Objective To investigate the effects of LncRNA MT1JP on the migration and invasion of uveal melanoma cell,and explore its regulatory mechanism.Methods The expression level of LncRNA MT1JP in uveal melanoma cell line and normal retinal pigment epithelium cell line were measured by qRT-PCR.The SP6.5 cell line was divided into three groups,MT1JP group transfect with pcDNA3.1-MT1JP,siMT1JP group transfect with pcDNA3.1-siMT1JP and NC group transfect with negative pcDNA3.1 plasmid.The migration and invasion ability were measured by cell wound scratch assay and transwell assay.Western blot was used to measure the expression level of P53 protein.Results The expression level of LncRNA MT1JP in SP6.5 and M23 cell line was 0.20 ± 0.02 and 0.31 ± 0.01,respectively,which was significantly lower than 1.0 in ARPE-19 cell line (all P ＜0.01).The wound healing rate in siMT1JP group was 61.50 ± 3.70％.while,NC group was 20.0％ ± 2.10％,and MT1JP group was 10.6％ ± 1.7％,there was no statistical difference among three groups (all P ＜ 0.01).The invasion cell number was 75.6 ±6.8 in siMT1JP group,which was significantly more than 29.5 ± 3.9 in NC group,and 10.3 ±2.6 in MT1JP group,there was no statistical difference among three groups (all P ＜ 0.01).The expression level of P53 protein in siMT1JP group was 0.41 ± 0.04,which was significantly lower than 1.0 in NC group (P ＜ 0.01).However,the expression level of P53 protein in MT1JP group was 5.73 ±0.62,which was significantiy higher than 1.0 in NC group (P ＜ 0.01).Conclusion LncRNA MT1JP inhibits the migration and invasion of uveal melanoma cell,which may be associated with upregulation of P53 expression.

It was recently found that the changes in activity of MAPKs occurred during ischemia/reperfusion, but the biological significance of the changes was still controversial.

Traditional Chinese Medicine Informatics is a new branch created by integration of Traditional Chinese Medicine and Information Science.To research the connotation and principle of TCM Informatics will enrich the theory and promote the development of disciplines.This article thoroughly discussed the theoretical foundation and discipline characters of TCM Informatics.Then it discussed the principle and tasks from TCM information formation,acquisition,identification,transformation,feedback control,activation and dissemination of experiential knowledge.

AIM: To investigate the change and the possible role of mitogen-activated protein kinase(MAPK) subfamilies in early recovery process following acute renal ischemia/reperfusion injury. METHODS: Ischemia/reperfusion renal injury model was made by placing an atraumatic vascular clamp in renal pedicel. The morphologic change was observed by transmission electron microscope. The proliferating cell nuclear antigen(PCNA) positive renal cells were detected by immunohistochemistry. Extracellular signal-regulated kinase (ERK) and c-Jun NH 2-terminal kinase(JNK) activity was assayed by specific substrate phosphorylation with immunoprecipitation. RESULTS: After acute ischemia, microvilli in renal tubular cells appeared again after 2h of reperfusion. At the same time, the PCNA-positive cells were initially increased. ERK activity decreased at 45 min of ischemia, and completely recovered at 5 min of reperfusion. JNK activity was not influenced by ischemia, but increased at 5 min of reperfusion, reaching its maximal activity at 20 min of reperfusion, and prolonged within 2h reperfusion. CONCLUSION: After renal ischemia/reperfusion injury, the early recovery of renal tubule damage was related to the changes of MAPKs in which the increase of JNK activity might be more important.

Objective To explore the influencing factors of routine blood test results in the laboratory ,to ensure the accuracy of test results .Methods The influence of the different area of drawing blood and different testing time after blood collection were ob-served .Results The leukocyte count ,red blood cell count ,hemoglobin level of finger blood were obviously higher than those of venous blood ,and the platelet count was lower than that of venous blood ,the difference was statistically significant(P<0 .05) .≥6 h test specimens of blood platelet count was significantly lower ,compared to immediate detection and less than 6 h ,the difference was statistically significant(P<0 .05) .Conclusion The improvement of the system ,operation specification ,improving the quality of samples are helpful to improve the accuracy of test results .

