1.Preliminary study on the prediction of chemotherapeutic efficacy in patients with advanced digestive cancer with CD+3CD28+T cell value

Objective To investigate the relationship between CD+3CD28+T cell value and chemotherapeutic efficacy in patients with advanced digestive cancer. Methods 38 digestive cancer patients with distant metastatic lesion were included.10 healthy persons were as control group. The absolute counts of CD+3CD28+T cell were measured with flow eytometry in these patients before chemotherapy. All patients received chemotherapy, gastric cancer: DCF regimen (docetaxel plus tegafur and cisplatin, one cycle), colorectal cancer:. Folfox regimen (L-OHP plus tegafur, two cycles), esophageal cancer: mitomycin plus tegafur and cisplatin (two cycles), gallbladder or pancreatic cancer: gemzar plus tegafur and cisplatin (one cycle). According to WHO standard, the chemotherapeutic efficacy was determined, 38 patients were divided into three groups: CR+PR (complete remission plus partial remission), SD (stable of disease) and PD (progress of disease) group. Statistical analysis was used upon all data. Results There was no significant difference in value of CD+3CD28+ T cell between CR+PR group and control group (P=0.549). The value of CD+3CD28+ T cell in SD group and PD group were significantly decreased (P=0.002, P=0.000). The value of CD+3CD28+T cell in CR+PR group was more than that in PD group (P =0.001), while in SD group was more than in PD group (P = 0.044). Conclusion The efficacy is poor, if the value of CD+3CD28+ T cell in patients with advanced digestive cancer is less than that of in healthy persons in peripheral blood. Therefore, the value of CD+3CD28+ T cell is important for the early prediction of the efficacy before chemotherapy.
2.Effect of TGF-? on the expression of type Ⅰ inositol 1,4,5-triphosphate receptor in WB rat liver epithelial cells

Objective:To investigate the effect of TGF-? on the expression of type Ⅰ inositol 1,4,5-triphosphate receptor in WB rat liver epithelial cells.Methods:The expression of type Ⅰ inositol 1,4,5-triphosphate receptor protein and mRNA were detected by Western blot and RT-PCR in WB rat liver epithelial cells in vitro stimulating with TGF-?.Results:The expression level of type Ⅰ inositol 1,4,5-triphosphate receptor protein and mRNA in WB rat liver epithelial cells were both increased after stimulating with TGF-? in several time points and reached the highest at 8 h stimulated, and then decreased.Conclusion:TGF-? enhance the IP_3R1 protein and mRNA expression in WB rat liver epithelial cells.

The conformity between bone implant and bone tissue plays an important role in osseointegration.Protein molecules such as extracellular matrix proteins,cytoskeletal proteins,and attachment proteins,which produced by osteoblasts,promote the attachment of osteoblasts to bone implant and the surrounding ceils.In vitro studies show that nano-plated implant material bears excellent biocompatibility and superior physical and chemical features.Nano-scale material has the advantage of high surface roughness,wettability and electrochemistry features which erdaance the adherence and interaction of extracellular matrix proteins and promotes the attachmerit,proliferation and differentiation of osteoblasts.The expression of cytokines,such as prostaglandin E2(PGE2)and transforming growth factor-β(TGF-β)are also increased in nano-structured bone implants which in turn promote the new bone formation.Further studies need to be done to explain the mechanism of the effect of nano-tructured bone implant on osteoblasts.
4.Treatment of Recurrent Oral Ulceration (Yin Deficiency Fire Excess Type) by Qianjin Kouchuang Jiawei Granule: a Clinical Observation.

