1.Expression of suppressors of cytokine signaling in mater-fetal interface of human first trimester gestation

Objective:To investigate the mRNA and protein expression of suppressors of cytokine signaling in human villa and decidua of first trimester gestation.Methods:We first obtained villous tissues and decidual tissues from 38 women who had undergone selective termination at 7-9 weeks of normal gestation.The transcripts of SOCS1,SOCS2,SOCS3 in villous tissues and decidual tissues were detected by using semi-quantitative reverse transcriptase polymerase chain reaction.The mRNA of SOCS1,SOCS2,SOCS3 from cultured trophoblasts and decidual stromal cells were detected.Also the protein expression of 35 pregnant women were detected by western blot.Immunohistochemistry analysis revealed the location of SOCS1,SOCS2,SOCS3 in materno-fetal interface.IL-10 and IFN-? secreted by cells in vitro were detected by ELISA.Results:The expression of SOCS1,SOCS2,SOCS3 were found in mater-fetal interface,while positive expression of SOCS3 in villi/decidua was 73.7%/71.1%,and that of SOCS2 was 50.0%/39.5%,then SOCS1 expression was 34.2%/31.6%.The protein expression of SOCS1,SOCS2,SOCS3 were similar to the transcription level.SOCS1,SOCS2,SOCS3 were mainly expressed in villi and decidual stroma.Trophoblasts and decidual stromal cells were not found the expression of SOCS1,but the low level mRNA of SOCS2,SOCS3 without serum,and IL-10 secreted by them increased significantly(P
2.The effects of allopurinol in different dose on the heart function of chronic heart failure rats induced by adrimycin

Objective To study the effects of allopurinol in different dose on the cardiac function of chronic heart failure rats induced by adrimycin. To explore dose-dependency of allopurinol in improving blood vessel endothe-lium function and cardiac ventricle remodeling of the rats heart, as to supply evidence and new sight in clinical treat-ment of congestive heart failure. Methods 40 Sprague-Dawley male rats were randomly divided into four groups: normal control group (group A)、model control group (group B)、low-dose allopurinol group (group C)、high-dose allopurinol group (group D). The heart failure model was made by administering adriamycin to rats. After the model myocardial pathological changes were detected. Results Compared with the normal control group, the weight and heart weight of rats in model control group and allopurinol groups were obviously lessen (group A=(300.10± 9.85)g,group B=(200.67±9.91)g, group C=(233.14±9.42)g,group D=(248.25±13.34) g;group A= (828.30±50.97) nag, group B=(681.50±16.97) mg, group C=(743.00±17.20) nag, group D=(784.88± 36.83) mg,P<0.05). Heart weight indexes were all incerased ( group A=(2.76±0.15) mg/g, group B=(3.41± 0.17) mg/g, group C=(3.26±0.76) mg/g, group D=(3.11±0.65) mg/g, P<0.05). The hemedynamics resuh showed that myocardial contractile force were enhanced in drug groups. The level of NO, SOD were increased in the allopurinol groups compared with the model control group (group B: NO=(41.55±6.28) μmol/L, group C: NO= (52.47±5.59) μmoL/L,group D:NO=(61.04±4.26) μmoL/L; group B:SOD=(63.83±6.40) U/ml,group C: SOD=(76.29±7.99) U/ml, group D: SOD=(100.13±7.43) U/ml, P<0.05) and MDA levels were obviously decreased (group B: MDA=(9.70±1.08) μmol/L, group C: MDA=(6.64±0.34) μmol/L, group D: MDA= (5.72±0.71)p.moVL,P<0.05). The level of NO, SOD were obviously increased in the allopurinol of high-dose group compared with low-dose allopurinol group(P<0.05). MDA levels were obviously decreased(P<0.05). The myocardial pathological changes were relieved obviously in the allopufinol groups. Conclusion Allopurinol improves blood vessel endothelium function dose-dependently. High-dese allopurinol obviously decreases MDA, improve NO, SOD, thereby can improve the cardiac function of heart failure.
4.Study on the health equity in urban dwellers from social stratification

Objective To investigate health equity status of urban dwellers with different social stratification and provide evidence for health promotion.Methods 1,007 urban dwellers were surveyed in Zhejiang province in a stratified sampling based on their income,occupation and education,with their health equity analyzed.Results The proportion of urban dwellers to participate in social health insurance reached 97.5%,a proportion of statistical significance (P<0.05).The two-week prevalence rate and two-week attendance rate of urban dwellers of different social stratification has no significant difference,but the rate of sickness without attending was significantly negatively correlated to social stratification.The proportion of no visiting because of poverty was 23.1% in the lower social class,3.2% in middle and lower class,and 0% in the middle and upper class,a difference being statistically significant (P<0.001).The outpatient clinic of first choice and ranking of the hospital was positively correlated to social stratification.Conclusion There is health inequity for urban dwellers with different social stratification,for which the government should pay more attention to lower social class.
5.Study on the Relationship between Cisplatin-resistance of Human Ovarian Cancer Cell and TLR9 under Hypoxia