Objective To explore the effect of Sunset Abelmoschus on podocyte injury in adriamycin-induced nephropathy rats.Methods Fifty male SD rats were randomly divided into five groups:sham operation group (n=10),model group (n=10),Sunset Abelmoschus low dose group (0.5 g·kg-1· d-1 n=10),middle dose group (1.0 g· kg-1· d-1,n=10) and high dose group (2.0 g· kg-1· d-1,n=10).Unilateral nephrectomy combined repeated adriamycin injection were performed to establish adriamycininduced nephropathy models.The rats were administered with the corresponding dose of Sunset Abelmoschus during the experiment period.Urinary protein,urinary N-acetyl glucose aminotransferase (NAG),serum albumin,serum creatinine and blood lipid were measured before operation and 2,4,6,8 weeks after operation.The rats were sacrificed on week 8 for the renal histological examination,including light microscope and electron microscope.Expression of nephrin was examined by immunofluorescence assays.Results As compared to model group,urinary protein and NAG significantly decreases in Sunset Abelmoschus groups in each time point,especially in high dose group (P＜0.01),meanwhile the serum albumin increased and the disturbance of lipid metabolism was improved in Sunset Abelmoschus groups (P＜0.05).Compared with sham group,Scr increased significantly in model group and Sunset Abelmoschus groups at the 4th week.At the 8th week,Scr in high dose group was lower than that in model group (P＜0.05),and the ratio of glomerular globe sclerosis and segmental sclerosis,tubulointerstitial damage reduced in Sunset Abelmoschus groups,especially in high dose group.The podocyte damage and the extent of foot process fusion were improved in Sunset Abelmoschus groups compared with model group.Expression of nephrin increased in Sunset Abelmoschus groups than that in model group.Conclusion Sunset Abelmoschus can ameliorate proteinuria and renal tissue damage of adriamycin-induced nephropathy rats,whose mechanism may be associated with the improvement of podocyte injury.

Objective To investigate the trends of colorectal cancer incidence in Shanghai Xuhui District from 2001 to 2011.Methods Total 3042 cases of colon cancer and 1958 cases of rectal cancer were collected from the Shanghai Cancer Reporting System.The results were age-adjusted according the census data of 2000 as the standardized incidence.Results The standardized incidence rate of colorectal cancer in Xuhui District rose from the 21.83/100 000 in 2001 to 27.35/100 000 in 2006 and increased by 25.29％ ; from 2009 to 2011 the incidence rate was stabilized.The ratio of colon to rectal cancer was 1.55:1,and the incidence of colon cancer was significantly higher than rectal cancer.The incidence rate of male and feinale colorectal cancer increased with age,especially at ＞ 45 y group accounting for 98.70％ and 98.37％ of total incidence rate respectively.The ratio of male to female in colon cancer incidence rate was 0.98∶ 1,while that of rectal cancer was1.25∶ 1.Conclusion The incidence of colorectal cancer in Xuhui District from the last 11 years is significantly higher than of national level.More effective preventive measures should be taken.

ObjectiveTo investigate the effects of scavenger receptor A on the catabolism of lipid emulsions and further to see if it differently affects long-chain triglyceride (LCT) and fish oil (FO) emulsions. MethodsA total of 24 C57BL/6J female mice, 10 to 12 weeks old, were randomly divided into 4 groups with 6 mice in each group. Two groups of mice were intravenously injected with dextran sulfate ( DexSO4 ) ( 1 mg/mouse) followed by intravenous injection of [1α, 2α(n)-3H] cholesteryl oleoyl ether [(3H)CEt] labelled LCT or FO emulsions (0.4mg triglycerde/mouse) at 2 minutes respectively, and other two groups were injected by saline as controls before injection of (3H)CEt labelled LCT and FO emulsions. Then, blood was drawn at fixed intervals to measure the radioactivities and the emulsion's fractional catabolic rates (FCR) were calculated. With the same procedures above mentioned, non-radiolabelled LCT and FO emulsions were intravenously injected to mice to determine liver uptake of lipid emulsions under electromicroscopy. Finally, THP1 cell line was used to examine the effects of DexSO4 on cell uptake of LCT and FO emulsions in vitro. ResultsPre-injection of DexSO4 to mice decreased the FCR of both LCT and FO emulsions at 72.38％ and 47.38％ respectively, as compared to controls ( P =0.020 ). Electromicroscopy showed that pre-injection of DexS04 decreased the uptakes of LCT and FO emulsions by Kupffer cells and sinusoidal endothelial cells similarly. In hepatocytes, no lipid droplets existed in mice with LCT emulsion injection, whereas some lipid droplets were still shown in mice with FO emulsions but with less quantities compared to control mice.In vitro, addition of DexSO4 to medium decreased THP1 cell uptakes of LCT and FO emulsions ( P =0.003 and 0.008) by 30.74％ and 41.60％ respectively. However, no differences were found in the effects of DexS04 on cell uptakes between LCT and FO emulsions ( P =0.080). ConclusionScavenger receptor A plays important roles in catabolism of lipid emulsions to some extent, and it's effects on FO emulsions may be less than LCT emulsions.

Objective To explore an effective evaluation method for students' self-and peer-assess-ment. Methods The students of 6 groups participating in extracurricular teaching activities were selected as research subject. Traditional method (final score = mean score of group/2 + teacher's score/2) and mean difference method [final score=teacher's score-(mean difference of group-mean difference of all groups)] were used to calculate final score of each group, and effect of two methods were compared. Results Scores of most groups were higher than the teacher's scores, and high scores were given by group 3 in self- and peer-assessment. The final score of all groups were higher than teacher's scores in traditional method. Compared with teacher's scores, final scores increased significantly in group 1, 4, 5 below mean difference, final score decreased significantly in group 2, 3 above mean difference, and final score did not differ in group 6 equal to mean difference in mean difference method. Conclusion The mean difference method can reflect the effect of student's self- and peer-assessment, and guide student to make objective and accurate evaluation. It is a more reasonable and scientific evaluation method for self-and peer-assessment.
