OBJECTIVE:To investigate the effects of pravastatin on serum levels of CRP and TNF-? in patients with diabetic nephropathy and study its possible mechanism in the treatment of diabetic nephropathy. METHODS:28 patients with diabetic nephropathy were treated with provastatin plus routine therapy (trial group),another 28 patients with diabetic nephropathy were treated with routine therapy alone (control group). Serum levels of CRP and TNF-? in the two groups were detected after treatment for 24 weeks. RESULTS:The total effect rate in the trial group was 67.9% versus 42.9% in the control group(P

4.HPLC Fingerprints of Chaihu Shugan Pills

Objective:To establish the high performance liquid chromatography-fingerprints( HPLC-FPS)of Chaihu Shugan pills. Methods:The analysis was performed on a Zirchrom Kromasil C18 column with the mobile phase of methanol-water(containing 0. 05%phosphoric acid)with gradient elution at a flow-rate of 0. 8 ml·min-1 . The detection wavelength was set at 294 nm and the column temperature was 30℃. Results:With baicalin as the reference substance,there were 24 common peaks in the fingerprints to establish the fingerprint common pattern. The similarity among the 16 batches of samples was calculated,and the results showed it was within the range of 0. 690-0. 993,suggesting the differences among the samples from different factories. Conclusion:The establishment of the fingerprints provides a simple and practical analysis method for more comprehensive regulation of medicinal material sources,effective quality monitoring of the products and the assurance of effective and safe clinical application.
5.Kaempferol inhibits cell proliferation, invasion and migration via Wnt/β-catenin signaling in HBx-HepG2 cells

AIM: To explore the effects of kaempferol on the proliferation, invasion and migration abilities of HBx-HepG2 cells and to examine the underlying molecular mechanisms.METHODS: The expression levels of related genes at mRNA and protein levels were determined by RT-qPCR and Western blot.The cell apoptotic rate was analyzed by flow cytometry.The cell proliferation, growth, invasion and migration abilities were measured by MTT assay, colony formation assay, Transwell invasion assay and wound healing assay, respectively.RESULTS: Kaemferol inhibited HBx-HepG2 cell proliferation in a concentration-and time-dependent manner.Kaempferol at 100 μmol/L significantly inhibited the colony formation, invasion and migration abilities of the HBx-HepG2 cells.Kaemferol at 100 μmol/L also increased cell apoptotic rate, increased the protein levels of cleaved caspase-3, cleaved caspase-9 and Bax, and decreased the expression level of Bcl-2.In addition, kaemferol at 100 μmol/L suppressed the mRNA and protein expression levels of β-catenin, c-Myc and cyclin D1 in the HBx-HepG2 cells.Kaemferol at 100 μmol/L also suppressed the protein level of p-GSK-3β and the β-catenin protein levels in both cytoplasm and nucleus.LiCl treatment reversed the inhibitory effect of kaempferol on the growth, invasion and migration of the HBx-HepG2 cells.CONCLUSION: Kaempferol inhibits cell proliferation, invasion and migration via activating Wnt/β-catenin signaling in HBx-HepG2 cells.
6.The correlation of distal latency and esophageal motility under esophageal high resolution manometry in gastroesophageal reflux disease

Objective To investigate the relationship between esophageal motility and distal latency (DL) in gastroesophageal reflux disease (GERD) using high resolution manometry (HRM).Methods A total of 51 GERD patients underwent HRM and 24 h-esophageal pH monitoring.According to the HRM topography (characterized as either break peristalsis or normal esophageal movement),all GERD patients were divided into two groups:hypomotility group (n =28) and normal group (n =23).Fourteen non-GERD controls were enrolled.The monitoring results were analyzed.Results The HRM DL of 28 esophageal hypomotility patients(54.9%,28/51) were the longest (7.27 ± 1.44) s.Patients with normal peristalsis also had longer latency (6.70 ± 1.41)s than the non-GERD controls (5.86 ± 0.96)s.All the differences were statistically significant (P < 0.01).DCI of hypotensive peristalsis patients (712.49 ± 703.10) mmHg · s · cm was lower compared with the other groups [(1 285.85 ± 850.83) mmHg · s · cm,(1 109.74 ± 611.70) mmHg · s · cm] (P <0.O1).Other indicators such as LES pressure,CFV and IBP showed no significant differences among groups (P > 0.05).Conclusion Esophageal manometry of GERD patients indicates that esophageal hypomotility is accompanied with prolonged DL.Because DL of all GERD sufferers are extended,esophageal dysmotility has great implications for GERD's development.
8.The characteristics of the gastroesophageal junction in gastroesophageal reflux disease under high-resolution esophageal manometry

