1.Comparison of five kits for quantitation of hepatitis B virus DNA

Objective To compare the sensitivity, specificity and consistency of five kits for quantitative detection of hepatitis B virus (HBV) DNA. Methods Four domestic fluorescence kits, A, B, C, and D, and Roche Cobas Ampliprep/Cobas TaqMan HBV test for quantitation of HBV DNA in serum, were applied to detect the National Standards, an additional plasma for sensitivity (7 times dilution), 15 plasma samples from healthy blood donor and 45 plasma samples from chronic hepatitis B patients. Results All five kits showed the correct results for the 9 positive specimens and 8 negative specimens from the National Standards. The quantitative results of specimens for sensitivity met requirements of the National Standards. The lowest concentration of HBV DNA by these three kits was 15.6 IU/mL. The lowest detection level of HBV DNA for domestic kit B was ≤500 IU/mL. There was linear correlation between the results by Roche kit and domestic kit C (r> 0.98). Kit D showed 2 false-negatives results in the 15 healthy blood donor samples. The coincidence rates between 4 domestic kits and the Roche kit ranged from 50% to 96% (A: 61%, B: 50%, C: 96%, D: 83 %). The consistency rate of kit C with the Roche kit was significantly higher than those of kit D, A and B (X2=5.62, P<0.05, X2=28. 93, P<0.01, X2=44.31,P<0.01, respectively). The consistency rates among these 5 kits were highest when testing samples with HBV DNA levels between 1×104-1×107 IU/mL. Conclusions The qualities of domestic kits vary remarkably and kit C has the greatest consistency rate with the Roche kit. Therefore, the qualities of domestic kits need to be further improved.
2.Short-term evaluation of clinical effect of bone ring grafting and immediate insertion.

OBJECTIVETo observe the short-term clinical effectiveness of bone ring graft technique and to summarize the key points of related surgical operation to provide comprehensive clinical guidelines.
METHODSFifteen patients with severe alveolar bone absorption were selected to receive bone ring grafting and immediate dental implant. Final fixed prostheses were cemented five months after initial implantation. Cone beam CT scans were conducted on all subjects before the procedure, as well as four months post-operation to evaluate alveolar bone height and level of bone height and absorption around the implants. Four to six months after prosthesis installation, each implant's Jemt classification, gingiva attachment, and probing depth (PD) were analyzed. The difference of PD between implants and adjacent teeth, as well as the difference of the bone absorption between labial and lingual sides, was compared. The survival rate of the bone ring and the retention rate of implants were calculated. Complications and patient satisfaction were also investigated.
RESULTSBone graft survival rate was 94.4% and dental implantation retention rate was 100% four months post-operation. Average bone level increase was (6.06 +/- 1.06) mm, average bone absorption was (1.33 +/- 0.84) mm, and average bone thickness at the neck of the dental implant body was (6.94 +/- 0.73) mm. Approximately 4 to 6 months after crown restoration, average bone level increase was (5.62 +/- 1.03) mm, average bone absorption was (1.51 +/- 1.02) mm, and average bone thickness at the neck of the dental implant body was (6.77 +/- 0.72) mm. The PD around the implant body and the adjacent teeth was statistically insignificant. No major post-operative complication was observed, restorations were successful, and patient satisfaction level was high.
CONCLUSIONBone ring graft technique and immediate dental implantation are relatively simple to perform, and these techniques facilitate reduction in required treatment time. Short-term effect is reliable and satisfactory, whereas long-term outcomes require further follow up and study.

3.Effect of team follow-up on continuous management of cancer pain patients

Objective To explore the effect of continuous management of cancer pain patients by group follow-up model. Methods A total of 86 cases of cancer pain patients discharged from oncology department from March to May 2016 were selected and grouped according to the number of inpatients. The control group(44 cases)was treated with conventional intervention, and the experimental group(42 cases) was followed by team follow-up. The cancer pain control effect and quality of life were evaluated in the two groups after 1 month. Results The average degree of pain degree within 24 h in the past pain after intervention respectively was (2.90 ± 0.85) (2.56 ± 0.79) points in the experimental group and (4.17 ± 0.92) (4.00 ± 1.18) points in the control group, there was statistically significant difference between the two groups (t=6.618, 6.492, P < 0.01). The quality of life in physical function, cognitive function, social function, general health level was(66.83 ± 20.02),(69.33 ± 15.45),(61.17 ± 16.50),(63.83 ± 17.93)points in the experimental group,(27.83 ± 11.97),(36.17 ± 16.24),(26.14 ± 17.29),(28.23 ± 17.43)points in the control group, there were significant differences(t =4.224-9.727,P<0.05 or 0.01). There was no significant difference in role function and emotion function between the two groups (P>0.05). Conclusions The continuous management of team follow-up can help the patients with cancer pain continue to receive standard treatment and care after discharge, which will help improve the patient's pain control level and improve the quality of life.
4.Three-dimension finite element analyses of interior stress of two kinds of Replace implant.