OBJECTIVETo observe the curative effect of Qianjin Kouchuang Jiawei Granule (QKJG) on recurrent oral ulceration [yin deficiency fire excess type (YDFET)].
METHODSTotally 120 patients who suffered from recurrent oral ulceration (YDFET) were randomly assigned to two groups, the experiment group and the control group, 60 in each group. Patients in the experiment group took QKJG, 20 g each time, twice per day, while those in the control group took Kouyanqing Granule (KG) , 20 g each time, twice per day. Fourteen days consisted of one therapeutic course, two for all. Scores for patients' symptoms and signs (ulcer area, exudation, hyperaemia, edema, the number of ulceration, burning sensation, and pain degrees) were assessed before treatment, at day 3 and 7 after treatment. Short-term efficacy was assessed by visual analogue scale (VAS). The total paralysis time and the total number of ulceration at month 12 after treatment were taken as judgment for long-term efficacy. Results Compared with before treatment in the same group, symptoms and signs were obviously improved at day 3 and 7 after treatment in the two groups (P < 0.05). Compared with the control group at day 3 after treatment, the improvement of edema, exudation, pain degree, and burning sensation was more obvious in the experiment group (P < 0.05, P < 0.01). The improvement of edema, pain degree, and burning sensation at day 7 after treatment was more obvious in the experiment group than in the control group (P < 0.05). As for short-term efficacy, the total effective rate was 86.67% (52/60 cases) in the experiment group and 83.33% (50/60 cases) in the control group, with no statistical difference between the two groups (P > 0.05). As for long-term efficacy, the total effective rate was 90.00% (54/60 cases) in the experiment group, significantly higher than that of the control group with statistical difference [81.67% (49/60 cases), P < 0.05]. At month 12 after treatment, the total number of ulceration was reduced and the paralysis time of ulcer attack prolonged in the experiment group, with statistical difference when compared with the control group (P < 0.05).
CONCLUSIONQKJG showed better long-term efficacy than that of KG in treating recurrent oral ulceration (YDFET).

5.Investigation on reference interval of serum non-esterified fatty acid among population in Wuhan area

Objective To preliminarily establish the reference interval of serum non‐esterified fatty acids(NEFA)among popula‐tion in Wuhan area by enzymatic assay .Methods NEFA level of serum samples from A total of 1 250 individuals undergoing healthy physical examination in our hospital were selected as the research subjects .The Olympus AU5400 biochemistry analyzer and SEKISUI reagents were adopted to detect serum NEFA level .The regression analysis was adopted to analyze the NEFA influencing factors .The NEFA 95% reference interval was estimated by using the normal distribution method and the reference interval was verified .Results The regression analysis showed that the body mass index (BMI)and age had significant effect on NEFA (P<0 .01) .NEFA was highest in the group aged ≥60 years old ,followed by the group of 18- <45 years old ,and lowest in the group of 45-60 years old ;NEFA was highest in the group of BMI<18 .5 kg/m2 .The 95% reference interval of NEFA for healthy popula‐tion in Wuhan area was estimated as 204 .6 -975 .2 μmol/L .The detected NEFA level for clinical verifiers was 242 .0 -831 .2μmol/L ,which conformed to the above reference interval .Conclusion The serum NEFA reference interval among healthy popula‐tion in Wuhan area is primarily established .
6.Studies of TLC and determination of three components in Wenweishu Tablets

AIM: To establish a simple and rapid method for the quality control of Chinese traditional medicine. METHODS: Pericarpium Citri Reticulatae,Fructus Psoraleae,Rhizoma Atractylodis macrocephalae and Radix Codonopsis in Wenweishu Tablets were identified by TLC at two thin-layer chromatographic plates.The contents for hesperidin,psoralen and isopsoralen in this preparation were determined simultaneously by HPLC. RESULTS: The methodological study showd that a good linear correlation existed in the range of 0.039 6-0.495 ?g(r=(0.999 99)) of hesperidin,0.018 5-0.231 ?g(r=0.999 98) of psoralen and 0.019 7-0.246 2 ?g(r=0.999 91) of isopsoralen applied respectively.The average recovery of hesperidin was 98.42%(n=9),RSD was 2.69%;The average recovery of psoralen was 98.29%(n=9),RSD was 1.08%.The average recovery of isopsoralen was(98.93%)(n=9),RSD was 2.41%. CONCLUSION: The method is simple、rapid and practicle.
7.Study on the relationship between the efficacy of IFN and ribavirin on chronic hepatitis C and the anti-IFN antibodies.

