3.Protective effect of polypeptides from Chlamys farreri on murine thymocytes damaged by 60Co γ-rays

Objective To study the effect and mechanism of polypeptide from Chlamys farreri(PCF) on the thymocytes damaged by 60Co γ-rays. Methods The ceils were randomly divided into six groups: control group, 60Co groups (2 Gy, model),60Co + 0.5% PCF, 60Co+ 0.25% PCF, 60Co + 0.125% PCF, 60Co +0.1% VitC. The concentration of GSH-Px, ROS, A-SAC, T-AOC and the cells' viability were determined. The mitochondria membrane potential were tested. The expressions of P53, Bax and Bel-2 proteins were examined.Results The activities f GSH-Px, A-SAC, T-AOC in cells were enhanced, and the amounts of ROS were decreased by PCF. The expression of Bcl-2 gene was up-regulated, and down-regulated for the expression of P53 and Bax. All observed indexes of the PCF groups were significantly different compared with model group (P <0.05). Conclusions PCF has the protective effects on damages of thymocytes caused by 60Co irradiation. The mechanisms might be related to inhibiting of lymphocyte apoptosis and enhancing the activities of lymphocytes.
4.Minimally Invasive Coronary Bypass by Using Bilateral Internal Mammary Arteries Combine with Percutaneous Coronary Intervention for Multivessel Coronary Artery Disease:Case Report

[Summary] A 60-year-old man was admitted with main complaint of chest pain on exertion.Coronary angiogram showed severe triple vessel disease.He underwent the hybrid technique of treatment through two left minithoracotomy incisions.Both internal thoracic arteries were harvested under direct vision.The left internal mammary artery (LIMA)and right internal mammary artery (RIMA) compositing Y grafts was constructed. The LIMA was anastomosed to the left anterior descending branch and the RIMA was anastomosed to the ramus intermedius.The operation time was 306 min.The amount of bleeding during operation was 50 ml.Five days later he underwent percutaneous coronary intervention at right coronary artery lesion successfully.He was discharged from hospital 8 days after operation,and didn’t complain of chest pain during fellow-up for one month.This technique demonstrates to be an effective and minimally invasive alternative to treat patients with multivessel coronary artery disease.
5.Establishment of animal model with gastric cold syndrome.

This study is to establish the gastric cold model of rats. After gastric feeding with cold water for 5 weeks and extra iced water bath in the last 2 weeks, model group show distinct physical sign of gastric cold syndrome. The pathology of gastrics reveals gastricism of model group, while treatment group(treated with Fanzuojin Wan) show mild lesion. Elisa detection of model group show that the solution of interleukin-2 (IL-2) is higher than blank group. The difference with significance among model group, treatment group and blank group reveals the success of the establishment of gastric cold syndrome.

6.Clinical analysis of nervous system non-Hodgkin's lymphoma resembling to inflammatory diseases

Objective To characterize the manifestations of non-Hodgkin's lymphoma in nervous system with in-flammation-like presentation. Methods We reviewed clinical and laboratory data obtained from 3 cases of non-Hodgkin's lymphoma in nervous system with inflammation-like presentation.Those data include clinical manifestations,CSF examina-tions neuroimaging,pathology of biopsies,treatment and prognosis.Results The clinical manifestations of NHL in nervous system were variable and the findings of cerebrospinal fluid and imaging were not characteristic.Parital relief of symptoms by steroid cortisone could be achieved in some cases which maght further increased the difficulty in differentiating NHL from CNS inflammation.Several signs including no evidence of CNS inflammation,multiple organ involyements,especially the organ involvements outside CNS,and deterioration after a transient relief of symptoms by steroid cortisone,strongly suggest the possibility of NHL.Condusions We should increase physicians'awareness to NHL to reduce the misdiagnosis even though the final diagnosis relies on pathological examination.
7.Investigation of the molecular mechanism how genistein combined with 5-fluorouracil inhibits proliferation of human colon cancer cell line SW480

