1.Determination of astragaloside IV in the Huangqijing by HPLC-RI

OBJECTIVE To establish a HPLC-RI assay for the determination of astragaloside IV in the Huangqijing solution.METHODS The column Hypersil ODS 2(4.6 mm×200 mm,5 μm) was used.The mobile phase was consisted of methanol ane water (67∶33).The flow rate was 1 mL.min-1.RESULTS The standard curve was linear over the range of 1~5 μg with the correlation coefficient 0.9999.The average recovery was 98.1% with the RSD 2.02%(n=6).CONCLUSION This method was sensitive,accurate,repeatable and easy to operate.
2.Investigation and control of nosocomial infection in inpatients of department of gynecology and obstetrics

ThO clinical data of infOctOd group and non -infOctOd group wOrO comparOd, and thO risk factors of nosocomial infOction in patiOnts of dOpartmOnt of gynOcology and obstOt-rics wOrO analyzOd by singlO factor analysis. MultivariatO logistics linOar rOgrOssion analysis was usOd to analyzO thO rOlatOd factors of nosocomial infOction in patiOnts of dOpartmOnt of gynOcology and obstOtrics. Results Of 150 patiOnts, thOrO wOrO 37 patiOnts with nosocomial infOction in thO hospital, thO hospital infOction ratO was 24.67%. Of 37 infOction patiOnts, 23 casOs wOrO from dOpartmOnt of obstOtrics, 14 casOs wOrO form dOpartmOnt of gynOcology, obstOtrics was morO than gynOcology(χ2 =4.378,P<0.05). ThO main infOction sitOs wOrO rOspiratory tract, urinary tract, surgical incision, and thO infOction ratOs wOrO 43.24% , 27.03% ,21.62% ,rOspOctivOly. UnivariatO analysis showOd that thOrO wOrO statistically significant diffOrOncOs bOtwOOn thO infOction group and non -infOction group in agO, BMI indOx, invasivO opOration, hospitalization timO, indwOlling cathOtOr, diabOtOs (χ2 =5.165,5.436,7.175, 6.034,8.134,7.334,all P<0.05). MultivariatO linOar rOgrOssion analysis showOd that agO, BMI indOx, invasion opOration, hospitalization timO, indwOlling cathOtOr, diabOtOs wOrO thO risk factors of nosocomial infOction in hospital-izOd patiOnts.Conclusion ThO nosocomial infOction of inpatiOnts in dOpartmOnt of gynOcology and obstOtrics arO affOctOd by agO BMI indOx, invasivO opOration, hospitalization timO, indwOlling cathOtOr, diabOtOs mOllitus and so on. ThO infOction occurrOd in obstOtrics, mainly rOspiratory tract infOction, urinary tract infOction, surgical incision infOc- tion. Clinical work should bO aimOd at thO risk factors to carry out prOvOntivO intOrvOntion.
3.Research on similarities and differences of information needs between cancer patients treated with radiotherapy and chemotherapy

Objective To study the similarities and differences of information needs between cancer patients treated with radiotherapy and chemotherapy, analyze the causes for cancer patients' different choices, and provide reasonable and disease-related information for cancer patients. Methods A total of 102 and 119 cancer patients treated with radiotherapy and chemotherapy relatively were asked to complete the questionnaire (IPQCP+2) and two questions about the total information capacity and content requirements. Results Two groups had different needs of therapeutic information and treatment in IPQCP. Cancer patients treated with chemotherapy needed more widely-ranged information than patients underwent radio-therapy. Conclusions Most of cancer patients treated with radiotherapy and chemotherapy need to be informed of disease-related information, among which the content of therapeutic information and total information is relative to the treatment. Nurses should provide disease-related medical information based on their different needs, as to improve the life quality of cancer patients.
4.Experimental study of recombinant human erythropoietin therapy for contusion injury of spinal cord

[Objective]To study the therapeutic effect of recombinant erythropoietin for contusion injury of spinal cord.[Method]Contusion injury of spinal cord was caused by weight dropping in 24 New Zealand rabbits.Twelve hours after injury,the rabbits in control group were given natural saline intravenously and rabbits in low,mediate and large-dose group were given rh-EPO 100 IU/kg,500 IU/kg and 1000 IU/kg respectively.Neurological status of lower limbs were scored at 24 hours,48 hours and one week after spinal cord injury.All rabbits were killed one week after injury and spinal cords were stained by HE and caspase-3 method.Electronic microscopy was used to evaluate ultrastructural injury.[Result]The neurological scores of EPO treated groups were significantly higher than that of control group.HE and Caspase-3 immunohistochemistry showed that histological and ultrastructural damage of EPO treated groups were less severe than that of control group.The caspase-3 positive neurons were significantly fewer than that of saline treated group.There was no significant difference of therapeutic effect between mediate and large-dose EPO treated groups.[Conclusion]Rh-EPO administered 12 hours after contusion injury of spinal cord could lessen histological and ultrastructural damage,prevent apoptosis of neurons and promote neurological recovery of spinal cord.Mediate dose of rh-EPO is the appropriate treatment choice for spinal cord injury.
5.Clinical observation of Fufangqiaoruisu granule in the treatment of 76 children with bronchial asthma