OBJECTIVE:To study the relationship between cisplatin-resistant of human ovarian cancer cells (SKOV3) and Toll-like receptor 9 (TLR9) under hypoxia. METHODS:Immunofluorescence method was used to detect the TLR9 expression in rupture and intact membranes SKOV3. SKOV3 was taken,adding cisplatin,then CCK-8 was used to detect cell inhibition rate af-ter 6,12,24 h of added into TLR9 specific agonists CpG-ODN 2006 (group A) and its isotype control CpG-ODN 2006 control (group B). SKOV3 or SKOV3 pretreated by 0(not added),1,10,102,103 μmol/L TLR9 specific antagonist chloroquine for 3 h were taken,adding cisplatin,cell inhibition rate was detected after 24 h of added into SKOV3 normoxia(21% O2),hypoxia(1%O2)incubating 6,12,24 h,HICR under hypoxia in pretreatment was calculated. Western blot method was used to detect the multi-drug resistance associated protein(MRP)expression in SKOV3 under the conditions of normoxia 24 h cell supernatant(group C), hypoxia 24 h cell supernatant(group D),hypoxia 24 h cell supernatant+10 μmol/L chloroquine(group E). RESULTS:TLR9 was expressed in both cell membrane and cytoplasm of SKOV3. Compared with group A,cell inhibition rate in group B was decreased (P<0.01). Compared with normoxia cell supernatant,cell inhibition rate was decreased after hypoxia cell supernatant (P<0.05). Compared with not added chloroquine,adding 10,102 μmol/L chloroquine can obviously decrease HICR(P<0.01),and the most significant decrease was showed when using hypoxia 24 h cell supernatant+10 μmol/L chloroquine. Compared with group C,MRP expression in group D was strengthened (P<0.01);compared with group D,MRP expression in group E was weakened (P<0.01). CONCLUSIONS:TLR9 receptors can be activated under hypoxia,and cause SKOV3 to cisplatin-resistance,the effect may be related with up-regulating MRP expression.
6.Clinical values of superoxide dismutase and malondialdehyde detection in cord blood of newborns with fetal distress

Objective To investigate the relations between intrauterine asphyxia and peroxidation and newborn hypoxic-ischemic encephalopathy(HIE). Methods The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in cord blood of 60 newborns with intrauterine asphyxia during labor(which was divided into two groups,39 cases with asphyxia in groupⅠ, and 21 cases with asphyxia in groupⅡ),and in 30 newborns without intrauterine asphyxia(control group) were determined. The levels of SOD and MDA in cord blood of newborns with HIE were compared with those in newborns without HIE. The incidence of HIE was estimated simultaneously. Results (1) The levels of SOD were (12 896?247) U/g Hb in groupⅠ, (9846?268) U/g Hb in groupⅡ, (17 282?134) U/g Hb in control group, significantly lower in the former two groups compared with control group, while the level of SOD in group Ⅰ was higher than that in group Ⅱ(P121 min group, and the levels of SOD was (9786?249) U/g Hb.(2)The levels of MDA were (6.3?0.4) ?mol/L in group Ⅰ, (8.6?1.5) ?mol/L in group Ⅱ, and (4.1?0.5) ?mol/L in control group, significantly higher in the former two groups compared with control group (P
7.Equity investigation of primary public health service for the migrant population

Objective To probe into the inequality found in primary public health service available to the migrant population and permanent residents. Methods With on-site interviews, 581 migrant workers and 581 permanent residents in a county in Zhejiang province were surveyed, to learn the inequalities between the two in the expenditure, access and outcomes of the public health service accessible to each. Results only 7. 8% of the migrant population ever had access to medical aid, a ratio far below that of the permanent residents; 40. 9% of the migrant population may turn down medical service beyond their affordability, a ratio far higher; for the migrant population, their ratio of gynecological checkup is 58. 7%, lower than that of the permanent residents (67. 7%). Conclusion Resources and financial allocation to institutions of public health should be enhanced, and social medical and insurance system should be developed and improved, in an effort to encourage the equality of primary public health services accessible to various sectors of the population.
8.Quality of life and influencing factors for migrant workers in Zhejiang province

Objective To identify the quality of life (QOL)and major influencing factors for migrant workers in Zhejiang province.Methods The QOL of 1217 migrant workers and 1387 permanent urban residents in Zhejiang province were measured by WHOQOL-BREF scale (Chinese version)in a stratified sampling.A multiple linear regression model was used to analyze the relationship between the factors and the scores of the four domains.Results There were no significant statistical differences(P>0.05)found in the scores of physical health,social relationship and general health between migrant workers and urban residents.Their scores of psychological health,surrounding conditions and general quality of life,however,were found lower than urban residents (3.5 ± 0.8 vs 3.6 ± 0.8).Influencing factors for their psychology were education,family conflicts and appetite; those for their surrounding conditions were daily average work hours and appetite.Conclusion Given improvements in the quality of life of migrant workers,rooms to improve were found in such scores as psychological health and surrounding conditions.
9.Management value of high-risk human papilloma virus detectionin the women with atypical squamous cell of undetermined significance and low grade squamous intraepitheliallesion of thin prep liquid-based cytology

