2.Value of red blood cell volume distribution width for predicting prognosis in patients with peritoneal dialysis-associated peritonitis

Objective To explore the evaluation significance of the red blood cell volume distribution width(RDW) for the prog‐nosis in the patients with peritoneal dialysis‐associated peritonitis .Methods The patients receiving the continuous ambulatory peri‐toneal dialysis(CAPD) and developing peritoneal dialysis‐associated peritonitis were prospectively collected in our hospital from Oc‐tober 2012 to October 2014 and served as the research subjects for conducting 18‐month followed up ,as well as their clinical data and laboratory detection data .Results There were 59 research subjects were enrolled ,the average dialysis time was (14 .98 ± 3 .6) months and the number of re‐peritonitis was (1 .4 ± 0 .5) times .Among them ,54 cases were survival and 5 cases died ,the dialysis time ,cardiovascular events ,RDW‐CV levels in the death group were higher .Cox regression analysis obtained that the cardiovascular events (HR=0 .765 ,95% CI:0 .107-0 .388 ,P=0 .072) and RDW level (HR=0 .681 ,95% CI:0 .518-0 .985 ,P=0 .058) were not the risk factors affecting patients′death ,while the dialysis time(HR=1 .03 ,95% CI:0 .788 -1 .857 ,P=0 .023) was related with the prognosis in the peritonitis patients .Conclusion The microinflammation status exists in the CAPD patients ,RDW may be in‐volved in the inflammatory process ,but it is not an independent risk factor affecting the prognosis of the patients with peritoneal di‐alysis‐associated peritonitis ,which needs more studies .
3.Relationship between tumor angiogenesis and invasion, metastasis and prognosis of hepatocellular carcinoma

Objective To investigate the relationship between the tumor angiogenesis and the clinicopathological variables of hepatocellular carcinoma (HCC) and determine whether tumor angiogenesis can be used as an indicator for predicting invasion, metastasis and prognosis of HCC. Methods Using the immunohistochemical S-P method, CD34 expression was detected in liver tissue from 20 normal individuals, 20 patients with liver cirrhosis and 85 patients with HCC and juxtacancerous tissues and the microvascular density (MVD) determined in those from 85 patients with HCC. Results CD34 was mainly located in the cell cytoplasm of endothelial cells. The CD34 staining was confined to vessels in the portal triad in the above-mentioned various tissues. In HCC tissues, however, sinusoid-like vessels reacted intensively with anti-CD34 and the MVD was 156.5±62.4 (per 0.74 mm2). MVD was correlated to tumor size, number, differentiation, emboli in the portal vein and capsule (P<0.05 or 0.01) but not to HBsAg and AFP (P>0.05). It was also correlated with prognosis of HCC. The mean tumor-free survival of MVD ≥156 was 14 months while that of MVD <156 was 53 months (P<0.01). Conclusion The expression of CD34 in sinusoid-like vessels in HCC tissues represents the neovascularization. Angiogenesis in HCC is associated with the progression of HCC. MVD can be used as an indicator to judge the invasion, metastasis and prognosis of HCC.
4.Epidemiological features and disease spectrum of hepatitis C in Qinghai Province,China:analysis of 1 10 cases

Objective To investigate the epidemiological features and disease spectrum of inpatients with hepatitis C in recent years and to provide reference for the control of hepatitis C.Methods A retrospective study of demography,epidemiology,and laboratory examination was performed in 1 10 hospitalized patients with hepatitis C.Results Of the 1 10 patients,60 were male and 50 were female,with a male-to-female ratio of 1.2:1.The main transmission routes included blood product transfusion (33.6%),surgery (30.0%),intravenous drug use (14.5%),and sexual transmission (4.5%).The other 19 cases (17.4%)were of unknown transmission route.The annual numbers of cases from 2008 to 2011 were 16,18,32,and 44,respectively.The genotype proportions in patients were as follows:1b,60.7%(17/28);2a,28.6%(8/28);1b/2a mixed type,7.1%(2/28);3b,3.6%(1/28).Of all patients,13.6%(15/110)were found with a-cute hepatitis C,67.3% (74/110)with chronic hepatitis C,19.1% (21/110)with liver cirrhosis,and 10.9% (12/110)with decom-pensated cirrhosis.Conclusion The incidence of HCV infection has been increasing year by year.The major HCV genotypes are 1 b and 2a.Hepatitis C is apt to develop into liver cirrhosis.It is necessary to reinforce monitoring and to carry out specific epidemiological study a-mong the target population.
5.Expression of RCAS1 in Different Tumor Cell Lines

Objective To investigate the expression of RCAS1,which is a receptor-binding cancer antigen expressed on SiSo cells in many tumor cell lines. Methods Using real-time polymerase chain reaction(PCR),Northern blot,immunohistochemical examination and enzyme-linked immunosorbent assay,we detected the expre-(ssion) of RCAS1 in 10 different tumor cell lines and two normal cell lines. Results The expression of RCAS1 in the tumor cell lines and the fetal renal cell line(293 cell) was detected by real-time PCR and Northern blot,but not in the normal liver cell line(LO2 cell).The results of immunohistochemical examination on the tumor cell lines and the 293 cell showed that RCAS1 existed in cytoplasma and on cell surface.Except HuH-7 cell and LO2 cell,the soluble RCAS1 protein in supernatants of the other cells was significantly more than that in DMEM(P
6.The effects of electroacupuncture combined with transcranial magnetic stimulation on VEGF164 mRNA and CD31 expression in rats with cerebral ischemia