Objective To study the clinical effect of Fufangqiaoruisu granule in the treatment of children with bronchial asthma.Methods 152 children with bronchial asthma were randomly divided into treatment group and control group.All the patients were treated with based therapy,and the patients in the control group were treated with montelukast,the patients in the treatment group received montelukast/Fufangqiaoruisu granule.The clinical symptoms such as cough,asthma were compared,and also the total effective rate.Results The disappear time of clinical symp-toms of cough,ashma,moist rale in the treatment group were (3.8 ±1.0) days,(3.5 ±0.8) days,and (2.1 ± 0.6)days,which were much shorter than (5.2 ±0.7)days,(4.7 ±0.9)days,(3.0 ±0.5)days in the control group, the differences were statistically significant(t =4.56,5.12,4.89,all P <0.05).After treatment,the FVC,FEV1 and PEF in the treatment group were (4.9 ±1.7)L,(3.8 ±0.6)L and (3.9 ±0.5)L/min,which in the control group were (3.2 ±1.2)L,(2.1 ±0.8)L and (2.8 ±0.5)L/min,(t =5.12,4.87,5.32,all P <0.05).The total effective rate of the observation group was 92.1%,which of the control group was 73.7%,the difference was statistically significant(χ2 =1.62,P =0.027).Conclusion Fufangqiaoruisu granule shows satisfactory clinical efficacy in the treatment of bronchial asthma,and it is worthy of clinical use.
6.The effect of non -xanthine adenosine receptor antagonist on ventricular remodeling and RAAS in chronic heart failure

Objective To analyze the effect of non -xanthine adenosine receptor antagonist (SLV320)on the ventricular remodeling and renin -angiotensin -aldosterone system (RAAS)in animal experimental models of chronic heart failure (CHF).Methods The 40 healthy male New Zealand rabbits were received adriamycin by intra-venous injection to establishing the experimental animal models and were randomly divided into 4 groups,which were high -dosage group (injected with SLV320,10.0μg·kg -1 ·d -1 ),medium -dosage group (injected with SLV320, 5.0μg·kg -1 ·d -1 ),low -dosage group (injected with SLV320,2.5μg·kg -1 ·d -1 )and furosemide group (fed with furosemide,2.0mg·kg -1 ·d -1 ).Each group had 10 rabbits and continuous treated for one week.The indexes of plasma renin activity (PRA),angiotensinⅡ (AngⅡ),aldosterone (ALD)and beta ntriuretic peptide (BNP)were detected at pre -and post -treatment,and compared among 4 groups.The indexes of left ventricular end -systolic dimension (LVESD),left ventricular end -diastolic dimension (LVEDD),left ventricular posterior wall (LVPW), left ventricle ejection fraction (LVEF),left ventricular fractional shortening (LVFS)and E /A at pre -and post -treatment were detected by echocardiography and compared among 4 groups.The wet weigh of the left and right ventri-cle were weigh accurately.And the indexes of left ventricle weight index (LVWI)and the body weight index (BWI) were calculated and compared among 4 groups.Results The plasma levels of PRA,AngⅡ,ALD and BNP were no different among 4 groups before study (all P >0.05).The sequence of 4 groups on the plasma levels of RAAS indexes was high -dosage group
7.Clinical observation of maintenance therapy with gefitinib after the first-line chemotherapy for patients with advanced non-small-cell lung cancer