[Abstract ] Objective To investigate the molecular mechanism how genistein increased the sensitivity of colon cancer cell to 5-fluorouracil(FU)and promoted colon cancer cell apoptosis.Methods SW480 colon cancer cell line was chosen as experimental object.Genistein 80 μmol/L and 5-FU 30 μg/mL used separated or combined for 48 hours were set as drug experiment group.There were four experiment groups in this study:control group (without any drug),5-FU group,genistein group,5-FU and genistein combined group.The expression of survivin,Bcl-2,p21 ,caspase3 and caspase 9 in the tumor cells of each group at mRNA and protein level was deteced by real-time quantitative polymerase chain reaction (PCR) and Western blotting.The relative expression quantity of genes was determined by comparative threshold method (2 -ΔΔCT )andβ-actin was taken as an internal reference.Semi quantitative analysis was performed for protein relative expression quantity.The DNA combination activity of nuclear factor(NF)-κB of each group was measured by electrophoretic mobility shift assay (EMSA).Single factor analysis of variance was used for mean comparison among multiple groups and LSD test was for mean comparison between two groups.Results The expression of caspase3,caspase 9 and p21 of 5-FU and genistein combination group at mRNA (1 .903±0.122,2.726±0.050 and 2.541 ±0.393)and protein level (0.534±0.077,1 .161 ± 0.172 and 0.463±0.016)were all higher than those of control group (1 .001 ±0.052,1 .000±0.014 and 1 .001 ±0.037;0.080 ±0.043,0.248±0.059 and 0.139 ±0.021 ),genistein group (1 .559 ±0.038, 2.394±0.095 and 1 .686 ±0.061 ;0.335 ±0.052,0.478 ±0.059 and 0.304 ±0.018)and 5-FU group (1 .198±0.063,1 .051 ±0.043 and 1 .399±0.055 ;0.194±0.015 ,0.337 ±0.036 and 0.231 ±0.011 );the expression of survivin of 5-FU and genistein combined group at mRNA and protein level (0.165 ± 0.018 and 0.216±0.014)were all lower than those of control group,genistein group and 5-FU group (1 .001 ±0.033,0.775 ±0.044 and 0.395 ±0.030;0.594±0.079,0.375 ±0.014 and 0.295 ±0.014), and all the differences were statistically significant (gene,F = 802.865 ,52.760,39.992,187.288, 37.435 ;protein,F =10.466,44.483,19.490,200.011 ,45 .238;all P <0.01);the difference was also statistically significant in the expression of caspase3 and p21 of 5-FU group and genistein group compared with those of control group (LSD test,P <0.05 ).The results of EMSA assay showed that the DNA binding activity of NF-κB protein of genistein group (461.64 ±15.41 )and combined group (585.28 ±7.82) significantly decreased compared with that of control group (1 067.97 ±36.01)and 5-FU group (718.83± 23.18,LSD-test,P < 0.05).Conclusions Genistein combined with 5-FU seemed to have synergistic effects on apoptosis of colon cancer cell.The mechanism was that genistein inhibited the DNA binding activity of NF-κB mediated by genistein,further up-regulated caspase 3,caspase 9,p21 and down regulated survivin.Therefore,genistein may provide assistance in colon cancer chemotherapy.

Elisa detection of model group show that the solution of interleukin-2 (IL-2) is higher than the blank group. The obvious difference among model group, treatment group and blank group reveals the success of the establishment of gastric hot model.

9. Advances in modern research on anti-liver disease of Tibetan medicine Semen Hertospermi

Semen Hertospermi (Bolengguazi in Chinese, BL), as well-known Tibetan medicine, is one of commonly used drugs by Tibetan healers for the treatment of liver diseases and cholic diseases. Modern research indicated that BL had multiple pharmacological effects including hepatoprotection, anti-hepatitis B virus and anti-liver fibrosis, etc. And the lignans are the major pharmacodynamic substances. The fatty acid, polysaccharides and other ingredients also have hepatoprotective effects. This article mainly reviews the pharmacodynamic substances, pharmacological effects, preparation research and development related to anti-liver disease, so as to provide reference and thinking for the research and application of BL and preparations development in the prevention of liver disease and promote the modern research and development of ethnic medicine.
10. Spectrum-effect relationship of dryness of Atractylodis Rhizoma volatile oil based on grey relational grade and orthogonal partial least squares analysis

Objective To study the spectrum-effect relationship between GC-MS fingerprint of the volatile oil of Atractylodis Rhizoma and the dryness effect of Atractylodis Rhizoma, and clarify the main dryness components of Atractylodis Rhizoma. Methods The volatile oil of different batches of Atractylodis Rhizoma (S1-S9) were analyzed by GC-MS. Using the daily drinking amount of water, the viscosity of whole blood, and the content of aquaporin 2 (AQP2) in the kidney of rats as the indexes of dryness effect of Atractylodes Rhizome, the spectrum-effect relationship was analyzed by the combination of grey relational grade and orthogonal partial least square method. Results The dryness effect of Atractylodis Rhizoma was the combined action of multiple components, and the peaks of the major contribution to dryness effect were 31, 28, and 33, respectively. The corresponding components of the three peaks were β-eudesmol, (-)-aristolene, and bulnesol, which were identified by the spectral library retrieval analysis. Conclusion The main dryness effect components of Atractylodis Rhizoma are β-eudesmol, (-)-aristolene, and bulnesol, which provide a reference for investigating the material basis of dryness effect of Atractylodis Rhizoma.
