Journal of Chinese Physician 2011;13(6):754-756
Objective To explore the relationship between metabolic syndrome and ApoA5 gene polymorphism and serum concentration. Methods 100 patients with metabolic syndrome(MS group) and 100 healthy people(control group) were enrolled in this study. The ApoA5 serum concentration of two groups were measured by using enzyme-linked immunosorbent assay(ELISA), the genotypes of ApoA5-1131T>C polymorphism were analyzed by polymerase chain reaction-restriction fragment length polymorphism, and fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL) and white blood cell, hemoglobin, platelet, liver and kidney function were measured. Results MS group was compared with control group. In MS group, the ApoA5 serum concentration was significantly lower[(96.68±18.09)ng/ml vs (128.32±23.78)ng/ml,P<0.01], while the triglycerides levels were obviously higher[(2.35±1.07)mmol/L vs (1.62±1.13)mmol/L,P<0.01], and ApoA5-1131C allele frequency was higher than that in control group (30% vs 16.5%,P<0.05). Conclusions The ApoA5 serum concentration in metabolic syndrome was decreased, and ApoA5-1131C was associated with metabolic syndrome.

Objective:To detect the expressions of survivin, galectin-3 and thyroid peroxidase in papillary thyroid microcarcinoma ( PTMC) and cancer adjacent normal tissue to explore the clinical significant. The correlations between the expressions of survivin,Gal-3 and TPO with BRAFV600E gene mutation in PTMC were analyzed. Methods: The expressions of survivin,Gal-3 and TPO were measured by immunohistochemical ( IHC ) method in 56 cases of PTMC tissue and adjacent normal tissue; BRAFV600E mutation was detected by PCR amplification and subsequently sequencing.Chi square test was used to analyse the relation in the expression rates of survivin,Gal-3 and TPO and BRAFV600E gene mutation. Results:The survivin and Gal-3 were strongly expressed but TPO was negatively expressed. The survivin and Gal-3 were negative in adjacent normal tissues but TPO was expressed. There were sig-nificant differences in the expression rates of survivin, Gal-3 and TPO between PTMC tissue and adjacent normal tissue (all P<0.001).The BRAFV600E mutation rate (32.1%) and the expression rates of survivin,Gal-3 and TPO in PTMC tissue were found to be positively related to lymph node metastases (P=0.009,P=0.025,P=0.007,P=0.008),and negatively related to gender and age (all P>0.05).There were no correlation was found between the expressions of survivin,Gal-3,TPO and the BRAFV600E gene mutation in PTMC(all P>0.05).Conclusion: The strong expressions of survivin and Gal-3,the mild expression of TPO and BRAF mutation may be important in the development of PTMC,and the detection of BRAFV600E gene mutation and the expressions of survivin, Gal-3 and TPO could be used to the judgment of pathogenetic condition and prognostic outcomes.

Objective:To explore the effect of sarpogrelate on platelet function in patients at the bridging stage before coronary artery bypass grafting (CABG).
Methods: A total of 40 consecutive patients with peripheral artery stent and scheduled for CABG in our hospital from 2011-05 to 2013-04 were enrolled in this study. The patients were randomly divided into 2 groups, Low molecular weight heparin (LMWH) alone group, n=19 and Sarpogrelate+LMWH group, n=21. The medications started at 5-7 days before CABG and stopped at 24 h before CABG. The platelet inhibition rates (platelet aggregation induced by collagen+ serotonin) were examined and compared between 2 groups at the baseline (before randomization), 24h and 1h before CABG respectively.
Results: The platelet inhibition rates were similar between 2 groups at the baseline (87.33 ± 6.82) % vs (86.11 ± 6.87) %, P=0.577 and 1h before CABG (62.60 ± 12.39) % vs (56.19 ± 14.99) %, P=0.148. At 24h before CABG, the platelet inhibition rate in Sarpogrelate+LMWH group was higher than that in LMWH alone group (83.87 ± 8.99)%vs (63.13 ± 10.88)%, P<0.001. Compared with the baseline, the falling range of platelet inhibition was lower in Sarpogrelate+ LMWH group at 24h before CABG, (3.46 ± 6.18) % vs (22.98 ± 9.43) %, P<0.001 and the falling range was similar between 2 groups at 1h before CABG (24.73 ± 14.19)%vs (29.92 ± 14.28)%, P=0.257.
Conclusion: Sarpogrelate + LMWH may result better platelet inhibition rate with quicker recovery of platelet function upon the medication stopping, which might be a feasible management in patients at the bridging stage before CABG.

OBJECTIVE:To explore the application of topic model in the study of type 2 diabetes treatment plan. METHODS:Clinical data of 650 inpatients with type 2 diabetes in our hospital during Jan. 2012-Jun. 2016 were analyzed retrospectively. The data of clinical diagnosis, lab indexes and clinical drug use were exchanged, summarized and merged by MATLAB R2014a software. Latent Dirichlet allocation and author topic model were adopted to extract the typical topics with topic probability value>0.1,and the topics was described by the complications with cumulative probability value>0.5. RESULTS:A total of 62 complications words,16 abnormal laboratory indexes groups and 20 treatment plans were obtained. A total of 4 typical topics were excavated(cumulative probability values for the first few complications were 0.8786,0.8247,0.8215,0.7536;topic probability value were 0.3364,0.2773,0.2035,0.1176,respectively) and were mainly characterized by peripheral neuropathy, peripheral vascular disease, abnormal lipid metabolism and microvascular lesions;abnormal lab indexes groups met the above characteristics. The complications with high distribution rate included diabetic peripheral neuropathy (0.5787), hypertension (0.3631),atherosclerosis (0.2789),hyperlipidemia (0.4578) and diabetic retinopathy (0.3143);main drugs included Insulin aspart injection,Insulin injection,Methylcobalamin dispersible tablets, etc. CONCLUSIONS:The complications of type 2 diabetes are characterized by peripheral neuropathy, peripheral vascular disease, abnormal lipid metabolism and microvascular lesions. The medication rules with clinical significance can be extracted from the clinical data by topic model.

A novel molecularly imprinted polymers (MIPs) was prepared based on multiwalled carbon nanotubes (MWNTs) by using chlorogenic acid as template, methacrylic acid(MAA) as the functional monomer, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. Infrared spectroscopy, scanning electron microscopy, and thermogravimetric analysis were employed to characterize the composite structure and morphology, the results showed that a stable imprinted layer was grafted on the MWNTs with the average thickness of 30-40 nm. The dynamic adsorption and static adsorption experiments were also executed and the results showed that two kinds binding sites with the maximum apparent binding constant 21.5 μmol/g and 32.7 μmol/g existed v.i. between the imprinted polymers and chlorogenic acid. As solid phase extraction (SPE) materials, the separation and enrichment of chlorogenic acid from honeysuckle were successful with a enrichment factor of 25.