OBJECTIVETo establish three-dimension finite element model of mandible with two kinds of Replace implant, and to study stress of implant and abutment.
METHODSThe data of components of the dental implant was measured, cross section of the mandible was scanned by spiral CT and image reconstruction technique was conducted. Three-dimension finite element analysis software UG and MSC. Marc/Mentat were used to built the conjunction model and bone model of two implant systems. Axial loading (200N) and 30 degrees oblique loading (100N) were applied on the models respectively, the stress distribution patterns of the implant and abutment of two implant systems were analyzed.
RESULTSThe peak stress district was concentrated on the structure of the implant cervix, which was more obviously displayed on the Replace Select implant. The peak stress of oblique loading was higher than that of axial loading. The peak stress on the implant cervix of Replace Select implant was higher than that of Replace External Hex implant in all loadings.
CONCLUSIONTo Replace Select especially, oblique force should be avoided on clinical practice in case of the implant fracture.

5.Study of labeling human periodontal ligament stem cells with enhanced green fluorescent protein by lentivirus vector infection.

OBJECTIVEThe aim of this study is to optimize conditions for labeling human periodontal ligament stem cells (PDLSCs) using enhanced green fluorescent protein (eGFP) infected by lentivirus vector and to obtain PDLSCs with high stable expressed eGFP.
METHODSPDLSCs were transfected with eGFP by lentivirus vector for 48 h via different multiplicity of infection (MOI) (25, 50, 100, 200 and 400) and the infection efficiency were analyzed by both fluorescent microscope and flow cytometry. The proliferation rate of infected PDLSCs was evaluated by MTT. The infected PDLSCs were further for detection of pluripotent, differentiation ability and alkaline phosphatase (ALP) expression ability.
RESULTSThe infection efficiency for each group were 44.7%, 60.9%, 71.7%, 85.8% and 86.9% respectively. Proliferation of PDLSCs was not affected when MOI was below 200; however, at MOI 400, the proliferation ability was affected compared with control group. The pluripotent and ALP abilities of PDLSCs were not changed by the infection.
CONCLUSIONInfection for 48 h at MOI 200 is optimal for labeling PDLSCs with eGFP using lentivirus vector, and the proliferation and differentiation abilities of PDLSCs are not affected obviously.

6.Mutation analysis of two pedigrees with suspected oculocutaneous albinism.

OBJECTIVE:
To analyze the clinical presentation and gene of 2 pedigrees with suspected oculocutaneous albinism(OCA), and provide basis for clinical classification, genetic counseling and prenatal diagnosis.
METHODS:
Variants were identified using next-generation sequencing(NGS) and confirmed by Sanger sequencing in 2 pedigrees with suspected OCA. The pathogenicity of the variants was analyzed according to the American College of Medical Genetics and Genomics (ACMG) standard.
RESULTS:
Two compound heterozygous mutations of TYR and OCA2 genes were identified respectively in 2 pedigrees with suspected OCA. The mutation of c.819+3insATATGCC in TYR and the mutation of c.1870G>C in OCA2 are first reported in this study. The pathogenicity analysis shows that two novel mutations are likely pathogenic by combination of prediction of SIFT, Polyphen-2 and Human Splicing Finder.
CONCLUSION
The findings of this study expand the mutational spectrum of OCA. Compound heterozygous mutations in the TYR and OCA2 gene may be responsible for clinical manifestations of 2 pedigrees with suspected OCA.

7.Research progress on molecular mechanism of transcription factor C/EBPβ in lung diseases

CCAAT enhancer binding protein β (C/EBPβ), as a nuclear transcription factor necessary for the development of liver, airway epithelium, and adipose tissue, plays a vital role in physiological processes related to cell proliferation, apoptosis, and differentiation. However, the up-regulation of C/EBPβ activates signal pathways related to inflammatory response, epithelial-mesenchymal transition, cell proliferation and invasion, immune response, and angiogenesis by regulating a series of downstream genes transcription promotes the development of lung diseases. Therefore, targeting C/EBPβ may be a potential treatment strategy for lung diseases. This paper summarizes the regulatory effects of C/EBPβ and related signaling pathways in lung infection, asthma, chronic obstructive pulmonary disease, lung injury, pulmonary fibrosis, and lung cancer to provide a theoretical basis for the precision medicine of lung diseases.