Objective To study the effect of electroacupuncture(EA) combined with transcranial magnetic stimulation(TMS) on the angiogenesis in rats with cerebral ischemia. Methods Fifty-five male Wistar rats were randomly divided into five groups: a normal group, a model group, an EA group, a TMS group and an EA plus TMS group. After the establishment of acute middle cerebral artery occlusion (MCAO), the rats in various groups were accordingly treated with EA, TMS or EA plus TMS, respectively. Then the expression of VEGF164 mRNA and CD31 was observed. Results The expression of VEGF164 mRNA and CD31 around the infarction locus of the three treatment groups increased, especially in the EA plus TMS group. Conclusion EA plus TMS can increase the expression of VEGF164 mRNA and CD31 around the infarction locus and promote the non-molecular angiogenesis in rats with cerebral ischemia.
7.Ouabain and asthenospermia.

Asthenospermia accounts for about 30% of the causes of male infertility. Currently, most drugs for asthenospermia lack specificity and desirable therapeutic efficiency. An insight into the pathogenesis of asthenospermia is important for the development of specific therapies for this disease. The protein Na+/K(+)- ATPase α4 isoform (NKA4) presents in both mature testis tissue and the sperm tail, the absence or reduced activity of which may significantly decrease sperm motility. Ouabain is a natural inhibitor of NKA4, suppressing its activity by specifically binding the ouabain site in it. The hypothalamus and adrenal cortex excrete an ouabain-like steroid hormone called endogenous ouabain (EO), which may be associated with the pathogenesis of asthenospermia by inhibiting the activity of NKA4, affecting Na+/H+ exchange, Na+/Ca2+ exchange and sperm cell membrane potential, and eventually reducing sperm motility.

8.Effects of proprioception training on the motor function and ability in the activities of daily living of hemiplegic stroke patients

Objective To study the effects of proprioception training on the motor function and ability in the activities of daily living (ADL) of hemiplegic stroke patients. Methods Forty hemiplegic stroke patients were divided into study group and a control group ( n =20 in each).The two groups received conventional rehabilitation treatment and pharmacotherapy,but the study group received additional proprioception training.The Technobody proprioceptive system assessment,the Fugl-Meyer lower limb assessment (FMA),the Berg balance scale (BBS) and the modified Barthel index (MBI) were used to evaluate their proprioceptive ability,lower limb motor function,balance and ADL ability,before and after 4 weeks of treatment. Results After 4 weeks,proprioception,lower limb motor function,balance and ADL ability had all improved significantly in the experimental group.Lower limb motor function,balance and ADL ability had also improved significantly in the control group,but they showed no significant change in their average proprioceptive ability.The improvement in the experimental group was superior to that of the control group in terms of proprioceptive ability,balance and ADL ability. Conclusions Proprioception training can improve the motor function (including the proprioception and balance) and ADL ability of hemiplegic stroke patients.
9.Expression and Purification of Enterovirus Type 71 Polyprotein P1 using Pichia pastoris system

Enterovirus type 71(EV71) causes severe hand-foot-and-mouth disease(HFMD) resulting in hundreds of deaths of children every year; However,currently,there is no effective treatment for EV71.In this study,the EV71 poly-protein(EV71-P1 protein) gene was processed and cloned into the eukaryotic expression vector pPIC9k and then expressed in Pichia pastoris strain GS115.The EV71 P1 pretein with a molecular weight of 100 kD was produced and secreted into the medium.The soluble EV71 P1 protein was purified by column chromatography with a recovery efficiency of 70%.The result of the immunological analysis showed that the EV71 P1 protein had excellent immunogenicity and could stimulate the production of EV71-VP1 IgG antibody in injected rabbits.We suggest that EV71-P1 protein is an ideal candidate for an EV71 vaccine to prevent EV71 infection.
10.In vitro introduction of recombinant trichosanthin on demethylation of p27 in HeLa cells

Objective To investigate the effects of recombinant trichosanthin(rTCS) on methylation status and expression level of p27 gene in HeLa cells.Methods HeLa cells was treated by different concentration(20 ?g/mL,40 ?g/mL,and 80 ?g/mL) of rTCS for 48 h and then methylation-specific polymerase chain reaction(MSP) was used to detect the promoter methylation status of the p27 gene,real-time PCR was used to detect levels of p27 and DNMT1 mRNA,and Western blotting assay was used to detect expression level of p27 protein before and after treatment with rTCS.Results Low expression level and promoter methylation status of the p27 gene were detected in HeLa cells.Treatment with 40 ?g/mL rTCS totally demethylated p27 promoter.Treatment with 20 ?g/mL,40 ?g/mL or 80 ?g/mL rTCS resulted in a 2.22-,4.00-or 6.03-folds increase in p27 mRNA level,respectively,and also a great increase in p27 protein level.A high DNMT1 expression level was observed in HeLa cells and treatment with 40 ?g/mL rTCS resulted in a 78% decrease at the DNMT1 mRNA expression.Conclusion rTCS could reverse promoter hypermethylation and re-activate the expression of p27 gene by inhibiting DNMT1 expression in HeLa cells,which indicates its potential use in cancer therapy.