Objective To analyze the characteristics of gastroesophageal junction(GEJ) in gastroesophageal reflux disease (GERD) using high resolution manometry (HRM),and the features in different GERD subtypes,reflux esophagitis (RE) and non-erosive gastrooesophageal reflux disease (NERD).Methods Sixty GERD patients were divided into two subtypes,RE (n =33) and NERD(n =27),based on the results of endoscopy.All patients received HRM,and were divided into three subtypes,type Ⅰ,type Ⅱ and type Ⅲ according to the GEJ pressure morphology.HRM results among different subtypes were investigated.Results In 60 GERD patients,the GEJ was divided into three subtypes,including 26 cases of type Ⅰ (RE =14,NERD =12),29 cases of type Ⅱ (RE =14,NERD =15) and 5 cases of type Ⅲ (RE =5).The characteristics of GEJ morphologic subtypes were analyzed.The total length of lower esophageal sphincter(LES) of three GEJ subtypes(type Ⅰ,type Ⅱ and type Ⅲ) were (3.35 ± 0.56) cm,(3.29 ± 0.87) cm and(2.43 ± 0.22)cm,respectively.The LES intra-abdominal length of three GEJ subtypes were(2.41 ± 0.68) cm,(3.07 ± 4.59) cm and (0.00 ± 0.00) cm,respectively.The LES rest pressure of three GEJ subtypes were (22.00 ± 8.00) mmHg,(16.46 ± 8.60) mmHg and (11.07 ± 5.80) mmHg,respectively.The total length of LES were significantly different among three subtypes (P < 0.05).The LES rest pressure were not significantly different among three subtypes (P > 0.05).Among type Ⅰ and type Ⅱ patients,there were no significant difference in LES-CD distance,total length of LES,LES intra-abdominal length and LES rest pressure between RE group and NERD group.Conclusion HRM can clearly distinguish the GEJ subtypes.To some extent,esophageal motility characteristics of the patients with different subtypes GEJ is various.However,it still needs further studies with larger sample size to confirm.
9.Effects of Acupuncture Combined with Massage on Shoulder Subluxation after Hemiplegia

ObjectiveTo observe the effects of acupuncture combined with massage on shoulder subluxation and motor function recovery of the upper extremities in patients with hemiplegia after stroke.Methods60 hemiplegic patients were randomly divided into two groups for treatment with acupuncture cooperates with massage or usual rehabilition, respectively. The recovery of the patient's shoulder subluxation and movement function of upper extremities were evaluated 2 months after treatment. ResultsThe shoulder subluxation and movement function of the upper extremities were improved after treatment with both therapies (P<0.01), and acupuncture cooperates with massage showed better effect (P<0.01). ConclusionAcupuncture cooperates with massage can facilitate the recovery of shoulder subluxation and motor function of the upper extremities after hemiplegia.
10.Application of rapid immunohistochemical staining technique in intraoperative frozen section diagnosis of thyroid neoplasm

ObjectiveTo investigate the application of rapid immunohistochemical staining technique in intraoperative frozen section diagnosis of thyroid neoplasm.Methods MaxVision one-step rapid immunohistochemical staining technique was used to detect the expression of CK19,HBME-1,and Gal-3 in frozen section of papillary thyroid carcinoma(PTC)andthyroid benign lesions.MaxVision conventional immunohistochemistry of frozen remaining tissue was served as control.ResultsMaxVision one-step rapid immunohistochemical staining technique could be completed in 20 minutes.The positive localizations of three markers detected by rapid immunohistochemistry were similar to conventional immunohistochemistry, in general.The expression of CK19 was located in cytoplasm and cellular membrane.Gal-3 and HBME-1 were mainly detected in follicular luminal border and/or surface of papilla. The staining intensity in rapid immunohistochemistry was stronger than that in conventional immunohistochemistry. The positive rates of CK19,HBME-1,and Gal-3 by rapid immunohistochemistry in frozen sections were: 0 (0/28),10.7 % (3/28),0 (0/28),respectively,for benign lesions (nodular goiter,Hashimoto thyroiditis,thyroid adenoma); and 94.9 %(37/39),92.3 % (36/39),92.3 % (36/39),respectively,for PTC.The expression of three markers between thyroid benign lesions and PTC had a significant difference (x2 =59.326,55.861,44.605,all P < 0.001).In benign lesions,the rate of same case with two and more positive markers was 0,while in PTC it was 100 % and significantly different (x2 =67.000,P < 0.05).ConclusionMaxVision one-step rapid immunohistochemical staining technique could be applied in intraoperative frozen section diagnosis.Detecting CK19,HBME-1,and Gal-3 expression in intraoperative frozen section has an auxiliary value for diagnosis of PTC.
