Chinese Journal of Tissue Engineering Research 2016;20(36):5364-5370
BACKGROUND:Stem cel transplantation is a promising treatment of advanced liver disease, and adipose-derived mesenchymal stem cel s have become another kind of popular cel s fol owing bone marrow mesenchymal stem cel s.
OBJECTIVE:To investigate the influence of adipose-derived mesenchymal stem cel transplantation on blood biochemical indices of liver cirrhosis rats.
METHODS:Sixty rats were equal y randomized into normal control, model and cel transplantation groups. Model rats of liver cirrhosis were made in the latter two groups through intragastric administration of carbon tetrachloride. One week after successful modeling, rats were given intraperitoneal injection of adipose-derived mesenchymal stem cel suspension in the cel transplantation group, and given normal saline in the other two groups.
RESULTS AND CONCLUSION:Compared with the normal control group, the model group showed a significant increase in the levels of alanine aminotransferase, aspartate aminotransferase, total bilirubin, total protein in liver tissues, serum level of malondialdehyde, 15-minute indocyanine green retention rate and degree of hepatic fibrosis, and a significant decrease in serum albumin level, serum albumin/globulin, levels of glutathione peroxidase and cyclic guanosine monophosphate in liver tissues. On the contrary, these indicators were al improved in the cel transplantation group compared with the model group.
Moreover, CM-Dil-positive cel s were visible in the liver tissue of rats undergoing adipose-derived mesenchymal stem cel transplantation. Al these findings indicate that adipose-derived mesenchymal stem cel transplantation can reduce liver cirrhosis in rats by acting on blood biochemistry levels.

Objective To investigate the effects of c-myc promoter binding protein(MBP-1)gene expression silencing on the pro-liferation in vitro in human gastric cancer cell line SGC-7901.Methods The cells divided into three groups:blank control group (cells without transfecting gastric cancer cell),negative control group(cells transfecting missense sequence)and experimental group (cells transfecting MBP-1 shRNA).Two MBP-1 shRNA sequences and one negative control shRNA sequence were designed,syn-thesized and cloned into pSIREN-retroQ plasma. Then the recombinant plasmids were constructed and transfected into human gas-tric cancer SGC-7901 cells by Lipofectamine 2000.After selection of cells with puromycin for two weeks, stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was obtained. The expressions of MBP-1 mRNA and protein in SGC-7901 were deter-mined by the real time PCR and Western blot, respectively. The effects of altered expression of MBP-1 on the cell proliferation were measured by MTT cell proliferation assay. Results PCR and sequencing indicated that the recombinant plasmids pSIREN-retroQ was constructed. Then the recombinant plasmids were transfected into human gastric cancer SGC-7901 cells by Lipofectamine 2000. After selection of cells with puromycin for two weeks, stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was ob-tained.The relative expression level MBP-1 mRNA in the MBP-1 siRNA transfection group was significantly decreased compared with the blank control group(P <0.05).Compared with the blank group, the expression levels of MBP-1 protein in the experimental group also significantly decreased. The proliferation abilities of SGC-7901 cells at 48,72,96,120 h after MBP-1 siRNA transfection were significantly increased compared with the blank control group (P < 0.05 ).Conclusion Down-regulating the expression of MBP-1 can obviously promote the proliferation of human gastric cancer cell line SGC-7901.MBP-1 gene may become the new target of gene therapy for gastric cancer.

OBJECTIVE: To explore the neuroprotective effect of electroacupuncture therapy on damaged cholinergic neurons in hippocampus in aged rats with Alzheimer's disease (AD). METHODS: Thirty six aged male rats were randomly divided into normal control group, sham-operated group, untreated group and electroacupuncture group. Animal model of AD was established with fimbria-fornix transection. The rats in the electroacupuncture group received electroacupuncture on Baihui (DU 20), Yongquan (KI 1), Taixi (KI 3) and Xuehai (SP10). The activity of choline acetyltransferase (ChAT) in septal area of brain was detected by radioimmunoassay, and the protein expressions of nerve growth factor (NGF) and c-fos in CA3 region of hippocampus were detected by immunohistochemical assay. RESULTS: The ChAT activity and the expression levels of NGF and c-fos proteins in the electroacupuncture group were significantly higher than those in the untreated group. CONCLUSION: Electroacupuncture therapy can protect cholinergic neurons in hippocampus in aged rats with AD by means of promoting synthesis of c-fos protein and increasing the expression level of NGF.

