1.Inhibitory effect of laminarin (acidic polysaccharide) J201A on the proliferation of Human Lung Fibroblasts and its related mechanism of action
Chinese Journal of Marine Drugs 1994;0(03):-
To study the inhibitory effects of laminarin (acidic polysaccharide) J201A on the proliferation of human lung fibroblasts (HLF) and its related mechanism of action. Methods: Effect of J201A on the proliferation of HLF was evaluated by MTT assay, and the effects on cell cycle and synthesis of proteins of HLF were assessed by flow cytometry. The existence of J201A receptors on HLF was confirmed by fluorescent staining assay. Results:J201 A inhibited the synthesis of proteins and the proliferation of HLF at G0/G1 stage of cell cycle and J201A receptors existed on HLF. Conclusion:J201A exerted its antifibrotic activity by inhibiting HLF at G0/G1 stage of cell cycle and the synthesis of proteins.

Objective To investigate the impact on renal fibrosis by inhibition of connective tissue growth factor( CTGF) by RNA interference in spontaneous hypertension rat( SHR) . Method Twenty SHR were randomly (random number) divided into SHR group ( n = 10) and RNAi group ( n = 10), eight Wistar-Kyoto rats were set as control. At the end of RNA interference procedure, all the rats were sacrificed and the kidneys were harvested. The mRNA and plasmosin of CTGF and fibronectin(FN) of renal tissue were extracted and measured by RT-PCR and Western Blotting. And the localization of CTGF and FN were analyzed with immunohistochernistry technique. The collagen deposition(shown as collagen volume traction, CVF) were evaluated with 0.1% sirius-picric staining, and the hydroxyproline of myocardium were detected by colorimetry. Results The mRNA and protein expression of CTGF decreased 66% and 62% in RNAi group (P < 0.01). The mRNA and protein expression of FN decreased 56% and 51% in RNAi group. The same inhibition effect was observed by hislological analysis. Immuno-histochemistry showed that CTGF localized both in renal parenchyma and renal interstitium, whereas FN majorly expressed in renal interstitium. Observation with light microscope showed that collagen deposition(CVF)decreased sharply in RNAi group versus SHR group. And the same effect was viewed in hydroxypnoline assay[SHR group: (0.596 ± 0.067) μg/mg, RNAi group: (0.368±0.084) μg/mg, P < 0.01 ] .Further study by polarized microscope displayed that RNA interference mainly suppressed type I collagen synthesis. Conclusions Targeted inhibition of CTGF by RNA interference leads significant decrease of extracellular matrix deposition in kidney. And the anti-fibrotic effect independent of lower the blood pressure. This study indicated CTGF take a key role in the development and progress of renal fibrosis.

BACKGROUND:Previous studies have shown that there are some significantly gender-related differences in coronary heart disease between women and men.It is not clear whether the outcomes have affected by gender.OBJECTIVE:To analyze the outcomes of follow-up after coronary stenting, and to investigate the sex diffefence.DESIGN,TIME AND SETTING:The prospective follow-up was performed at the Wuhan Asia Heart Hospital from January 2004 to December 2005.PARTICIPANTS:267 patients who suffered from coronary artery disease and treated with percutaneous coronary intervention were involved in the present study and followed up for 3 years, including 52 females and 215 males,averagely(59.4±7.5)years old.Imfarction in 3 levels were considered to be scccessful.Follow-up in out-patient clinic was conducted regularly.Six monks after implantation, telephone follow-up was done;questionnaire follow-up was performed every year.Average follow-up time was(20±7)months.MAIN OUTCOME MEASURES:Baseline clinical and angiographic characteristics,follow-up outcomes after 3 years of implantation after coronary stenting were compared between 52 females and 215 males.lower significantly than those in men.During the 3-year follow-up,no significant differences were observed between women and men regarding death, major adverse cardiovascular events, and recurrent typical ischemia. The proportion of symptom remission, life quality improvement was similar between women and men.In multivariable analysis, sex was not anindependent predicator for long-term outcomes of coronary stenting(OR 0.819,95% CI 0.41 to 1.79).CONCLUSION:Women with coronary artery disease are older than men and have more comorbidities.Long-term outcomes have improved in women after coronary stenting.Therapeutic outcome is not correlated to sex.

OBJECTIVETo explore the genetic and clinical characteristics of isodicentric Ph chromosomes [idic(Ph)] in lymphoid blast crisis of chronic myeloid leukemia (CML-BLC).
METHODSBone marrow aspirates of 2 patients with CML-BLC were analyzed by R banding after 24 hours of culturing. Genomic copy number variations (CNV) were analyzed by single nucleotide polymorphism array (SNP array) in case 1. The results were confirmed with fluorescence in situ hybridization (FISH). Variations of acute lymphoblastic leukemia-related genes including CDKN2A/AB and PAX5 were detected by multiplex ligation-dependent probe amplication (MLPA).
RESULTSDeletions and duplications on derivative chromosome 9 detected by FISH were confirmed by SNP array analysis. The distances between the BCR/ABL fusion signals on the idic(Ph) chromosomes in the two patients have differed greatly. The idic(Ph) in the second patient was supposed to be formed by two Ph chromosomes joined at their q terminals, where as the idic(Ph) in the first patient have been shown to be fused at the satellite regions of their p arms.
CONCLUSIONThe idic(Ph) chromosomes presented in CML-BLC may predict resistance to Imatinib and response to Dasatinib.