Objective To discuss the significance of high-risk human papilloma virus ( HR-HPV) detection by hybrid capture 2 (Hc-2) in managing the women with atypical squamous cell of undetermined significance (ASCIIS) and low grade squamous intraepithelial lesion (LSIL) of thin prep liquid-based cytology (TCT). Methods One hundred and seventy-eight women with ASCUS and 108 women with LSIL were studied. The infection of HR-HPV in the women was analyzed. And sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) were used to evaluate the significance of HR-HPV detection by Hc-2 in managing the women with ASCUS and LSIL of TCT. Results Sensitivity,specificity, PPV and NPV of HR-HPV DNA detection by Hc-2 in screening CIN II or above in ASCUS were 89.83% (53/59),53.78% (64/119), 49.07% (53/108), 91.43% (64/70) respectively,and in LSIL were 97.62%(41/42),22.73%(15/66), 44.57%(41/92), 93.75%( 15/16) respectively. Conclusion HR-HPV detection by Hc-2 is an effective measure in managing the women with ASCUS and LSIL of TCT.
10.Antidepressant effect of curcumin in mice

BACKGROUND: The development of safe and powerful antidepressant agents from traditional Chinese herbs has become a hotspot in studies on anti-depression therapy. OBJECTIVE: To investigate the anti-depressive effect and possible mechanism of curcumin by behavioral and neurochemical procedures. DESIGN: Randomized grouping design and controlled experiment. SETTING: Depart, ment of Pharmacology, School of Basic Medical Sciences of Peking University.MATERIALS: This study was carried out in the laboratory of the Department of Pharmacology, School of Basic Medical Sciences of Peking University, between November 2003 and October 2004. A total of 240 male ICR mice were recruited. METHODS: The whole experiment was divided into 4 tests. ① Antagonism of reserpine-induced hypothermia: Totally 60 mice were randomly chosen and divided into 6 groups: normal control group, groups of various doses of curcumin (1.25, 2.50, 5.00 and 10.00 mg/kg), and positive control group (imipramine 10 mg/kg). Normal temperature of the mice was measured before experiment. The animals were given a single injection of reserpine (2.5 mg/kg). The mice were administered with drugs 18 hours later, namely, curcumin of different concentrations by gastric perfusion, groundnut oil (0.1 mL/10g by gastric perfusion) as well as imipramine (10 mg/kg by intraperitoneal injection). Rectal temperature was measured 60, 90, 120,150 and 180 minutes after administration, respectively. ② Potentiation of 5-hydroxytryptophan (5-HTP)-induced head twitches: animal grouping was the same as above, and the drug in positive control group was replaced by fluoxetine. The mice received gastric perfusion and the dose of curcumin given was the same as above. Groundnut oil and fluoxetine (10 mg/kg) and 5-HTP (70 mg/kg) were injected into the vein of the tail one hour later.The number of head twitches was counted within 5-10 minutes after 5-HTP treatment. ③ Antagonism of apomorphine-induced hypothermia: Mice grouping was the same as above; the drug in positive control group was replaced by imipramine. Curcumin was give as above at 4 doses, and groundnut oil and imipramine were also given. Large-dose apomorphine was injected subcutaneously (16 mg/kg). Rectal temperature was measured before injection, as well as 30 minutes and 60 minutes after injection. ④Determination of monoamine and metabolites: Mice grouping was the same as above. The drug in positive control group was replaced by imipramine.Curcumin was give as above at 4 doses, and groundnut oil and imipramine were also given. The content of monoamine and metabolites in the mice was measured with high performance liquid chromatography. ⑤ Dunnett's t test was used for comparison between groups.MAIN OUTCOME MEASURES: ① In reserpine-induced hypothermia test, the change of body temperature before and after administration. ② In 5-HTP-induced head twitches test, whether the times of head twitches were increased. ③ In apomorphine-induced hypothermia test, the change of body temperature after administration. ④ Effect of drugs on the content of monoamine.RESULTS: Totally 240 mice entered the result analysis. ① Experiment results of reserpine-induced hypothermia: Curcumin (5 mg/kg and 10 mg/kg)produced an antagonism against reserpine-induced hypothermia, and the results were significantly different from those in control group (P < 0.05,P<0.01). Curcumin of 10.00 mg/kg produced the similar effect compared as that of imipramine in positive control group. ② Results of 5-HTPinduced head twitches: Curcumin (5 and 10 mg/kg) could significantly increase the times of 5-HTP-induced head twitches (P <0.05, P<0.01). ③Results of apomorphine-induced hypothermia test: 2.50, 5.00 mg/kg and 10.00 mg/kg of curcumin could significantly increase the content of 5-HTP, and 10 mg/kg of curcumin could significantly increase the content of norepinephrine and dopamine. There was significant difference from that in control group (P < 0.05). By contrast, curcumin had no obvious effect on the content of metabolite 5-hydroxyindol acetic acid and 3,4-dihydroxyphenylacetic acid. Imipramine of 10 mg/kg as the positive control drug could significantly increase the content of 5-hydroxyindol acetic acid and norepinephrine (P<0.05).CONCLUSION: Curcumin has an antidepressant effect and the effect exerted may be related to monoaminergic neurotransmitter system.