Chinese Journal of Experimental Ophthalmology 2016;34(5):420-425
Background p21 is a cyclin-dependent kinase inhibitor, and it can prevent cells from going through the G1/S phase checkpoint and inhibit cell proliferation.Stuies determined that the expression level of p21 WAF1/CIP1 is associated with proliferative diseases.Traumatic proliferative vitreoretinopathy (PVR) is a proliferative response of eye.Understaining the relationship of dynamic expression levels of p21 WAF1/CIP1 in PVR is of significance for the prevention and management of PVR.Objective This study was to investigate the expression of p21 WAF1/CIP1 during the course of experimental traumatic PVR in rabbits. Methods Fifty-four pigmented rabbits were randomized into the normal control group and different experimerital groups, and one lateral eye of each rabbit served as experimental eye.PVR models were established by intravitreal injection of human platelet-rich plasma (PRP) (0.4 ml)combined with cryotherapy for 5 seconds, and vitreous and retinas were examined with B type sonography. The rabbits were sacrificed in 7,14,21 and 28 days after operation, and histopathological examination of the retinas was performed by haematoxylin and eosin stain. The expression levels of p21WAF1/CIP1 protein and gene were detected by immunohistochemistry,Western blot and reverse transcription-PCR (RT-PCR).The use and care of the rabbits complied with Statement of ARVO.Results B type sonography showed that the retinal morphology was normal in the normal control group. However, the proliferative membrane was gradually thickened 1 to 7 days after operation.Retinal folds of rabbits were seen in 7 days, and tractional retinal detachment was found in 14 days and 28 days after operation. The histopathological examination of the retinas showed epiretinal membrane and infiltration of inflammatory cells 7 days and fixed ruffle 28 days after operation. The p21WAF1/CIP1 was strongly expressed in the cell nucleus of retinal ganglion cell layer (GCL) and inner nuclear layer (INL) in the normal control group, and the expression was gradually weakened after modeling, with the weakest expression in the retinas in 14 days after modeling. The relative expression levels of p21 WAF1/CIP1 protein was 0.74±0.08,0.60±0.05,0.56±0.03,0.74±0.02 and 0.65 ±0.04 in the normal control group, postoperative 7-day group, postoperative 14-day group, postoperative 21-day group and postoperative 28-day group, respectively, showing a significant difference among the groups (F =20.55,P =0.00),and the expression levels of p21WAF1/CIP1 protein were significantly lower in the postoperative 7-day group and postoperative 14-day group than those of the normal control group, postoperative 21-day group and postoperative 28-day group (all at P<0.05).The relative expression levels of p21 WAF1/CIP1 mRNA was 0.65 ± 0.09,0.57 ± 0.05,0.45 ±0.04,0.46±0.02 and 0.47±0.04 in the normal control group, postoperative 7-day group, postoperative 14-day group, postoperative 21-day group and postoperative 28-day group, respectively, with a significant difference among the groups (F =18.06,P =0.00),and the expression levels were significantly lower in the postoperative 14-day group, postoperative 21-day group and postoperative 28-day group than those of the normal control group and postoperative 7-day group (all at P<0.05).Conclusions The dynamic expression of p21WAF1/CIP1 in the retinas is consistant with the prograssion of traumatic PVR,and the reduce tendency of p21 WAF1/CIP1expression is similar to cell prolieration change, indicating that reduce of p21WAF1/CIP1 expression in the retinas may promote the development of traumatic PVR.