Objective To evaluate RR,DCR,PFS,OS and toxicities in a population affected by NSCLC using gefitinib as maintenance therapy after the first line chemotherapy.Methods From January,2006 to January,2008,seventy-one patients were enrolled with stable disease or partial response after firstline chemotherapy.They were divided into the observing group and the control group.Gefitinib was administered at the dose of 250 mg in observing group;placebo was also administered everyday in control group.Results Of 71 assessable patients ,overall response rate was 36.1% (13/36) in observing group and 14.3 % (5/35) in control group respectively (x 2 = 4.633,P = 0.036),and one patient (2.8 %) was observed complete response (CR) in observing group.The disease control rate was 83.3 % (30/36) in observing group and 42.9 % (15/35) in control group,respectively,(x2 = 14.782,P < 0.001).The median PFS (progression free survival time) was 13 weeks in observing group and 11 weeks in control group respectively (x2 = 10.401,P =0.001).The median overall survival time (OS) was 13.2 months in observing group and 10.4 months in control group respectively (x2 = 7.696,P= 0.006).The median OS was 18.5 months in women and 11.2 months in men(x2 =22.864,P=0.011) .The median OS was 15.3 months in non-smokers and 10.3 months in smokers (x2 =10.389,P =0.007).The median OS was 16.0 months in adenocarcinoma and alveolar cell carcinoma patients and 10.2 months in squamous cell carcinoma patients (x 2 = 4.638,P = 0.001).The most common toxicity was rash ,diarrhea,fatigue and dry skin itching,and most of them were 1 or 2 grade.Conclusion Maintenance therapy with gefitinib after first-line chemotherapy may improve overall survival time and progression free survival time in patients with NSCLC,and it is effective and safe.
8.Clinical Observation of Oxytocin Combined with Compound Leonurus japonicus Oral Solution in the Treat-ment of Hemorrhage after Artificial Abortion

OBJECTIVE:To observe therapeutic efficacy of Compound Leonurus japonicus oral solution in the treatment of hemorrhage after artificial abortion and its effect and safety on normal menstruation recovery. METHODS:100 cases of artificial abortion selected and randomly divided into treatment group and control group,with 50 cases in each group. Control group re-ceived oxytocin intramuscularly after surgery,10 U/time,12 h/time,for 3 times;treatment group was additionally given Com-pound L. japonicus oral solution,2 piece/time,3 times/d,for 5 days,on the basis of control group. Metrorrhagia duration,hemor-rhage amount,normal menstruation recovery and duration of abdominal pain and ADR were all observed in 2 groups. RESULTS:The metrorrhagia duration [(5.38±0.95)d] of treatment group was significantly lower than that [(8.58±1.32)d] of control group, with statistical significance(P<0.05);hypomenorrhea cases(24 cases)of treatment group was higher than those(9 cases)of con-trol group;menorrhagia cases(8 cases)was lower than those(24 cases)of control group;with statistical significance(P<0.05). The normal menstruation recovery time [(27.34±3.87)d] of treatment group was significantly shorter than that [(40.13±2.58)d] of control group,with statistical significance(P<0.05);the duration of abdominal pain [(2.85±0.89)d] in treatment group was sig-nificantly shorter than [(5.02 ± 1.33)d] in control group,with statistical significance (P<0.05).Obvious ADR was not found in both groups. CONCLUSIONS:Compound L. japonicus oral solution is effective in the treatment of hemorrhage after artificial abor-tion,can significantl shorten metrorrhagia duration and hemorrhage amount,recover normal menstruation early,improve the symp-toms of abdominal pain after operation and is easy to take and good safety.
9.Optimization of mandibular CT scanning

Objective To Optimize the mandibular CT scanning methods with changing the scanning position,to reduce the radiation dosage.Methods Totally 50 consecutive patients with mandibular CT examination were enrolled in the study and divided into position-optimized group and routine group (n =25,respectively) randomly.The image quality,volume CT-dose index (CTDIvol),scanning length,and dose length product (DLP) were compared.Results There was no significant difference in subjective image quality and CTDIvol between two groups.But the scanning length and DLP in positionoptimized group were significantly reduced by about 20 % (t =4.986,P < 0.05) and 16% (t =3.682,P <0.05) compared with routine group.The lens were out of scanning field with the position-optimized protocol.Conclusions The scanning length would be efficiently reduced and the lens avoided with the position-optimized protocol,which would subsequently reduce the effective radiation dose.
10.Effect of Rho signaling pathway on SiO2 induced α-SMA expression in human bronchial epithelial cells

Objective To investigate the role of Rho in SiO2 induced α-SMA expression in human bronchial epithelial cells (HBECs). Methods HBECs were cultured and stimulated with SiO2. Immunocytochemistry and Western blot were used to detect the expression of α-SMA. The activity of Rho was determined by GST pull down assay. In the prevention experiment,SiO2-stimulated HBECs were incubated with Rho inhibitor Y27632,and the expression of α-SMA was examined by Western blot. Results With SiO2 (0-300 μg/mL) treatment,the expression of α-SMA increased gradually,and 200 μg/mL of SiO2 led to the highest expression of α-SMA which was (5.09±1.98) times of the expression of α-SMA in the control group(P<0.01). HBECs treated with SiO2 (200 μg/mL) for indicated time (1,2,6,12,and 24 h)showed an obvious increase of Rho activity(P<0.01). Y27632 inhibited SiO2-induced α-SMA expression significantly,and the inhibition rate of 20 and 30 μmol/L Y27632 was 68% and 75%,respectively (P<0.01).Conclusion Rho signaling pathway may mediate SiO2 induced α-SMA expression in HBECs.
