid atherosclerosis in the patients with ICVD in Foshan.There is a positive correlation between carotid atherosclerosis and ICVD.

Objective: To study the possible role and way of ALR in the immune regulation in vitro. Methods: The proliferation of spleen mononuclear cells of rat was detected with 3H-TdR method in vitro under the following different treatments:(1)the administration of the different concentration of rALR with 5 ?g/ml ConA at same time;(2)the addition of 30 ?g/ml ALR after 5 ?g/ml ConA pretreatment in 10 h and 32 h, respectively;(3)the addition of 5 ?g/ml ConA after 30 ?g/ml ALR pretreatment until 30 h. Cyclosporin A and the supernatant from cultures of yeast were used as positive and negative controls, respectively. The IL-2 levels in the supernatants from the mononuclear cells by various treatments were detected with RIA test kit. Results: rALR inhibited the proliferation and the production of IL-2 of the mononuclear cells from rat spleen stimulated by ConA dose-dependently. The mononuclear cell proliferations were still inhibited by 30 ?g/ml rALR after stimulated by ConA for 10 h or 32 h, but the pretreatment of 30 ?g/ml rALR for 30 h had no influence on the reactivity of mononuclear cell to ConA compared with control. Conclusion: rALR could inhibit in dose-dependent way the proliferation of mononuclear cells from rat spleen stimulated by ConA in vitro, but it couldn’t influence the rest mononuclear cells. This suggests that the rest mononuclear cells might not express the receptor of ALR.

OBJECTIVE: To establish a HPLC method for the determination of content of entecavir and its related substances. METHODS: The HPLC condition was consisted of a Luna C18 column with a column temperature at 25 ℃, using gradient eluate method, the mobile phase was acetonitrile,with a flow rate of 1.0 mL?min-1 and detection at 254 nm. RESULTS: The calibrated linear curve of entecavir was within 0.033 45～0.167 2 mg?mL-1(r=0.999 98). The average recovery was 100.09%,RSD=0.20%, The content of related substance was 0.54%～1.48%. CONCLUSION: This accurate and reliable HPLC method is applicable for the quality control of entecavir and its related substances.