Objective In order to study the relationship between the efficacy of IFN and ribavirin in treating the patients with chronic hepatitis C and the anti-IFN antibodies.Methods 20 patients received 3MU rIFN ?-2b three times a week and ribavirim 1?050mg once a day for 24 weeks;32 patients were treated with 3MU rIFN ?-2b and without ribavin.These patients were all followed up 24weeks after stopping treatment,meanwhile the anti-IFN antibodies were detected.Results In first group,50% of the patients had complete response,which was higher obviously than 18.7% in the second group;25%relapse developed in the first group,which was lower than 56.3% in the second group.25% of the patients had no response in each groups.The relapse was related to the anti-IFN antibodies,especially neutralizing antibodies.Conclusion The efficacy of combined treatment with IFN?-2b and ribavirin is superior to the treatment with only rIFN?-2b.The relapse is related to anti-IFN antibodies,especially neutralizing antibodies.
9.Degeneration of Dendritic Cells from Human Peripheral Blood and Acquirement Antigen from Apoptotic Hepatochlangioma Cells

Objective:To eastablish the efficient presentation of antigen from apoptotic cells by human DC from peripheral blood. Methods: using recombinant human granulocyte/macrophage colonystimulating factor(GM- CSF) and interleukin 4 (IL- 4 ) we have established dendritic cells (DC)from peripheral blood monocyte that maintain the antigen capturing and processing capacity characteristic of immature dendritic cells in vivo. GM - CSF 50ng/ml , IL- 41 000ng/ml once two days(total four). on the 3 rd day of culture, immature DC and apoptotic hepatochlangioma cells were in coculture lasting 7 days. Results:these cells had typical dendritic morphology, express high levels of CD1a ,B7 and acquired antigen from apoptotic cells and induced an increased T cell stimulatory capacity in MLR. Conclusions:we have established DC from blood mononuclear tells using GM- CSF and IL- 4 and DC can be efficiently drived from apoptotic cells and can induce the increase of T cells obviously. It probably becomes an effective approach of antigen transduced with DC.
10.Sensitized dendritic cells with apoptotic hepatoma cells induced by mitomycin

BACKGROUND: Dendritic cells play an important role in antigen present in vivo, and the mechanism of tumor cells in escaping the antigen presentation of dendritic cells existed in the patients with tumor.OBJECTIVE: To sensitize dendritic cells from human peripheral blood with apoptotic hepatoma cells induced by mitomycin.DESIGN: A randomized control trial by taking apoptotic hepatoma cell sensitized dendritic cells as the observed subjects.SETTINGS: Institute of Field Surgery, Daping Hospital, the Third Military Medical University of Chinese PLA; Institute of Radiation Medicine,Chinese PLA Academy of Military Medical Sciences.MATERIALS: The experiments were carried out in the Institute of Radiation Medicine, Chinese PLA Academy of Military Medical Sciences from April 1998 to May 1999. The cell strain was the QBC939 bile duct cancer cell strain, and mitomycin was used as the antitumor drug.METHODS: Mononuclear cells were isolated from the peripheral blood of normal people, 50μg/L granulocyte-macrophage colony stimulating factor(GM-CSF) and 1 000 U/mL interleukin-4 (IL-4) were added, once every other day for 4 times. On the 3rd day of culture, the apoptotic bile duct cancer cells induced by mitomycin was added, and then cultured in vitro for 4 days, finally the dendritic cells were collected.MAIN OUTCOME MEASURES: ① The identification of the cultured dendritic cells was observed; ② The dendritic cells were co-cultured with necrotic and normally cultured bile duct cancer cells respectively, and the phagocytized apoptotic body loaded antigens were observed; ③ The immunostimulatory activity of dendritic cells (1×103, 5×103 and 1×104/well)and that after loaded by antigen were detected, and the mononuclear cells were taken as controls.RESULTS: ① The cultured and amplified dendritic cells expressed high levels of costimulatory molecules of CD1a and B7, and there were typical irregular processes on the surface. ② The tumor cells formed apoptotic bodies when they were induced by mitomycin, which were arrested and phagocytized by dendritic cells. ③ The ability of the antigen loaded dendritic cells in stimulating the proliferation of allogenic lymphocyte T was further enhanced.CONCLUSION: The apoptotic tumor cells induced by mitomycin can induce the mononuclear cells from human peripheral blood differentiating into the dendritic cells with the concomitance of GM-CSF and recombinant IL-4 and amplify dendritic cells. Meanwhile, the dendritic cells can effectively present the antigens of apoptotic bile duct cancer cells, and it probably becomes a new effective approach for tumor antigen to sensitize dendritic cells.
